Ecofisiologia e biomarcadores de estresse oxidativo na caracterização do ciclo de vida pós-embrionário de uma espécie de peixe anual neotropical
Os peixes de água doce são considerados um dos grupos mais ameaçados do mundo, visto que os ecossistemas aquáticos atualmente estão enfrentando constantes interferências de ações antrópicas, principalmente, com a intensificação da agricultura que tem impactado e descaracterizado diversos habitats de muitas espécies. Frente a isto, para a conservação da diversidade biológica nesses ambientes aquáticos é de extrema importância obter informações sobre as espécies constituintes destes habitats. Desta forma, ferramentas fisiológicas podem ser utilizadas para caracterizar a diversidade biológica e suas implicações ecológicas, e abranger também, as respostas dos organismos às alterações ambientais e agentes estressores. Os peixes anuais são organismos que vivem em áreas úmidas temporárias que secam sazonalmente. Devido a isto, apresentam um ciclo de vida curto e rápido, mostrando um grande potencial para serem utilizados como modelos biológicos para diversas pesquisas, principalmente, aquelas ligadas ao estudo de envelhecimento. O objetivo do presente trabalho foi analisar marcadores fisiológicos relacionados ao balanço redox do peixe anual Cynopoecilus fulgens, endêmico da região sul do Brasil, com variáveis ambientais e estimativas do período reprodutivo. Foram coletados 443 indivíduos compreendendo juvenis, adultos e senis, entre o mês de junho e novembro de 2016, nos arredores do Parque Nacional da Lagoa do Peixe (PNLP), localizado na Planície Costeira Externa do Rio Grande do Sul, em três diferentes unidades amostrais (P1, P2 e P3). Estes animais foram eutanasiados em campo e em laboratório foram medidos (comprimento padrão) e pesados (massa corporal). Após, foram homogeneizados para a realização dos ensaios da Superóxido Dismutase, Catalase, Glutationa Peroxidase e Glutationa S-transferase, e para determinação dos níveis de Lipoperoxidação, através do TBARS, Proteínas Carboniladas e Proteínas Totais. Os resultados mostraram que machos e fêmeas de C. fulgens empregam estratégias fisiológicas diferentes em seu balanço redox durante o ciclo de vida, sendo influenciados pelo período reprodutivo e variáveis ambientais (bióticas e abióticas). Dentre as estratégias, as fêmeas mostram investir bastante na reprodução, principalmente no período de maturação sexual e produção dos ovos, evidenciado pelos níveis mais elevados de TBARS quando comparadas aos machos que parecem investir no crescimento somático e massa corporal. Além disso, as variáveis que apresentaram maiores influências no balanço oxidativo destes animais foram: temperatura, abundância de predador, abundância do peixe anual Austrolebias minuano e abundância de C. fulgens. Também, a população de cada área úmida temporária apresenta respostas fisiológicas distintas frente à adaptação ao seu ciclo de vida, corroborando a influência de algum componente ambiental como modulador deste ciclo. Adicionalmente, foi observado que os animais das poças P2 e P3 se comportam de maneira semelhante entre si apresentando um ciclo mais longo; sugerindo que o sistema antioxidante destes animais apresenta alta eficiência mantendo os níveis de lipoperoxidação e proteínas carboniladas baixos. Enquanto os da P1 têm seu ciclo de vida pós-eclosão mais curto possivelmente devido ao incremento do estresse oxidativo evidenciado pelo aumento da lipoperoxidação. Estes resultados demonstram que a utilização dos peixes anuais neotropicais na pesquisa é altamente promissora, permitindo um estudo aprofundado sobre o envelhecimento e ensaio toxicológico. Além disso, enfatizam a importância do desenvolvimento de estratégias de conservação ambiental para preservar as características das áreas úmidas temporárias, estabelecendo estes animais como espécies sentinelas.Freshwater fish are considered one of the most threatened groups in the world, as aquatic ecosystems are currently facing constant interference from anthropic actions, mainly with the intensification of agriculture that has impacted and de-characterized diverse habitats of many species. The aim of the present study was to analyze physiological markers related to the redox balance of the annual fish Cynopoecilus fulgens, endemic to southern Brazil, with environmental variables and estimates of the reproductive period. A total of 443 individuals were collected, including