Silver stain after isoelectric focusing of unconcentrated cerebrospinal fluid: Visualization of total protein and direct immunofixation of immunoglobulin G
A silver stain technique has been developed to study proteins in unconcentrated cerebrospinal fluid (CSF) after isoelectric focusing. This method is highly sensitive, and bands containing 25 to 50 ng protein can be clearly distinguished, so that small volumes (40 μl maximum) of native CSF can be used. Individual proteins (e.g., immunoglobulin G) can be detected easily by specific immunofixation. It is also possible to perform direct precipitation and direct specific immunofixation on a single gel in order to compare side by side the patterns of the whole and of specific proteins from different samples of CSF. The technique is simple and highly reproducible, and results are obtained 6 h after sample deposition (if immunofixation is used, a further 24 h washing is necessary). The sensitivity and versatility of this technique (immunofixation can be applied to the detection of any antigen) should permit its extension to other biological fluids with a low protein content
Effects of methimepip and JNJ-5207852 in Wistar rats exposed to an open-field with and without object and in Balb/c mice exposed to a radial-arm maze
The role of the histamine H3 receptor (H3R) in anxiety is controversial, due to limitations in drug selectivity and limited validity of behavioral tests used in previous studies. In the present report, we describe two experiments. In the first one, Wistar rats were treated with an H3R agonist (methimepip), and exposed to an open-field. In the second one, Balb/c mice were treated with H3R agonist (methimepip) or antagonist (JNJ-5207852), and exposed to an open space 3D maze which is a modified version of the radial-arm maze. C57BL/6J saline treated mice were included for comparisons. When exposed to an empty open field, Wistar rats spent more time in the outer area and made very low number of brief crossings in the central area. However, when an object occupied the central area, rats crossed frequently into and spent a long time in the central area. Administration of a range of different doses of methimepip (selective H3R agonist) reduced the entries into the central area with a novel object, indicating enhanced avoidance response. In the 3D maze, both Balb/c and C57BL/6J saline-treated mice crossed frequently onto the bridges that radiate from the central platform but only C57BL/6J mice crossed onto the arms which extend the bridges. This suggests that Balb/c mice are more anxious than C57BL/6J mice. Neither methimepip nor JNJ-5207852 (selective H3R antagonist/inverse agonist) induced entry into the arms of the maze, indicative of lack of anxiolytic effects

Histamine H3 and H4 receptors are expressed on distinct endocrine cell types in the rat fundic mucosa
The present study was designed to explore the expression and distribution of histamine H3 and H4
receptors within the rat gastric fundus by immunohistochemistry using our unique immunological
probes. Groups of rats were either fed ad libitum or fasted for 24h before the sacrifice. Tissue
samples from the fundic region of the stomach were removed, fixed and processed for
immunohistochemistry, using our validated anti-H3 (349-358) and anti–H4 (374-390) antibodies.
The results showed that cells immunoreactive for H3R and H4R are located in the fundic epithelium.
The vast majority of H3R immunopositive cells occupied the lower half of the glands, and were
absent in the superficial portion. Their number per gland was 5.38 ± 0.19 in freely fed rats and 3.21
± 0.40 in fasted rats. Conversely, cells immunoreactive for H4R were dispersed throughout the
glands. Their number per gland was 0.75 ± 0.03 in fed rats and 0.90 ± 0.05 in fasted rats. Double
immunostaining revealed that the H3R is not colocalized with the H4R, while both H3R and H4R
positive cells were immunoreactive for chromogranin A, suggesting that these immunopositive cells
are endocrine cells. Because five different endocrine cell types have been identified in the rat fundic
epithelium, we used double immunostaining to identify the cell types expressing H3R and H4R,
respectively. Approximately 90% of cells positive for H3R were also immunoreactive for histidine
decarboxylase, demonstrating that the H3R is located on ECL cells. Cells positive for H4R were
immunoreactive for ghrelin, originating from A-like cells.
These findings may imply that histamine, via a balanced interaction with H3R and H4R expressed
on ECL and A-like cells respectively, regulates not only acid secretion and mucosal protection but
could also have a role in feeding behaviour and growth hormone release

Oxygen isotope and trace element compositions of peridotite xenolitlis from Nushan volcano, SE China and implications for mantle metasomatism
Oxygen isotopic compositions and trace element contents of minerals from 9 peridotite xenoliths hosted by Cenozoic basalts\ in Nushan, SE China have been obtained by laser fluorination method and LA-ICP-MS respectively. Some samples dont show oxygen isotope equilibrium between pyroxene and olivine with the lowest Delta(Cpx)(18)O ( = delta(18)O(Cpx)-delta(18)O(OI)) of -0.5 parts per thousand. Clinopyroxenes from the amphibole-bearing xenoliths are enriched in LILE (Sr, Th, LREE) and depleted in HFSE (Nb, Zr, Ti). Oxygen isotope data suggest that the Nushan upper mantle experienced metasomatism by oceanic fluids from the subducted plate. Based on the negative correlation between Delta(Cpx-01)(18)O and formation depth of Nushan xenoliths, we propose an "ascending + buffering" model for mantle metasomatism in Nushan. The degree of oxygen isotopic disequilibrium is not related with LILE enrichment and HFSE depletion, suggesting at least two metasomatic events happened in the Nushan mantle, one was responsible to LILE enrichment/HFSE depletion and another to oxygen isotopic disequilibrium

The authors present a new technique for the analysis of the cerebro spinal fluid (C.S.F.) proteins: silver stain after isoelectric focusing with possibility of immunofixation. This technique is simple and results are obtained after six hours only after sample deposition. If immunofixation is used a further 24 hours washing is necessary. It is highly sensitive and bands containing 25 ng of proteins can be clearly distinguished so that small volumes (10 microl. at mean) of unconcentrated C.S.F. can be used. With this technique abnormal distinct bands of IgG specificity are seen in the majority of central and peripheral nervous system inflammatory disorders. No distribution of these bands, particularly in the more cathodal zone, seems to be specific of any disease including multiple sclerosis (M.S.). The abnormal IgG bands in M.S.C.S.F. seem to have their counterpart in the serum. These results need further verification. The usefulness of this technique for research purposes is stressed. The protein pattern obtained must be interpreted in the light of the concepts of genetic polymorphism, microheterogeneity and heterogeneous mode of antibody production
