    Mechanisms of nanoparticles or cigarette smoke induced inflammation : study of signalization pathway mediated by ST2 receptor and NLRP6

    Les maladies pulmonaires, responsables de 3,1 millions de décès de part le monde représentent un problème majeur de santé publique. En particulier, la fibrose pulmonaire et la broncho-pneumopathie chronique obstructive (BPCO) conduisent à la perte de la fonction pulmonaire. Aucun traitement efficace n’a été identifié à ce jour pour lutter contre ces maladies, la seule alternative étant la transplantation. Au cours de ma thèse, j’ai exploré les mécanismes du développement de ces maladies en utilisant différents modèles chez la souris, soit par l’instillation de nanoparticules de métaux ou de bléomycine, conduisant à l’inflammation et/ou à la fibrose pulmonaire, soit par exposition à la fumée de cigarette provoquant une inflammation. Nous avons montré le rôle de la voie de signalisation IL-33/ST2 dans les réponses inflammatoires induites par les nanoparticules ou la bléomycine et identifié de nouveaux mécanismes de régulation de l’IL-33 au sein des macrophages, différents de ceux décrits pour les cellules épithéliales. Nos résultats indiquent que l’expression intracellulaire de l’IL-33 et de son récepteur ST2, joue un rôle important dans l’inflammation, ainsi que la translocation nucléaire de l’IL-33. D’autre part, mes travaux de thèse ont permis d’identifier le rôle clef du senseur intracytosolique NLRP6 dans l’inflammation provoquée par l’exposition à la fumée de cigarette. Nos résultats indiquent que NLRP6, aux fonctions pulmonaires inexplorées, contrôle l’activation des cellules épithéliales et le recrutement des neutrophiles de façon indépendante de la formation d’un inflammasome mais dépendante de la signalisation par les récepteurs des interférons de type I et III.Pulmonary diseases are a major health problem with 3.1 million deaths in the worldwide. Among them pulmonary fibrosis and chronic obstructive pulmonary disease (COPD), which occur after repeated lung epithelium injury, are characterized by impaired lung functions. To date, no effective therapy against pulmonary fibrosis and COPD were developed, lung transplantation being the only alternative. During my thesis, I studied the mechanisms leading to disease development using different experimental models in mice in particular by metal dioxide nanoparticles or bleomycin instillation leading to inflammation and/or pulmonary fibrosis, or by cigarette smoke exposure promoting pulmonary inflammation which may lead to emphysema. We show the crucial role of IL-33/ST2 signaling pathway in response to nanoparticles or bleomycine and identify new mechanisms for IL-33 regulation in macrophages which are different from those described in epithelial cells. Our results indicate that intracellular expression of IL-33 and of its receptor ST2, together with nuclear IL-33 translocation, play an important role in inflammatory response to nanoparticles instillation. On the other hand, my thesis work allowed identifying that the cytosolic sensor NLRP6 as a key player in pulmonary inflammation developed upon mouse cigarette smoke exposure. Interestingly, our results show that the receptor NLRP6, whose pulmonary functions are still unexplored, controls epithelial cells activation leading to neutrophils recruitment in the airways, in an inflammasome-independent manner but dependently of type I and III interferon receptors signaling

    Le tabagisme est le premier facteur déclencheur de la broncho-pneumopathie chronique obstructive, induisant une inflammation pulmonaire chronique et notamment le recrutement de neutrophiles. Cette pathologie est actuellement sans traitement curatif. L’objectif de mon travail a été de mieux comprendre les mécanismes de la mise en place de l’inflammation pulmonaire provoquée par l’exposition aigüe à la fumée de cigarette en étudiant trois axes: le rôle du récepteur NLRP6, l’implication de l’ADN du soi et de la voie STING et enfin les effets de la cytokine BAFF. Nous avons exposé des souris à la fumée de cigarette durant 4 jours, puis nous avons évalué le recrutement cellulaire, en particulier les neutrophiles, et les paramètres inflammatoires résultant de cette exposition. Nous montrons que le récepteur NLRP6 contrôle la sécrétion de la chimiokine des neutrophiles CXCL5 produite par les cellules épithéliales pulmonaires indépendamment de la formation de l’inflammasome. De plus, NLRP6 exprimé au niveau de l’intestin contrôle la sécrétion de CXCL5 par les cellules épithéliales intestinales mais aussi par les cellules épithéliales pulmonaires via son action sur la composition du microbiote intestinal, mettant en évidence l’existence un axe intestin-poumon dans la régulation de l’inflammation pulmonaire par la fumée de cigarette. Nous montrons pour la première fois que cette exposition induit une libération d’ADN du soi dans l’espace broncho-alvéolaire, détecté par le senseur cGAS et conduisant à l’activation de STING, à la production d’interféron de type I et à l’afflux de neutrophiles dans les voies respiratoires qui vont mourir par NETose, libérant également leur ADN dans le compartiment extracellulaire. Les neutrophiles sont également une source précoce de la cytokine BAFF qui est sécrétée dans l’espace broncho-alvéolaire, celle-ci étant impliquée dans la formation de follicules lymphoïdes lors d’expositions chroniques à la fumée de cigarette. Cette étude a permis de mettre en évidence de nouveaux mécanismes de l’inflammation provoquée par la fumée de cigarette et de proposer de nouvelles cibles thérapeutiques.Cigarette smoking is the main trigger for chronic obstructive pulmonary disease, inducing chronic inflammation with cellular recruitment and in particular neutrophils. This pathology has no curative treatment. The aim of my work was the better understanding of the inflammatory mechanisms developed in response to acute cigarette smoke exposure by studying three axes: the role of the NLRP6 receptor, of self-DNA release and its sensing by the cGas/STING pathway and the production of the cytokine BAFF in the setting up of this inflammation. We exposed mice to cigarette smoke for 4 days and we evaluated cellular recruitment, in particular neutrophils influx in the bronchoalveolar space and the inflammatory parameters resulting from this exposure. We have demonstrated that the NLRP6 receptor controls the secretion of the CXCL5 neutrophil chemokine produced by pulmonary epithelial cells independently of the classical inflammasome in the lung and the gut. Moreover, we demonstrate that NLRP6 expressed in the intestine controls the secretion of CXCL5 by the intestinal epithelial cells but also by the pulmonary epithelial cells via its action on the composition of the intestinal microbiota, thus highlighting the existence of a gut-lung axis in the regulation of pulmonary inflammation to cigarette smoke. We show for the first time that this exposure induces a release of self DNA in the bronchoalveolar space, detected by the cGAS sensor and leading to the activation of the STING pathway and the production of type I interferon and neutrophils influx into the respiratory tract. These recruited neutrophils will die by NETose, also releasing their DNA into the extracellular compartment. In addition, neutrophils are also an early source of the BAFF cytokine secreted into the bronchoalveolar space and which has been involved in the formation of lymphoid follicles during chronic exposures to cigarette smoke. This study allowed to highlight new inflammatory mechanisms caused by cigarette smoke exposure and to propose new therapeutic targets

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    Les mécanismes de défense de l’organisme contre des pathogènes ou des particules toxiques sont regroupés sous le terme d’Immunité. L’Immunité a développé plusieurs familles de récepteurs dont les NLR (Nod Like Receptors) spécialisés dans la reconnaissance de motifs de pathogènes (PAMP), de signaux de danger endogènes (DAMP) et de désordres métaboliques et capables d’engager une réponse inflammatoire. L’inflammation est dite stérile si elle n’est pas déclenchée par des agents infectieux mais par des molécules endogènes en excès comme l’acide urique et l’ATP ou par des polluants environnementaux tels que la silice et les nanoparticules. Nous nous sommes d’une part intéressés aux mécanismes d’activation de l’inflammasome NLRP3 qui permet la maturation d’une cytokine pro-inflammatoire, l’Interleukine (IL)-1β. Nous montrons que les cristaux d’acide urique, de silice et d’Alum ainsi que les nanoparticules de titane et de silice induisent une libération active d’ATP dépendante d’hémicanaux par les cellules. Nous mettons en évidence un nouveau mécanisme d’activation de l’inflammasome NLRP3 qui fait intervenir l’ATP et ses différents métabolites, en particulier l’adénosine, via des familles de récepteurs purinergiques. Nous nous sommes d’autre part focalisés sur l’inflammation pulmonaire induite par l’administration de nanoparticules dans un modèle murin. Nous avions montré dans une étude précédente que cette inflammation est dépendante des IL-1α et IL-1β et de leur récepteur IL-1R1. Nos résultats montrent que l’adénosine est impliquée in vivo dans la réponse inflammatoire aux nanoparticules et que l’inflammation des voies respiratoires est modulée par l’IL-33 et son récepteur ST2.The mechanisms of defense of the organism against pathogens or oxious particles are termed Immunity. The Immunity developed several receptors’ families among which the NLR (Nod-Like Receptors), specialized in the recognition of pathogen patterns (PAMPs), of endogenous danger signals (DAMPs) and of metabolic disorders, and capable of triggering an inflammatory response. The inflammation is named “sterile” if it is not activated by infectious agents but by endogenous molecules in excess, as uric acid and ATP, or by environmental pollutants, such as silica and nanoparticles. We first became interested in the mechanisms of activation of the NLRP3 inflammasome which allows the maturation of a pro-inflammatory cytokine, the Interleukine (IL) -1β. We show that uric acid, silica or Alum crystals but also titanium and silica nanoparticles induce an active release of ATP by cells, dependent of hemichannels. We highlight an innovative mechanism of activation of the NLRP3 inflammasome which involves ATP and its metabolites, in particular adenosine, through purinergic receptor families. We then focused on the nanoparticle-induced lung inflammation in a murine model. We had shown in a previous study that this inflammation is dependent on IL-1α, IL-1β and their common receptor IL-1R1. Our in vivo results show that adenosine is involved in nanoparticle-induced inflammatory response and that airway inflammation is modulated by IL-33 and its receptor ST2

