    Etude par imagerie TEP SV2A de la synaptogenèse pharmaco-induite dans un modèle préclinique de dépression

    In 2019, 5% of the world's population suffers from depressive disorders (World Health Organisation). Major depressive disorder (MDD) requires psychological and pharmacological treatment, in particular selective serotonin reuptake inhibitors (SSRIs), which are the first line of treatment. However, a third of patients are suffering from treatment-resistant depression (TRD). Ketamine, an N-methyl-D-aspartate (NMDA) receptor antagonist, is now FDA approved for this TRD-patients. However, its mechanism of action remains poorly understood. While synaptic loss has been observed post-mortem in the brain of MDD patients, antidepressants appear to act on varying degrees on synaptogenesis, i.e. the formation of new synapses. The aim of this thesis was to assess the pharmaco-induced synaptic modulation non-invasively using PET (positron emission tomography) imaging. For this, we chose [11C]UCB-J, a radiotracer targeting specifically the SV2A (synaptic vesicle 2A) glycoprotein, which is used as a marker of synaptic density. This imaging was carried out to study the effect of different treatments in a neuroendocrine model of depression (CORT model).A SV2A PET imaging study was carried out in CORT mice after a behavioural study had confirmed their anxiety-depressive endophenotype. This confirmed the relevance of this model, which shows significant synaptic loss in several brain regions. PET imaging using [11C]UCB-J also revealed the effects of fluoxetine, a classic SSRI, on synaptogenesis in this model after 5 weeks of treatment. In a second study, the acute (24h post-injection), sub-acute (1 week) and sub-chronic (3 weeks) effects of repeated administration of ketamine were studied in this same model, but also in naïve -non-corticosterone-treated mice. This provided a better understanding of the dynamics of ketamine action, which differ in CORT and healthy mice. It also enabled a correlation to be established between SV2A PET results in vivo and SV2A expression measured ex vivo. The SV2A PET results were then compared with those obtained with [18F]fluoro-D-desoxy-glucose PET under the same conditions, demonstrating the value of SV2A PET compared with this conventional marker of cerebral metabolism. Finally, as ketamine is only approved for use in combination with an SSRI, the study of a “booster” effect of ketamine (repeated dose at the induction of treatment) on a conventional treatment (fluoxetine) was investigated. This longitudinal study highlighted the complementary effect of these molecules in stimulating synaptogenesis rapidly and over a long period.This work showcase the full advantage of the non-invasive and translational nature of PET imaging by validating the relevance of [11C]UCB-J as a marker of drug-induced synaptogenesis in a mice model of depression. The results provide information about the dynamics of synaptogenesis induced by fluoxetine, ketamine and their combination in a preclinical model, validated by behavioural studies. Tandis qu'une perte synaptique a été observée en post-mortem chez les patients atteints de TDM, les antidépresseurs semblent agir à différents degrés sur la synaptogenèse, i.e la formation de nouvelles synapses. Ce travail de thèse a eu pour objectif d'étudier de manière non-invasive la modulation synaptique pharmaco-induite grâce à l'imagerie TEP (tomographie par émission de positons). Pour cela, nous avons choisi le [11C]UCB-J, un radiotraceur spécifique de la glycoprotéine SV2A qui est utilisé comme marqueur de la densité synaptique. Cette imagerie a été réalisée pour étudier l'effet de différents traitements dans un modèle neuroendocrinien de dépression (modèle CORT).Dans un premier temps, une étude d'imagerie TEP SV2A a été réalisée chez des souris CORT après une étude comportementale ayant confirmé leur endophénotype anxiodépressif. Cela a permis de confirmer la pertinence de ce modèle, qui présente une perte synaptique significative dans plusieurs régions du cerveau. Aussi, l'imagerie TEP utilisant le [11C]UCB-J a permis de mettre en évidence les effets de la fluoxétine, un ISRS classique, sur la synaptogénèse dans ce modèle après 5 semaines de traitement. Dans une deuxième étude, les effets aigus (24h post-injection), subaigus (1 semaine) et subchroniques (3 semaines) d'une administration répétée de kétamine ont été étudiés sur ce même modèle, mais aussi chez des souris saines. Cela a permis de mieux comprendre la dynamique d'action de la kétamine qui diffèrent chez les souris CORT et les souris saines. De plus, cela a permis d'établir une corrélation entre les résultats TEP SV2A in vivo et l'expression de SV2A mesurée ex vivo. Ensuite, les résultats TEP SV2A ont pu être comparés à ceux obtenus avec la TEP au [18F]fluoro-D-desoxy-glucose dans les mêmes conditions permettant de montrer l'intérêt de la TEP SV2A par rapport à ce marqueur classique du métabolisme glucidique cérébral. Enfin, l'utilisation de la kétamine étant approuvée seulement en association avec un ISRS, l'étude d'un effet « booster » de kétamine (dose répétée en début de traitement) sur un traitement classique (fluoxétine) ont été étudiés. Cette étude longitudinale a permis de mettre en évidence la complémentarité de ces molécules pour stimuler la synaptogénèse de manière rapide et prolongée.Ce travail de thèse tire pleinement parti du caractère non-invasif et translationnel de l'imagerie TEP en validant la pertinence du [11C]UCB-J comme marqueur de la synaptogénèse pharmaco-induite dans un modèle de dépression. Les résultats nous renseignent sur la dynamique de la synaptogénèse induite par la fluoxétine, la kétamine et leur combinaison dans un modèle préclinique, validé par des études de comportements.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
