    Radiocarbon evidence for the early Bronze Age Levant: The site of tell Fadous-Kfarabida (Lebanon) and the end of the early bronze III period

    Absolute dates for the end of the Early Bronze Age ancient Near East are of crucial importance for assessing the nature and extent of mid- to late 3rd millennium BC transitions in the Near East and their alleged link to the 4.2ka BP climatic event. This article presents a radiocarbon sequence for the Early Bronze Age site of Tell Fadous-Kfarabida (Lebanon) and argues that the end of the Early Bronze III period has to be dated considerably higher than previously estimated.     Adaptive Smolyak Pseudospectral Approximations

    Polynomial approximations of computationally intensive models are central to uncertainty quantification. This paper describes an adaptive method for nonintrusive pseudospectral approximation, based on Smolyak's algorithm with generalized sparse grids. We rigorously analyze and extend the nonadaptive method proposed in [P. G. Constantine, M. S. Eldred, and E. T. Phipps, Comput. Methods Appl. Mech. Engrg., 229--232 (2012), pp. 1--12], and compare it to a common alternative approach for using sparse grids to construct polynomial approximations, direct quadrature. Analysis of direct quadrature shows that O(1)\mathcal{O}(1) errors are an intrinsic property of some configurations of the method, as a consequence of internal aliasing. We provide precise conditions, based on the chosen polynomial basis and quadrature rules, under which this aliasing error occurs. We then establish theoretical results on the accuracy of Smolyak pseudospectral approximation, and show that the Smolyak approximation avoids internal aliasing and makes far more effective use of sparse function evaluations. These results are applicable to broad choices of quadrature rule and generalized sparse grids. Exploiting this flexibility, we introduce a greedy heuristic for adaptive refinement of the pseudospectral approximation. We numerically demonstrate convergence of the algorithm on the Genz test functions, and illustrate the accuracy and efficiency of the adaptive approach on a realistic chemical kinetics problem.American Society for Engineering Education. The final chapter of the theoretical part of the work is devoted to the fashion industry itself, fast fashion and trends. Next, Generation Z, its perception, purchasing and consumption behavior is brought up. In the practical part, based on the theoretical part, the author defines three areas of questioning for use in qualitative research. The interviews are conducted in a semi-structured manner. The analysis of the interviews then tries to answer the research question of the perception of greenwashing by Generation Z

Teoretická část nejprve popisuje koncept green marketingu, jeho definici, přínos pro firmy i společnost a také jeho kritiku. Následující kapitola se věnuje greenwashingu, jeho historii, uvádí nejčastější praktiky a následně se věnuje strategii pro udržitelnou komunikaci. Závěrečná kapitola teoretické části práce se věnuje samotnému módnímu průmyslu, fast fashion a trendům. Dále je pak přiblížena generace Z, její vnímání a nákupní a spotřební chování. V praktické části autorka na základě teoretické části stanovuje tři otázkové okruhy pro využití v kvalitativním výzkumu. Rozhovory jsou vedeny semi strukturovanou formou.     Anwendungen des Empirischen Likelihood-Schätzers der Fehlerverteilung in AR(1)-Prozessen

