HER2 breast cancer therapies: a review
Conleth G Murphy, Shanu ModiBreast Cancer Medicine Service, Department of Medicine, Memorial Sloan-Kettering Cancer Center, New York, NY, USAAbstract: Amplification of the HER2 gene and/or overexpression of its protein product have been found in up to 25% to 30% of human breast cancers and have been shown to be associated with poorer outcomes compared to ‘HER2 normal’ breast cancer. Research has focused on developing therapies directed to the HER2 receptor and its pathway. These include the monoclonal antibody trastuzumab, which has improved outcomes when used in patients with both advanced and early breast cancer. Lapatinib is a small-molecule tyrosine kinase inhibitor which has demonstrated activity in advanced breast cancer and is currently being evaluated in early stage disease. We discuss the therapeutic rationale and clinical trial experience with these agents. Other novel and emerging strategies targeting the HER2 receptor and its pathway are also discussed. These strategies include novel HER2 antibodies and small-molecule inhibitors, antibody–drug conjugates, agents targeting downstream components of the HER2 signaling pathway, and heat shock protein 90 (HSP90) inhibitors.Keywords: HER2, human epidermal growth factor receptor 2, breast cancer, trastuzumab, lapatini

Filmmaker Noel Murphy describes the creation and content of his film, "The Last Dymaxion: Buckminster Fuller's Dream Restored." Murphy talks about the genius and legacy of Richard Buckminster "Bucky" Fuller, an American architect, author, inventor, and futurist as he introduces the film. The discussion is interrupted for the special screening of the film and picks up again after the screening. The soundtrack is not recorded. Murphy talks about re-editing the film to emphasize the design science thesis and synergy motif and make connections between Fuller's vision and technology being used today. He responds to audience questions and entertains additional questions after his presentation. Murphy is introduced by Michigan State University Librarian Michael Rodriguez. Collaboration and interconnectivity: Nottinghamshire Fire and Rescue Services and higher education institutions in Nottingham
This paper will describe the developing relationship between Nottinghamshire Fire and Rescue Services and the two Higher Education Institutions in Nottingham. It will chronicle how a very traditional relationship has been transformed, initially by a simple consultancy project, into a much closer working relationship characterised by a much richer variety of collaborative projects. It demonstrates the potential mutual benefits that greater trust and reciprocity between the institutions can bring to both academia and to practice and the impact it has already had on curriculum development, teaching and learning in Nottingham

The role of CDK4/6 inhibitors in breast cancer
Oral inhibitors of CDK4/6 have been shown to increase response rates and prolong disease control when combined with endocrine therapy in hormone-responsive (HR+) HER2-negative advanced breast cancer. Palbociclib, ribociclib and abemaciclib are all approved in combination with non-steroidal aromatase inhibitors in first-line therapy for post-menopausal women, with a 40–45% improvement in progression-free survival seen with the addition of any of these CDK4/6 inhibitors. Additional approved indications, including first- and second-line combination therapy for pre-menopausal women, combination with fulvestrant and use as monotherapy, vary with each agent and are reviewed fully in the subsequent texts. These agents also differ in their toxicity profiles and monitoring requirements, and prescribers should be aware of the individual requirements for each agent. Current clinical trials are investigating the expanded use of these agents in other breast cancer subtypes, such as HER2-positive and triple-negative breast cancer, as well as in the adjuvant and neoadjuvant treatments of early breast cancer. Resistance to CDK4/6 inhibition can occur through multiple mechanisms. Rational combinations with other therapies, such as PI3K inhibitors, HER2-directed therapies and immunotherapy, are being explored
Mode-II fracture behaviour of aerospace-grade carbon fibre/epoxy composites interleaved with thermoplastic veils
Thermoplastic veils based on Polyethylene-terephthalate (PET), Polyphenylene-sulfide (PPS) and Polyamide-12 (PA) fibres (∼10μm in diameter) were used to interlay unidirectional (UD), non-crimp fabric (NCF) and 5-Harness satin weave (5H) carbon fibre laminates. The PET and PPS veils remained in a fibrous form and the PA veils melted during the laminate curing process. The results of an end-loaded split test demonstrated significant improvements in the mode-II fracture performance in all cases. In general, interlaying thermoplastic veils was most efficient for toughening the UD laminates, with reduced improvements observed for the 5H and NCF laminates, respectively. The main toughening mechanism of the intact PET and PPS veils was thermoplastic fibre bridging. The melted PA veils mainly improved the fracture toughness of the epoxy at the mid-plane. The different toughening mechanisms of the veils, combined with different fracture mechanisms between the UD, NCF and 5H laminates, resulted in significantly different toughening levels

Rather than reducing Brecht's use of the Bible to the purely satirical, the author interprets the full dramatic function of Brecht's complex use of scripture. Using examples from plays written throughout the span of Brecht's career, Murphy shows how Brecht invokes the stories of Old Testament figures such as Job and Isaiah as well as the crucifixion accounts of the New Testament in order to build sympathetic characters and explore his more political themes