juveniles, adults and senile, between June to November 2016, around the Parque Nacional da Lagoa do Peixe, located in the Coastal Plain of Rio Grande do Sul, in three sampling units (P1, P2 and P3). After, they were homogenized for the Superoxide Dismutase, Catalase, Glutathione Peroxidase and Glutathione S-transferase assays, and for the determination of the levels of Lipoperoxidation, through TBARS, Carbonylated Proteins and Total Proteins. The results showed that males and females of C. fulgens use different physiological strategies of their redox balance during the life cycle, being influenced by the reproductive period and environmental variables (biotic and abiotic). Among the strategies, females show a great deal of investment in reproduction, especially in the period of sexual maturation and egg production, evidenced by the higher levels of TBARS when compared to males that seem to invest in somatic growth and body mass. In addition, the variables that had the greatest influence on the oxidative balance of these animals were: temperature, abundance of predator, abundance of annual fish Austrolebias minuano and abundance of C. fulgens. Also, the population of each temporary wetland presented different physiological responses to the adaptation to their life cycle, corroborating the influence of an environmental component as a modulator of this cycle. Additionally, it was observed that the animals of the pools P2 and P3 behave in a similar way among each other presented a longer cycle; suggesting that the antioxidant system of these animals presents high efficiency keeping the levels of lipoperoxidation and carbonylated proteins low. Fish of P1 have their shorter post-hatch life cycle possibly due to increased oxidative stress evidenced by increased lipoperoxidation. These results demonstrate that the use of Neotropical annual fish in the research is highly promising, allowing in-depth study on aging and toxicology testing. In addition, they emphasize the importance of developing environmental conservation strategies to preserve the characteristics of temporary wetlands, establishing these animals as sentinel species
Efeitos adversos de herbicidas à base de glifosato e de ácido diclorofenoxiacético (2,4D) em diferentes estágios de desenvolvimento pós-eclosão de peixes anuais
Os ambientes aquáticos são constantemente expostos e suscetíveis a entrada de agroquímicos. Estes são aplicados em culturas para o controle e eliminação de pragas, visando o aumento da produtividade. Entretanto, quando presentes no meio ambiente podem alcançar ecossistemas aquáticos e prejudicar espécies não-alvo. Os agrotóxicos mais utilizados no Brasil são os herbicidas, destacando-se os à base de glifosato e o ácido 2,4-Diclorofenoxiacético (2,4-D), que são aplicados em diferentes culturas e podem apresentar risco aos habitats ocupados por peixes anuais que estão próximos a estas plantações. Cabe ressaltar que estes animais apresentam uma distribuição restrita, ocupando brejos, poças e lagoas rasas formadas pelas chuvas, mas que secam anualmente no período de estiagem. Além disso, são considerados modelos potenciais para estudos em laboratório, visto que apresentam um ciclo de vida curto e rápido que está intimamente relacionado ao local que vivem. Para isso, requerem características específicas para sobreviver como um crescimento acelerado, um intenso processo reprodutivo e grande deposição de ovos como uma estratégia para manutenção da população em ambiente natural. Dessa forma, buscamos avaliar o efeito dos herbicidas Roundup Original© (princípio ativo glifosato nas concentrações de 65 μg/L, 130 μg/L e 260 μg/L) e Dez© (princípio ativo ácido 2,4-Diclorofenoxiacético (2,4-D) nas concentrações de 4 μg/L, 15 μg/L e 30 μg/L) em dois estágios de desenvolvimento (adulto-jovem e senil) e em ambos os sexos do peixe anual Cynopoecilus sp. Analisamos marcadores de balanço oxidativo e de metabolismo intermediário em animais coletados em campo e transportados até o laboratório para as experimentações. Estes foram medidos, aclimatados por 10 dias e após, expostos a cada um dos herbicidas por sete dias, em todas as etapas os peixes foram alimentados diariamente com ração comercial. Parâmetros abióticos (pH, O2 dissolvido, sólidos totais dissolvidos, condutividade de temperatura da água) foram controlados. Após a exposição, os animais foram medidos e crioeutanasiados (nitrogênio líquido) para as análises dos