    Mit den Arbeiten von Owen wurde das empirische Likelihood-Prinzip (kurz EL-Prinzip) in die mathematische Statistik eingeführt. Es ermöglicht im Modell unabhängig und identisch verteilter Daten unter anderem die nichtparametrische Schätzung von Verteilungen unter Zusatzinformationen mittels einer modifizierten empirischen Verteilungsfunktion. Darüber hinaus wurde es in jüngster Zeit (z. B. in der Diplomarbeit des Verfassers) in Zeitreihenmodellen erfolgreich zur Schätzung der Fehlerverteilung angewendet. In der vorliegenden Arbeit werden weitere mögliche Anwendungen des EL-Prinzips diskutiert. Der erste Teil der Arbeit beschäftigt sich mit Anpassungstests auf parametrische Verteilungsklassen unter Voraussetzung der Zentriertheit. Mithilfe der Theorie empirischer Prozesse mit geschätztem Parameter werden adäquate Tests vom Kolmogorov-Smirnov-Typ sowohl im Modell unabhängig und identisch verteilter Daten als auch im AR(1)-Modell konstruiert. Dabei ist jeweils ein funktionaler Grenzwertsatz unter der Hypothese des Testproblems grundlegend. Die Konsistenz der zugehörigen Bootstraps-Prozesse wird bewiesen, um die Ergebnisse praktisch anwendbar zu machen, d.h. Bootstrap-Tests angeben zu können. Simulationsstudien schließlich zeigen die durch die Ausnutzung der Zentriertheit erhaltenen Güteverbesserungen gegenüber den klassischen Tests auf. Der letzte Teil der Arbeit widmet sich nichtparametrischen Ein- und Zweischritt-Prognoseintervallen in stabilen AR(1)-Prozessen auf Basis der gewöhnlichen wie der modifizierten empirischen Verteilungsfunktion der Residuen. Dazu wird zunächst die Konsistenz der so konstruierten Intervalle gezeigt. In einem zweiten Schritt werden in beiden Fällen die asymptotischen Verteilungen ihrer bedingten Überdeckungswahrscheinlichkeiten ermittelt und verglichen.The empirical Liklihood principle (EL-principle) was introduced into mathematical statistics by Owen. It allows for the estimation of an unknown probability distribution of independent and identically distributed random variables if some auxiliary infomation is available. It has also been successfully used recently in autoregressive models to estimate the innovation distribution (in the author´s diploma thesis, for example). In the present thesis further applications of the EL-principle are discussed.The first part of the thesis is concerned with goodness-of-fit tests for parametric classes of distributions when the mean of all distribution is zero under the hypotheses. Using the theory of empirical processes with estimated parameters tests of Kolmogorov-Smirnov type are established for independent and identically distributed random variables as well as in AR(1)-models. For these results two functional central limit theorems are essential. In a second step consistency of the associated bootstrap processes is shown to make the results applicable by deriving bootstrap tests. Simulations show the gain of power in comparison to the classical tests. The last part of the thesis examines nonparametric prediction intervals of lag one and two in stable AR(1)-models based on the common empirical distribution function of the residuals as well as the one modified according to the EL-principle.     This construct aims to identify the pedagogical experiences that contributed to the formation of a student who participated in the practices of the music curriculum applied to Elementary School - Early Years, set in the NEI/CAp-UFRN institution. It constitutes a qualitative research, which appropriates the narratives of life and the (auto)biographical approach as a methodology. It aims to present the formative experiences when visiting self-knowledge (JOSSO, 2002) together with a reflective look at academic training, the relationship of the subject and decision-making before the teacher, through self-writing and self-reflection. The discussions raised about the path of teacher training in music education become present in the dialogue of teaching practice, professional identification and teaching knowledge. When entering the narratives of life, the self-listening and recollection of the experience was initially experienced, in order to approach the teaching action, by presenting the first experiences of the educational practices experienced, promoted by the ambience of the training site. In this, the formative experiences made possible through the teaching performance are evident, when debuting as an actor and author of academic training. It is observed that in music teaching performances, being an educator also evokes skills that must be acquired and refined throughout teaching, permeated by singularities and peculiarities that must be experienced according to the environment in which it is inserted as an actor. Finally, the point is that the place where the apprentice moves tends to do it and remake it, in an active and perpetual (re)construction by him wherever he goes.Este constructo tem como objetivo identificar as vivências pedagógicas que contribuíram para a formação de um licenciando participante das práticas do currículo de música aplicado ao Ensino Fundamental - Anos Iniciais, ambientadas na instituição NEI/CAp-UFRN. Constitui-se uma pesquisa qualitativa, a qual se apropria das narrativas de vida e da abordagem (auto)biográfica como metodologia. A mesma visa apresentar as experiências formadoras ao visitar o conhecimento de si (JOSSO, 2002) junto a um olhar reflexivo à formação acadêmica, a relação do sujeito e as tomadas de decisões ante o fazer docente, por meio da escrita de si e da autorreflexão. As discussões levantadas quanto ao trajeto da formação docente no ensino da música tornam-se presentes no diálogo da prática docente, da identificação profissional e dos saberes da docência. Ao adentrar nas narrativas de vida vivenciou-se, inicialmente, a auto-escuta e rememoração da experiência para, aproxima-se da ação docente, ao apresentar as primeiras experiências diante das práticas educativas vivenciadas, promovidas pela ambiência do local formativo. Nisso, evidencia-se as experiências formadoras oportunizadas por meio da atuação docente, ao estrear como ator e autor da formação acadêmica. Observa-se que nas atuações do ensino de música, estar como educador evoca também habilidades que deverão ser adquiridas e lapidadas ao longo da docência, permeadas por singularidades e peculiaridades que deverão ser vivenciadas conforme o ambiente em que estiver inserida como atuante.     Although cause-related marketing (CRM) is being increasingly applied, little is known about GenZ skepticism. This study aimed to understand the antecedents and consequences of skepticism towards CRM offers among GenZ consumers. Quantitative research was used to test the conceptual model using structural equation model (SEM). The analysis confirmed the proposed hypothesis. The author\u27s findings provide empirical evidence that altruism and the perceived company’s motivation negatively affect skepticism toward a CRM campaign. This study further proves the effect of consumer skepticism on purchase intention. As skepticism increases, the willingness to buy cause-related products decreases. The findings are relevant for both scholars and marketers. Scientists will be enriched with knowledge about the prosocial behaviour of Generation Z. From a managerial perspective, this research could help practitioners to design CRM campaigns effectively. To the best of the author’s knowledge, no such study has been conducted in the context of Poland.Marketing społecznie zaangażowany jest coraz częściej praktykowany, jednakże niewiele jest wiadomo na temat sceptycyzmu pokolenia Z. Celem badań jest zrozumienie przyczyn i konsekwencji sceptycyzmu wobec programów CRM wśród Z konsumentów. Badania ilościowe wykorzystano do przetestowania modelu koncepcyjnego za pomocą modelu równań strukturalnych (SEM). Wszystkie hipotezy zostały potwierdzone. Badania dostarczają empirycznych dowodów na to, że altruizm konsumencki i postrzegana motywacja firmy negatywnie wpływają na sceptycyzm wobec kampanii CRM. Ponadto pokazują jego wpływ na intencję zakupu konsumentów pokolenia Z. Wraz ze wzrostem sceptycyzmu zmniejsza się ich skłonność do nabycia produktów wspierających sprawę społeczną. Wyniki badań są istotne zarówno dla akademików, jak i praktyków. Pracownicy naukowi zostaną wzbogaceni o wiedzę na temat prospołecznych zachowań pokolenia Z.     Regulation of Trypanosoma brucei Polyamine Biosynthesis

