    Characteristics of human monocytes activation by monophosphoryl lipid A

    Le lipopolysaccharide (LPS), un agoniste du Toll-like récepteur 4 (TLR4) est un puissant immunomodulateur dont l’utilisation est limitée en pratique clinique par sa toxicité. Le monophosphoryl lipide A (MPLA) est un dérivé non toxique du LPS commercialisé comme adjuvant vaccinal et d’intérêt majeur dans de nombreux domaines. Les mécanismes sous-jacents aux propriétés non-toxiques et immunomodulatrices du MPLA sont mal caractérisés chez l’homme. Un modèle in vitro de monocytes humains de donneurs sains nous a permis de définir les caractéristiques spécifiques de l’activation par le MPLA. Le MPLA induit une production moindre de cytokines proinflammatoires (interleukin-1ß-IL-1ß et tumor necrosis factor-TNF) que les LPS. La production de cytokines anti-inflammatoires telles que l’IL-10 (interleukin-10) et l’IL-1Ra (IL-1 receptor antagonist) n’est pas significativement différente. Pour un même donneur, les productions respectives d’IL-1ß et TNF après stimulation par LPS ou MPLA ne sont pas corrélées, cela suggère une activation de voies de signalisation différentes par chaque agoniste. L’utilisation d’inhibiteurs du TLR4 (LPS Rhodobacter spheroides et Eritoran) a démontré que le MPLA et les LPS sont dépendant du TLR4. En revanche, contrairement au LPS, le MPLA ne nécessite pas le corécepteur CD14 pour induire une production de cytokines. L’utilisation d’inhibiteurs de polymérisation de l’actine (cytochalasine D et latrunculine A) a démontré la dépendance du MPLA aux mécanismes d’endocytose actine-dépendants pour la production de cytokines. Après stimulation par le LPS, une dépendance partielle à l’endocytose actine-dépendante est observée pour la production d’IL-1ß et de TNF dans la sous-catégorie des donneurs forts producteurs. Cela suggère que les voies de signalisation engagées par le MPLA sont intracellulaires et qu’elles sont également activées dans la sous-catégorie des donneurs forts producteurs après exposition au LPS. La voie de signalisation endosomale du TLR4 est la voie TRIF-dépendante. Cette voie de signalisation est partagée par le TLR3. Contrairement à ce qui a été publié sur modèle murin, il n’a pas pu être montré que la signalisation du MPLA dans les monocytes humains est préférentiellement conduite via la voie TRIF-dépendante. En effet, contrairement au Poly I :C, un agoniste du TLR3, le MPLA et les LPS n’induisent pas de CXCL-10 après stimulation. Le fait que la production de CXCL-10 après stimulation par poly I :C soit totalement inhibée par la cytochalasine D suggère cependant des mécanismes communs d’endocytose. La signalisation SYK-dépendante en aval de TLR4 a été impliquée à la fois dans l’endocytose de TLR4 CD14-dépendante mais aussi dans les productions de haut niveau de cytokines proinflammatoires. L’utilisation d’un inhibiteur de la phosphorylation de SYK, R406, n’a révélé aucune différence entre le MPLA et les LPS. Contrairement au modèle murin, les monocytes produisent de l’IL-1ß après une stimulation unique par un agoniste du TLR4, démontrant une capacité d’activation en un temps de l’inflammasome. Une corrélation significative entre la production d’IL-1ß et de TNF est observée après stimulation par MPLA. Cette corrélation n’est pas retrouvée après stimulation par LPS. Ainsi l’activation de l’inflammasome pourrait jouer un rôle-clé dans la signalisation induite par le MPLA. Ceci est conforté par l’utilisation d’un inhibiteur de pan-caspase (Z-VAD) dont l’effet inhibiteur sur la production de TNF est significativement plus élevé pour le MPLA que pour les LPS. En conclusion, la non-toxicité du MPLA se traduit dans les monocytes humains par une production moindre