marcadores de balanço oxidativo e de metabolismo intermediário. Para as análises de balanço oxidativo foram determinados os níveis de Lipoperoxidação (TBARS) e Proteínas Totais (PT), além da quantificação da atividade das enzimas antioxidantes Superóxido Dismutase (SOD) e Catalase (CAT), e da enzima de biotransformação Glutationa S-Transferase (GST). Já para o metabolismo foram realizadas a determinação dos níveis de Glicogênio (GG), Proteínas Totais (PT) e Ácido Úrico (AU) dos animais. Todos os resultados obtidos foram analisados no programa SPSS Statistics com os devidos testes estatísticos. Os resultados estão organizados em três artigos. O primeiro artigo intitulado “Development stage-dependent oxidative stress responses to the exposure to Roundup Original© in a neotropical annual killifish”, publicado na Revista Environmental Toxicology and Pharmacology, onde são apresentados os resultados obtidos com a exposição ao herbicida Roundup Original© de machos adultos-jovens e senis. Neste capítulo, observamos que a capacidade do sistema antioxidante de machos de Cynopoecilus sp. é eficiente nas duas fases do desenvolvimento biológico analisadas impedindo a ocorrência de mortalidade. Apesar disto, os senis apresentaram níveis 2 vezes maiores de peroxidação lipídica estando estes associados à manutenção dos níveis de atividade das enzimas antioxidantes (SOD e CAT). Cabe ressaltar que os indivíduos senis apresentaram uma diminuição da atividade da glutationa S-transferase o que pode afetar negativamente estes indivíduos possivelmente reduzindo sua capacidade reprodutiva e/ou seu ciclo de vida. Neste capítulo também mostramos que os marcadores fisiológicos estabelecidos, bem como a espécie, parecem promissores para serem usados em estudos futuros de senescência e toxicologia. O segundo artigo intitulado “Toxic effect of a Glifosate-Based herbicide associated with different developmental stages in neotropical annual killifish females”, está submetido a Revista Environmental Toxicology and Pharmacology. Neste capítulo nós avaliamos os efeitos do herbicida Roundup Original© (princípio ativo glifosato: 65, 130 e 260μg/L) sobre o balanço oxidativo e marcadores do metabolismo intermediário em diferentes estágios de desenvolvimento pós-eclosão (adulto-jovem e senil) em fêmeas de Cynopoecilus sp. Esses marcadores incluíram lipoperoxidação (TBARS), superóxido dismutase (SOD), catalase (CAT), glutationa S-transferase (GST), relação SOD/CAT, proteínas totais (PT), glicogênio (GG) e ácido úrico (AU). Fêmeas senis apresentaram aumento de atividade da CAT e GST, uma diminuição da relação SOD/CAT, combinado com um aumento de PT e AU quando comparados aos valores do grupo controle. A análise de variância de duas-vias mostrou uma diferença significativa entre faixas etárias e grupos para vários biomarcadores sugerindo uma maior sensibilidade das fêmeas ao glifosato. Novamente, o impacto deste herbicida parece ser mais pronunciado em animais senis. Por fim, no terceiro artigo intitulado “Análise do efeito do composto químico DEZ© (2,4-diclorofenoxiacético) sobre o ciclo de vida do peixe anual neotropical”, a ser submetido a Revista Chemosphere, aborda sobre a exposição de machos e fêmeas nos dois estágios de desenvolvimento expostos ao Dez© (4 μg/L, 15 μg/L e 30 μg/L). Foram realizadas análises em parâmetros de estresse oxidativo e metabolismo intermediário após a exposição ao herbicida de fêmeas e machos da mesma espécie. Nossos resultados mostraram que o estágio de desenvolvimento de maior vulnerabilidade tanto para machos como para fêmeas foi na fase senil, principalmente, em relação aos parâmetros oxidativos. As concentrações mais elevadas de 15 e 30μg/L foram as mais prejudiciais a espécie determinando níveis elevados de TBARS, indicando dano oxidativo. Além disso, as fêmeas de Cynopoecilus sp. parecem ser mais suscetíveis ao princípio ativo 2,4D do que os machos, visto que foram observados um maior número de alterações nos marcadores bioquímicos tanto de metabolismo como de balanço oxidativo. Mesmo não sendo considerada uma espécie-alvo para estes agrotóxicos, Cynopoecilus sp. é impactada com a exposição a concentrações ambientalmente relevantes destes herbicidas. Outro aspecto a se destacar, é que tanto o glifosato como o 2,4D estão presentes em diferentes formulações comerciais no Brasil, sendo estes os agrotóxicos mais vendidos em nosso país. Dessa forma, sugerindo maiores restrições no seu uso