    Human African Trypanosomiasis (HAT), also known as Sleeping Sickness, is a disease caused by the protozoan parasite Trypanosoma brucei. About 70 million people are at risk of infection in sub-Saharan Africa. While current treatments have efficacy, they are difficult to administer and serious toxicity is associated with one of the key compounds, emphasizing the need for novel therapeutic approaches. The polyamine biosynthetic pathway is vital for parasite viability and the first committed step catalyzed by ornithine decarboxylase is the target of Eflornithine, which is used to treat late stage disease. Our lab has investigated this pathway to identify other potential targets for drug development. During this work I also demonstrated that the parasite employs unique regulatory strategies to control metabolism through this pathway. Among them, I found T. brucei S-adenosylmethionine decarboxylase (TbAdoMetDC), which is a homodimer in other eukaryotes, is a heterodimer in the trypanosomatids. The active enzyme is composed of two paralogs, one with limited activity (AdoMetDC) and the other inactive, which I termed prozyme. Prozyme activates trypanosomatid AdoMetDC through an allosteric mechanism that involves relief of autoinhibition through a conformational change. Prozyme is found in limited quantities relative to TbAdoMetDC and upregulated upon knockdown or chemical inhibition of TbAdoMetDC, providing a mechanism to regulate polyamine flux in the cell. The cellular mechanism of regulation is not fully understood, as gene expression in T. brucei differs significantly from that of mammals. Herein, I show prozyme is regulated at the level of protein translation. I found that TbAdoMetDC suppresses prozyme expression at the protein level in an enzyme activity-independent manner. Upon its loss, prozyme is expressed constitutively in an unregulated manner. I also show that the enzymatic product of TbAdoMetDC, dcAdoMet, acts as a metabolic signal for prozyme upregulation. Under chemical inhibition of AdoMetDC by Genz-644131 I show dcAdoMet is the only significant metabolite to correlate with prozyme protein upregulation. I further support this hypothesis by characterizing the effects of knockdown of S-adenosylmethionine synthetase by RNAi and through methionine starvation to correlate dcAdoMet depletion with prozyme upregulation independent of AdoMetDC manipulation. Through this work I also demonstrated that TbAdoMetSyn is an essential enzyme and validate the activity of its allele.