    Dans le cadre de notre projet, nous avons étudié l'implication des récepteurs NOD-like receptor family, pyrin domain containing 3 et 6 (NLRP3 et NLRP6) et de la Gasdermine D dans l'inflammation pulmonaire induite par une exposition à la fumée de cigarette d'une part et à l'ozone d'autre part, chez la souris. Dans une première partie, nous avons montré que l'inflammation pulmonaire induite par une exposition à la fumée de cigarette était dépendante de NLRP3 puisque les souris déficientes (Nlrp3⁻/⁻) et sauvages traitées avec un inhibiteur de NLRP3 ont une inflammation réduite. L'activation de la Gasdermine D dans les cellules épithéliales bronchiques et les macrophages recrutés au niveau de l'espace bronchoalvéolaire dépend de NLRP3. Dans une seconde partie, nous avons mis en évidence que l'inflammation pulmonaire induite par une exposition à l'ozone était dépendante de NLRP6 puisque les souris déficientes (Nlrp6⁻/⁻) ont une diminution de cette inflammation, des paramètres de fibrose et d'emphysème. L'activation de la Gasdermine D dans les pneumocytes de type 1 et une proportion réduite de macrophages recrutés dans l'espace bronchoalvéolaire dépend de NLRP6 et de l'activation de la caspase 11.In our project, we studied NOD-like receptor family, pyrin domain containing 3 and 6 (NLRP3 and NLRP6) and Gasdermin D implications in smoke cigarette or ozone induced pulmonary inflammation in mice.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Innate immunity and inflammasome : role of endogenous danger signals

    La théorie du danger développée par Polly Matzinger stipule que l’attrait principal du système immunitaire ne réside pas dans la distinction entre le soi « à protéger » et le non-soi « à combattre ». Toute situation potentiellement délétère pour l’hôte, avec émission de signaux de dangers endogènes, est à ce titre capable de générer une réponse immunitaire afin de mobiliser les acteurs capables de permettre un retour à une situation basale. L’exposition des poumons de façon répétée à des agents toxiques environnementaux se traduit par une inflammation et une fibrose pulmonaire en condition stérile, c'est-à-dire sans intervention de micro-organisme. L’administration de Bléomycine dans les poumons de souris est un bon modèle pour étudier les molécules endogènes ou signaux de dangers engagés et les voies de signalisations associées. Nous avons identifié l’ATP extracellulaire et l’acide urique, provenant des cellules stressées ou endommagées, comme capables d’induire l’activation d’un complexe protéique cytoplasmique appelé inflammasome Nlrp3. Celui-ci conduit à la maturation de l’interleukine-1β, cytokine pro-inflammatoire. Dans une seconde partie, nous nous sommes intéressés aux mécanismes moléculaires d’activation de l’inflammasome Nlrp3 en réponse à des agents particulaires, responsables notamment de pathologies pulmonaires après inhalation. Nos résultats montrent que de l’ATP endogène est libéré activement par les cellules mises en contact avec des particules. Par un mécanisme autocrine/paracrine l’ATP va ensuite signaler sur plusieurs récepteurs purinergiques membranaires. Cette signalisation purinergique est importante dans la capacité des cellules à produire de l’interleukine-1β mature. Au final, les travaux présentés dans ce manuscrit attestent du rôle critique de molécules endogènes dans la mise en place d’une réponse immunitaire innée basée sur l’activité de l’inflammasome Nlrp3.The Danger Theory developed by Polly Matzinger specifies that what really matters for the immune system is not to distinguish between self « to protect » and non-self « to fight ». Any potentially detrimental situation for the host would be able to generate endogenous danger signal that are able to mount an immune response to eventually return to baseline. Lung repeated expositions to toxic environnemental pollutants lead to inflammation and then fibrosis in sterile condition, meaning without a role of micro-organism. Lung administration of Bleomycin in mice is a good model to define what are the endogenous molecules or danger signals and associated pathways. We have identified extracellular ATP and uric acid as danger signals released by stressed or damaged cells and able to produce pro-inflammatory interleukin-1β through activation of a cytoplasmic complex called the Nlrp3 inflammasome. We then became interested in the molecular mechanism of particle-induced Nlrp3 inflammasome activation in vitro. We demonstrate that particles such as silica or alum hydroxide lead to endogenous ATP release and further purinergic signaling via an autocrine/paracrine manner. We identified purinergic signaling as an essential triggering of interleukin-1β maturation in response to particles. As a whole, the work presented here highlights the critical role of two endogenous molecules, ATP and uric acid, in the Nlrp3-mediated immune response

    Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods
