    Karakterisering en functionaliteit van drafproteïnen en hun enzymatische hydrolysaten

    Draf (of bierbostel) is een belangrijke bijstroom die verkregen wordt tijdens de productie van bier. Hoewel draf rijk is aan proteïnen en dieetvezels, wordt het voornamelijk gebruikt in de veevoeding. Vandaag de dag is er een toenemende vraag naar alternatieven om draf (en zijn hoofdbestanddelen) op te waarderen. Het doel van deze thesis is een wetenschappelijke basis te leggen voor de valorisatie van de proteïnen uit draf met behulp van enzymtechnologie. Aangezien weinig informatie in de literatuur beschikbaar is over de proteïnen uit draf en hun interacties met andere polymeren, werden de drafproteïnen allereerst grondig gekarakteriseerd. Proteïnen uit gerst, mout en draf werden gefractioneerd op basis van hun sequentiële extraheerbaarheid in verschillende media en de fracties werden vervolgens karakteriseerd met behulp van SDS-PAGE en HPLC. De prolaminefractie uit gerst (hordeïnen genoemd) bestaat uit monomere en polymere proteïnen, waarbij de laatstgenoemden gestabiliseerd worden door intermoleculaire disulfidebindingen. Monomere C-hordeïnen en enkele B-hordeïnen werden uit gerst geëxtraheerd onder niet-reducerende voorwaarden, terwijl de meeste B-, C- en D-hordeïnen geëxtraheerd werden onder reducerende voorwaarden. Tijdens het mouten worden disulfidebindingen gereduceerd en worden B- en D-hordeïnen gedegradeerd door proteolytische enzymen. Ongeveer gelijkaardige gehaltes B-hordeïnen werden verkregen na extractie onder zowel niet-reducerende als reducerende voorwaarden en bijna geen D-hordeïnen werden geëxtraheerd uit mout. B-hordeïnen werden enkel geëxtraheerd uit draf onder reducerende voorwaarden. Hoewel de meeste C-hordeïnen uit draf werden geëxtraheerd onder niet-reducerende voorwaarden, werden meer C-hordeïnen geëxtraheerd onder reducerende voorwaarden. Tijdens het maïschen worden disulfidebruggen gevormd met aggregatie als gevolg. Hoewel literatuur suggereert dat dit aggregaat is samengesteld uit B- en D-hordeïnen (en glutelinen), wordt in dit werk het aggregaat voorgesteld als B-hordeïnen waarin C hordeïnen kunnen ingesloten zitten. De proteïnen uit draf zijn grotendeels onoplosbaar waardoor ze weinig functionaliteit vertonen. Via enzymatische hydrolyse van deze proteïnen werd getracht proteïnehydrolysaten uit draf met verbeterde oplosbaarheid en technofunctionele eigenschappen te verkrijgen. Alkalische extractie gevolgd door precipitatie van de gesolubiliseerde proteïnen resulteerde in een proteïnerijk materiaal (verder Brewers’ spent grain Protein Concentrate of BPC genoemd). De aminozuursamenstelling van de proteïnen uit draf en BPC was gelijkaardig wat suggereert dat draf en BPC uit dezelfde proteïnen zijn samengesteld. BPC werd vervolgens gehydrolyseerd via de pH-stat methode met verschillende peptidasen gedurende verschillende hydrolysetijden en met variërende enzymconcentraties. Hierdoor werden hydrolysaten met verschillende hydrolysegraden (DH) verkregen. Naast de molecuulgewicht (MW) verdeling en de (oppervlakte)hydrofobiciteit, werden eveneens de oplosbaarheid, kleur en emulgerend en schuimvormend vermogen bepaald. Enzymatische hydrolyse van BPC verhoogde de oplosbaarheid en verbeterde de emulgerende en/of schuimvormende eigenschappen. Voor de hydrolysaten verkregen na hydrolyse met Alcalase en pepsine, leidde een stijgende DH echter tot verminderde emulgerende en schuimvormende eigenschappen. Bovendien blijken de eigenschappen van het gebruikte enzym een duidelijk effect te hebben op de uiteindelijke technofunctionaliteit van het hydrolysaat. De hydrolysaten verkregen na enzymatische hydrolyse met Flavourzyme vertoonden goede technofunctionele eigenschappen, onafhankelijk van de DH. Fysicochemische karakterisering van de hydrolysaten duidde het belang aan van proteïnefragmenten met relatief hoog MW (hoger dan 14.5 k) en hoge oppervlaktehydrofobiciteit voor gunstige technofunctionele eigenschappen. De emulgerende en schuimvormende eigenschappen van de BPC hydrolysaten waren gelijkaardig (of hoger) in vergelijking met deze van commerciële proteïnehydrolysaten. De BPC hydrolysaten waren echter donkerder dan de commerciële producten. BPC hydrolysaten waren samengesteld uit een heterogeen mengsel van peptiden met variërende hydrofobiciteit met zowel laag MW als hoog MW. Daarom werd getracht door fractionering van de hydrolysaten op basis van hun MW verdeling en/of hydrofobiciteit meer homogene peptidemengsels te verkrijgen. Twee hydrolysaten met hoge en lage DH, verkregen na enzymatische hydrolyse met Alcalase op ‘grote schaal’, werden gefractioneerd met ethanol en ammoniumsulfaat. De MW verdeling, oppervlaktehydrofobiciteit, aminozuursamenstelling en emulgerende en schuimvormende eigenschappen van deze fracties werden bepaald. Stijgende ethanol- en ammoniumsulfaatconcentraties resulteerden in proteïnefragmenten met afnemende MW verdeling en oppervlaktehydrofobiciteit. Het was duidelijk dat de proteïnefragmenten met goede emulgerende eigenschappen andere fysicochemische eigenschappen vertoonden dan deze met goede schuimvormende eigenschappen. Het emulgerend en schuimvormend vermogen van BPC hydrolysaten werd niet uitsluitend door de MW verdeling of door de oppervlaktehydrofobiciteit bepaald. Deze hydrolysaten vertoonden gunstige emulgerende eigenschappen wanneer tenminste 50 % van de peptiden een MW tussen 1.7 k en 14.5 k hadden. Bovendien hadden deze proteïnefracties een hoge oppervlaktehydrofobiciteit. BPC hydrolysaten met goede schuimvormende eigenschappen bevatten minder dan 40 % van hun proteïnefragmenten met MW onder 1.7 k. De oppervlaktehydrofobiciteit van deze hydrolysaten was laag, behalve voor de proteïnehydrolysaten waar het gehalte aan proteïnefragmenten met MW boven 14.5 k hoger was dan proteïnefragmenten met MW tussen 1.7 k en 14.5 k. Inhibitie van het angiotensin I-converterend enzym (ACE) kan een bloeddrukverlagend effect veroorzaken. De ACE inhiberende activiteit van BPC hydrolysaten, verkregen na enzymatische hydrolyse met verschillende enzymen gedurende verschillende hydrolysetijden, was vergelijkbaar met deze van andere plantproteïnehydrolysaten, beschreven in de literatuur. De actieve componenten werden gedeeltelijk opgezuiverd met behulp van zowel ultrafiltratie als chromatografische technieken. Dit leverde BPC peptidefracties op met meer dan zes maal hogere ACE inhiberende activiteit in vergelijking met het hydrolysaat. Als alternatief werden ACE inhiberende peptiden opgezuiverd met een chromatografische methode gebaseerd op affiniteit. Hiervoor werd een longextract rijk aan ACE geïmmobiliseerd op een N-hydroxysuccinimide geactiveerde Sepharose 4 Fast Flow matrix en werden de peptiden die bonden aan ACE specifiek van de kolom geëlueerd met captopril, een gekende ACE inhibitor. Aangezien captopril samen met de gebonden BPC peptiden elueert, werden deze peptiden verder opgezuiverd met behulp van reversed phase HPLC. Deze procedure kan echter nog verder geoptimaliseerd worden. Op deze manier werden mengsels van peptiden verkregen met meer dan 2 000 keer hogere ACE inhiberende activiteit dan de BPC hydrolysaten. Dit mengsel bevatte een peptide met een MW van 432, bepaald met behulp van massaspectrometrie (ESI-MS). De aminozuursequentie van dit peptide was Cys-Asn-Cys-Pro.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