principalmente, em locais próximos aos ocupados por peixes anuais, visto que muitas espécies se encontram em algum grau de ameaça. Tanto o conjunto de biomarcadores escolhido como a espécie se mostraram adequados e promissores para o desenvolvimento de ensaios toxicológicos de caráter ontogenético, evidenciando uma sensibilidade diferenciada, especialmente para os senis; tais aspectos devem ser considerados em estudos de impacto de xenobióticos sobre estes grupos.Aquatic environments are constantly exposed and susceptible to the entry of agrochemicals. These are applied to crops to control and eliminate pests, aiming at increasing productivity. However, when present in the environment, they can reach aquatic ecosystems and harm non-target species. The most widely used pesticides in Brazil are herbicides, especially those based on glyphosate and 2,4-Dichlorophenoxyacetic acid (2,4-D), which are applied to different crops and may pose a risk to habitats occupied by annual fish. that are close to these plantations. It should be noted that these animals have a restricted distribution, occupying marshes, puddles and shallow lakes formed by the rains, but which dry up annually during the dry season. In addition, they are considered potential models for laboratory studies, since they have a short and fast life cycle that is closely related to the place they live. For this, they require specific characteristics to survive, such as accelerated growth, an intense reproductive process and large egg laying as a strategy for maintaining the population in a natural environment. Thus, we sought to evaluate the effect of the herbicides Roundup Original© (active ingredient glyphosate at concentrations of 65 μg/L, 130 μg/L and 260 μg/L) and Dez© (active ingredient 2,4-Dichlorophenoxyacetic acid (2,4 -D) at concentrations of 4 μg/L, 15 μg/L and 30 μg/L) in two developmental stages (young-adult and senile) and in both sexes of the annual killifish Cynopoecilus sp. We analyzed markers of oxidative balance and intermediary metabolism in animals collected in the field and transported to the laboratory for experimentation. These were measured, acclimatized for 10 days and after, exposed to each of the herbicides for seven days, in all stages the fish were fed daily with commercial feed. Abiotic parameters (pH, dissolved O2, total dissolved solids, water temperature conductivity) were controlled. After exposure, the animals were measured and cryoeuthanized (liquid nitrogen) for analyzes of markers of oxidative balance and intermediary metabolism. For the analyzes of oxidative balance, the levels of Lipoperoxidation (TBARS) and Total Proteins (PT) were determined, in addition to the quantification of the activity of the antioxidant enzymes Superoxide Dismutase (SOD) and Catalase (CAT), and of the biotransformation enzyme Glutathione S-Transferase (GST). As for the metabolism, the determination of the levels of Glycogen (GG), Total Proteins (PT) and Uric Acid (AU) of the animals were carried out. All results obtained were analyzed using the SPSS Statistics program with the appropriate statistical tests. The results are organized into three articles. The first article entitled “Development stage-dependent oxidative stress responses to the exposure to Roundup Original© in a neotropical annual killifish”, published in the journal Environmental Toxicology and Pharmacology, where the results obtained with exposure to the herbicide Roundup Original© of males are presented young and senile adults. In this chapter, we observed that the capacity of the antioxidant system of males of Cynopoecilus sp. it is efficient in the two phases of biological development analyzed, preventing the occurrence of mortality. Despite this, the senile showed 2 times higher levels of lipid peroxidation, which are associated with the maintenance of activity levels of antioxidant enzymes (SOD and CAT). It should be noted that senile individuals showed a decrease in glutathione S-transferase activity, which may negatively affect these individuals, possibly reducing their reproductive capacity and/or their life cycle. In this chapter we also show that the established physiological markers, as well as the species, seem promising to be used in future studies of senescence and toxicology. The second article entitled “Toxic effect of a Glifosate-Based herbicide associated with different developmental stages in neotropical annual killifish females”, is submitted to Revista Environmental Toxicology and Pharmacology. In this chapter we evaluated the effects of Roundup Original© herbicide (active ingredient glyphosate: 65, 130 and 260μg/L) on the oxidative balance and intermediary metabolism markers at different stages of post-hatching development (young-adult and senile) in females from Cynopoecilus sp. These markers included lipoperoxidation (TBARS), superoxide dismutase (SOD), catalase (CAT), glutathione S-transferase (GST), SOD/CAT ratio, total protein (PT), glycogen (GG) and uric acid (AU). Senile females showed increased CAT and GST activity, a decrease in the SOD/CAT ratio, combined with an increase in PT and UA when compared to control group values. Two-way analysis of variance showed a significant difference between age groups and groups for several biomarkers suggesting a greater sensitivity of females to glyphosate. Again, the impact of this herbicide appears to be more pronounced in senile animals. Finally, in the third article entitled “Analysis of the effect of the chemical compound DEZ© (2,4-Dichlorophenoxyacetic) on the life cycle of neotropical annual killifish”, to be submitted to Chemosphere Magazine, addresses the exposure of males and females in two developmental stages exposed to Dez© (4 μg/L, 15 μg/L and 30 μg/L). Analyzes were performed on parameters of oxidative stress and intermediary metabolism after exposure to the herbicide in females and males of the same species. Our results showed that the most vulnerable stage of development for both males and females was in the senile phase, mainly in relation to oxidative parameters. The highest concentrations of 15 and 30μg/L were the most harmful to the species, determining high levels of TBARS, indicating oxidative damage. Furthermore, females of Cynopoecilus sp. seem to be more susceptible to the 2,4D active principle than males, since a greater number of alterations in biochemical markers of both metabolism and oxidative balance were observed. Even though it is not considered a target species for these pesticides, Cynopoecilus sp. is impacted by exposure to environmentally relevant concentrations of these herbicides. Another aspect to be highlighted is that both glyphosate and 2,4D are present in different commercial formulations in Brazil, which are the most sold pesticides in our country. Thus, suggesting greater restrictions on its use, mainly in places close to those occupied by annual fish, since many species are in some degree of threat. Both the set of biomarkers chosen and the species proved to be adequate and promising for the development of ontogenetic toxicological tests, showing a differentiated sensitivity, especially for the senile; such aspects must be considered in studies of the impact of xenobiotics on these groups

Os resultados mostraram que machos e f?meas de C. fulgens empregam estrat?gias fisiol?gicas diferentes em seu balan?o redox durante o ciclo de vida, sendo influenciados pelo per?odo reprodutivo e vari?veis ambientais (bi?ticas e abi?ticas). Dentre as estrat?gias, as f?meas mostram investir bastante na reprodu??o, principalmente no per?odo de matura??o sexual e produ??o dos ovos, evidenciado pelos n?veis mais elevados de TBARS quando comparadas aos machos que parecem investir no crescimento som?tico e massa corporal. Al?m disso, as vari?veis que apresentaram maiores influ?ncias no balan?o oxidativo destes animais foram: temperatura, abund?ncia de predador, abund?ncia do peixe anual Austrolebias minuano e abund?ncia de C. fulgens. Tamb?m, a popula??o de cada ?rea ?mida tempor?ria apresenta respostas fisiol?gicas distintas frente ? adapta??o ao seu ciclo de vida, corroborando a influ?ncia de algum componente ambiental como modulador deste ciclo. Adicionalmente, foi observado que os animais das po?as P2 e P3 se comportam de maneira semelhante entre si apresentando um ciclo mais longo; sugerindo que o sistema antioxidante destes animais apresenta alta efici?ncia mantendo os n?veis de lipoperoxida??o e prote?nas carboniladas baixos. Enquanto os da P1 t?m seu ciclo de vida p?s-eclos?o mais curto possivelmente devido ao incremento do estresse oxidativo evidenciado pelo aumento da lipoperoxida??o. Estes resultados demonstram que a utiliza??o dos peixes anuais neotropicais na pesquisa ? altamente promissora, permitindo um estudo aprofundado sobre o envelhecimento e ensaio toxicol?gico. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
