En el siguiente documento, se verán consignadas las experiencias adquiridas por el estudiante en cuestión, quien optó por realizar las prácticas profesionales en la empresa EMTELCO CX & BPO, la cual es una compañía enfocada en el sector de Contac center y brinda ayuda a diferentes clientes quienes contratan sus servicios para brindar asesorías en las áreas específicas de cada uno de los mismos. A su vez, se verá reflejado cómo se puso en práctica los conocimientos adquiridos a lo largo de la carrera y cómo estos, ayudaron a realizar aportes mientras se sorteaban y superaban obstáculos que se presentaban en las diferentes ramas del cargo.
También, se verá como el estudiante logró tener un acercamiento por medio de su cargo a grandes compañías nacionales y otras internacionales las cuales conforman el gran portafolio de EMTELCO, siendo algunas de ellas como el grupo éxito, Keralty, Belcorp y un sinfín de compañías que permitieron expandir el horizonte del estudiante a la vez que lo guiaban por el mismo.In the following document, the experiences acquired by the student in question, who chose to perform the professional practices in the company EMTELCO CX & BPO, which is a company focused on the contact center sector and provides assistance to different customers who hire their services to provide advice in specific areas of each of them, will be recorded. Unveiling the significance of His160 residue in CK2α-CX-4945 interaction with implications for innovative diabetes treatment
reservedDiabetes mellitus is a chronic metabolic disorder characterized by disturbances in the body's sugar metabolism due to inadequate insulin production or utilization. Current therapeutic approaches focus on management rather than a cure necessitating the pursuit of novel treatments. Understanding the molecular mechanisms of the disease may aid innovative therapy development. The “Diabetic Kinome” encompasses protein kinases including DYRK1A, GSK3β, and CK2 responsible for regulating functions involved in diabetes. Targeted inhibition of these kinases shows potential as an alternative treatment, although this approach is still under investigation. Challenges arise due to the significant similarity in the kinase domains of diabetic kinome members, which requires meticulous design of highly specific inhibitors. Precision in this regard is crucial for minimizing off-target effects and promoting the advancement of effective anti-diabetic treatment. CK2, also known as Casein Kinase 2, is a tetrameric enzyme consisting of two catalytic subunits (CK2α and CK2α') and two regulatory subunits (CK2β). It is a key regulator of various cellular processes. Additionally, the recent studies have indicated an involvement of this particular kinase in the endocrine functions of pancreatic β-cells. Therefore, CK2 becomes one of the molecular targets in the treatment of diabetes. One of the most renowned CK2 inhibitors is CX-4945, a potent ATP-competitive inhibitor. However, this inhibitor also binds to other kinases within the diabetic kinome such as DYRK1A, making it challenging to attribute the observed effect to a specific kinase. Further investigation is essential to elucidate the intricate interaction mechanism between the inhibitor and the kinases. The analysis of the available CK2α-CX-4945 and DYRK1A-CX-4945 structures indicated a crucial role of the His160 residue in CK2α, in facilitating the unique inhibitor-kinase interaction.
The exact role of His160 was further examined in the presented thesis. Two mutants of His160 (Ala and Glu) were created using site-directed mutagenesis, expressed in E. coli, and subsequently purified. The thermal shift assay analysis confirmed that introduced changes did not affect the stability of the protein. Moreover, these alterations did not affect the enzymatic activity, and the analysed CK2α variants remained equally active, as demonstrated in the ADP-GLO assay. Substitution of His160 by Ala or Glu affected the kinase-CX-4945 interaction. The strongest inhibition was observed for the wild-type protein, while both mutants (H160A and H160E) displayed higher IC50 values. Then, crystallization trials were conducted, yielding crystals of three CK2α kinase variants (WT, H160A, and H160E) in complex with the CX-4945 inhibitor. This will enable the explanation of the observed differences in the interaction between CK2α variants and CX-4945 at the molecular level.
These results emphasize His160 residue role in CK2α-CX-4945 interaction, guiding the design of potent and selective inhibitors. These findings hold promise for design innovative ATP-competitive inhibitors tailored for CK2α, advancing potential therapies for diabetes treatment
Optical duobinary transmission system featuring improved receiver sensitivity and reduced optical bandwidth
We propose and demonstrate a simple method to improve both the receiver sensitivity and spectral efficiency of the optical duobinary transmission system. Compared to the conventional duobinary signal generated by using an electrical low-pass filter, more than 1-dB improvement of receiver sensitivity and similar to20% reduction in the spectral width are achieved simply by filtering the 10-Gb/s duobinary signals with a narrow-bandwidth (7 GHz) optical filter
Characterization of East London Culex pipiens s.l. in relation to the risk of transmission to humans of the West Nile virus in Great Britain.
PhDConcerns that West Nile Virus (WNV) may arrive in the UK, prompted investigations
of Cx. pipiens s.l., as this species complex has been most often implicated in European
urban WNV human outbreaks and the rapid spread of the virus across North America
from 1999.
Two members of the complex are present in Britain, Cx. p. pipiens and Cx. p. molestus.
These distinct biotypes do not interbreed and are respectively ornithophilic and
anthropophilic. Across Europe, these traits vary with latitude, presenting a major
taxonomic problem.
Research was conducted in an urban area of East London in and around the Beckton
Sewage Treatment Works (STW), where Cx. p. molestus was a nuisance. Most of the
study was conducted using a DNA based assay to distinguish the forms, which was first
fully evaluated against the typical characteristics normally used for identification
Temporal and spatial surveys of larval breeding sites indicated that Cx. p. molestus
breeds all year round inside artificial enclosures both above and below ground in a
range of both contained and non-contained pools varying widely in size, depth and
water quality.
Cx. p. molestus larvae were not found outside of enclourses even in the summer.
However, adults were found biting humans. Hence, Cx. p. molestus appears to move
from the Beckton STW to obtain its blood meals and returns.
Despite a search of all samples no hybrids were found. The two biotypes did not seem
to form hybrids confirming them as essentially separated.
The biting specificity of the two biotypes was studied in an urban farm where human,
animals and birds live in close contact. While Cx. p. pipiens was found to be exclusively
ornithophilic, Cx. p. molestus resulted to be anthropophilic and capable to take 5.5% of
its blood meals from birds, thus being a potential bridge vector for the transmission of the WNV to humans in Britain