d’IL-1ß et de TNF en rapport avec une signalisation différente de celle engagée par les LPS. Le MPLA nécessite une endocytose du TLR4 actine-dépendante et CD14-indépendante. Le TLR4 pourrait soit engager une signalisation intracellulaire soit être un transporteur pour le MPLA qui activerait lors directement l’inflammasome.Lipopolysaccharide (LPS), a Toll-like receptor 4 (TLR4) agonist is a potent immunomodulator whose use is limited in clinical practice by its toxicity. Monophosphoryl lipid A (MPLA) is a nontoxic derivative of LPS marketed as a vaccine adjuvant and of major interest in many fields. The mechanisms underlying the non-toxicity and immunomodulatory properties of MPLA are poorly characterized in humans. An in vitro model of human monocytes from healthy donors allowed us to define specific characteristics of MPLA activation. MPLA induces a lower production of proinflammatory cytokines (interleukin-1ß-IL-1ß et tumor necrosis factor-TNF) than LPS. The production of anti-inflammatory cytokines such as IL-10 (interleukin-10) and IL-1Ra (IL-1 receptor antagonist) are not significantly different. For a same donor, the respective productions of IL-1ß and TNF after stimulation by LPS or MPLA are not correlated, suggesting an activation of different signaling pathways by each agonist. The use of TLR4 inhibitors (LPS Rhodobacter spheroides and Eritoran) has demonstrated that MPLA and LPS are TLR4 dependent. In contrast, unlike LPS, MPLA does not require the CD14 co-receptor to induce cytokine production. The use of actin polymerization inhibitors (cytochalasin D and latrunculin A) has demonstrated the dependence of MPLA on actin-dependent endocytosis mechanisms for cytokine production. Following LPS stimulation, partial dependence on actin-dependent endocytosis is observed for IL-1ß and TNF production in the high-producers donor subset. This suggests that the signaling pathways initiated by MPLA are intracellular and are also activated in the high-producers donor subset after exposure to LPS. The endosomal signaling pathway of TLR4 is the TRIF-dependent pathway. This signaling pathway is shared by the TLR3. Contrary to what has been published on a mouse model, it has not been possible to show that MPLA signaling is human monocytes is preferentially carried out via the TRIF-dependent pathway. Indeed, unlike poly I:C, a TLR3 agonist, MPLA and LPS do not induce CXCL-10 after stimulation. The fact that the production of CXCL-10 is totally inhibited by cytochalasin D, however, suggests shared mechanisms of endocytosis. SYK-dependent signaling downstream of TLR4 has been implicated in both CD14-dependant TLR4 endocytosis and in high-level productions of proinflammatory cytokines. The use of a SYK phosphorylation inhibitor, R406, revealed no difference between MPLA and LPS. In contrast to the murine model, monocytes produce IL-1ß after a single TLR4 agonist stimulation, demonstrating a one-step inflammasome activation ability. A significant correlation between IL-1ß and TNF production is observed following MPLA exposure. This correlation is not found following LPS exposure. Thus, inflammasome activation could play a key role in MPLA-induced signaling. This is supported by the use of a pan-caspase inhibitor, (Z-VAD) whose inhibitory effect on TNF production is significantly greater for MPLA than for LPS. In conclusion, the non-toxicity of MPLA is reflected in human monocytes by a lower production of IL-1ß and TNF in relation to a signaling different from that initiated by LPS. MPLA requires actin-dependent and CD14-independant TLR4 endocytosis. TLR4 could either initiate intracellular signaling or be a cargo for MPLA which would then directly activate the inflammasome

    Cellules NK et inflammation systémique : compartimentalisation et réponse mémoire

    L'inflammation systémique est une réaction qui implique l’ensemble de l’organisme suite une agression sévère, potentiellement mortelle, illustrée par le syndrome de réponse inflammatoire systémique (SIRS). De nombreux acteurs cellulaires et moléculaires contribuent au développement de cette cascade inflammatoire parmi lesquels les cellules NK jouent un rôle clé. Malgré l'accumulation de preuves sur l’existence de propriétés spécifiques à chaque organe en réponse à l'inflammation systémique, en termes de cellules NK, on sait peu de choses sur la dynamique compartimentalisée de l’activation des cellules NK pendant un SIRS. En outre, le statut immunitaire des cellules NK après la résolution d’un SIRS est également mal connu. Dans le présent travail, nous avons étudié les réponses des cellules NK provenant de différents organes en utilisant un modèle d’endotoxinémie murine. Nous avons caractérisé la réponse des cellules NK au sein de la rate, du poumon, de la moelle osseuse, de la cavité péritonéale, et dans la circulation. Nous avons trouvé que, malgré une dynamique similaire de la réponse dans les différents organes, les réponses des cellules NK sont compartimentalisées avec des seuils différent et spécifiques. A l’aide de transferts adoptifs, nous avons constaté que la réactivité des cellules NK spécifiques d'organes peut refléter le compartiment d’origine lors des phases initiales de l'inflammation. Cependant, les cellules NK ont la capacité de s’adapter rapidement à leur nouvel environnement et d'ajuster leurs niveaux de réponse à ceux des cellules NK résidentes. Ainsi, cette étude fournit une preuve de concept qui confirme la compartimentalisation de la réponse des cellules NK lors de l'inflammation systémique. Dans une deuxième partie, nous avons analysé le statut des cellules NK à différents moments après une endotoxinémie. Les réponses des cellules NK au sein d’une préparation de cellules de la rate sont fortement supprimées en réponse à une restimulation in vitro, 14 jours après l'endotoxinémie. Cependant, nous avons montré que la réactivité intrinsèque des cellules NK est en fait augmentée après l'injection d’endotoxine, aboutissant à des cellules NK présentant des caractéristiques de cellules NK mémoires. Des expériences de transfert adoptif ont confirmé les propriétés de mémoire des cellules NK après endotoxinémie. Nos résultats accroissent la connaissance concernant le rôle des cellules NK dans un contexte d'inflammation systémique, révélant des réponses compartimentalisés et l’induction d’une mémoire suite à l’endotoxinémie. L'observation selon laquelle les cellules NK développent des propriétés de mémoire après une inflammation systémique dans le contexte d'un environnement suppressif est d’une grande nouveauté et ce phénomène est rapporté pour la première fois.Systemic inflammation is whole-body reaction to a triggering insult that often results in life threatening illness like systemic inflammatory response syndrome (SIRS). Contributing to the development of this inflammatory cascade are numerous cellular and molecular players, among which, NK cells have been shown to play a key role. Despite accumulating evidence on the organ-specific properties of both systemic inflammation and NK cells, little is known about the compartmentalized dynamics of NK cell activation during SIRS. Furthermore, the status of NK cells after the resolution of SIRS is also poorly characterized. In the present work, we investigated NK responses in different organs using a mouse model of endotoxinemia and characterized the compartmentalized response of spleen, lung, bone marrow, peritoneal and circulating NK cells. We found that despite similar dynamics of response in different organs, NK cells responses, are compartmentalized with seemingly specific thresholds of maximum activation. Using a series of adoptive transfers, we found that while organ-specific NK cell responsiveness can affect the initial phases of inflammation, these cells have the capacity to quickly adapt to a new environment and adjust their response levels to that of resident NK cells. Thus, this study provides proof of concept data on the compartmentalization of the NK cell responses during systemic inflammation. In a second part, we assessed the status of NK cells at different times after endotoxemia. NK cells responses in the context of whole spleen preparations were severely suppressed in response to in vitro restimulation at 14 days after endotoxemia. However, intrinsic NK cell responsiveness was increased after endotoxemia, showing characteristics of NK cell memory. Adoptive transfer experiments confirmed memory properties of NK cells after endotoxemia. Overall, these results expand on the role of NK cells in the context of systemic inflammation revealing compartmentalized responses during and memory properties following endotoxemia. The observation that NK cells develop memory properties after systemic inflammation in the context of a suppressive environment is of the highest novelty and the first one to report such a phenomenon

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.     “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Free Radicals in Inflammation

    This chapter sheds some light on the enigmatic world of "free radicals" as it relates to human physiology and disease, with particular emphasis on inflammation. Inflammatory processes are generally associated with changes in cellular status toward a more oxidized redox tone compared to a normal cell, a situation characterized by an increased production of reactive oxygen species (ROS) and accompanying "oxidative stress". With the increasing knowledge of free radical species interactions, it has become increasingly clear that ROS, reactive nitrogen species (RNS), and reactive sulfur species (RSS) have broad signaling effects well beyond individual cells. The chapter briefly reviews the properties of some important classes and key representatives and look at their role in the intricate network of regulatory processes that fuel the cellular machinery, allowing cells and tissues/organs to quickly adapt to changes in environmental conditions, energetic and metabolic demand, and nutritional status.</p

    Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods
