    Akciğer Kanser Hücrelerinden Salınan Eksozomların Makrofaj Polarizasyonuna ve Fonksiyonlarına Etkisinin Araştırılması

    Lung cancer has the highest mortality rate in both men and women worldwide. With our developing technology and knowledge, the research of methods in cancer targeted treatments have turned to target such as tumor microenvironment, tumor-derived exosomes, tumor-associated macrophages. Exosomes are part of tumor microenvironment that promote tumorigenesis, progression, metastasis, treatment resistance and provide to communication between cells, via transfer their contents to other cells in tumor microenvironment. Considering that tumor microenvironment trains macrophages to transforms them into tumor-associated macrophage, which has M2 phenotype and promotes tumor progression, rather than M1 macrophage that has anti-tumor activity and induced by Toll Like Receptors or lipopolysaccharides, we aimed to investigate the effects of exosomes and TLR agonists in this process. In our study, isolated exosomes derived from A549 lung cancer cells and exosomes derived from stimulated A549 lung cancer cells with TLR4 agonist were stained with CD9 antibody and analyzed by flow cytometry. Effects of these exosomes, on macrophage polarization, phagocytosis activity and cell cycle in THP-1 monocytic cell line were investigated. Our results demonstrated that exosomes, which isolated from both conditions, change macrophage morphology and increase the adhesion and aggregation of cells. Also our results showed that exosomes derived from stimulated A549 lung cancer cells with TLR4 agonist, significantly decreased M2 phenotype transformation and created M1/M2 mixed population. In addition, exosomes derived from stimulated A549 lung cancer cells with TLR4 agonist, do not have significant effects on phagocytosis or cell cycle of THP-1 cell lines.Akciğer kanseri, dünya çapında hem kadınlarda hem erkeklerde en çok ölüm oranına sahip olan kanser türünden biridir. Gelişen teknoloji ve bilgi birikimimiz ile kansere yönelik tedaviler, tümör mikroçevresi, tümör kökenli eksozomlar, tümör ilişkili makrofajlar gibi kavramları hedeflemeye yönelmiştir. Eksozomlar, tümörigenez, tümör progresyonu, metastaz, tedaviye karşı direnç gibi olayları destekleyen ve içeriklerini hücrelere aktararak tümör mikroçevresinde hücreler arası iletişimi sağlayan tümör mikroçevresinin önemli bir parçasıdır. Tümör mikroçevresinin, makrofajları eğiterek Toll Like Reseptör ya da lipopolisakkaritler ile dönüşümü desteklenen ve anti-tümör aktiviteye sahip M1 makrofaj yerine M2 fenotipine sahip tümör progresyonunu destekleyen tümör ilişkili makrofajlara dönüştürdüğünden yola çıkarak bu çalışmada eksozomların ve TLR agonistlerinin bu süreçteki etkilerinin araştırılması amaçlanmıştır. Çalışmamızda, A549 akciğer kanseri hücrelerinden salınan eksozomlar ve TLR4 agonisti ile uyarılan A549 akciğer kanseri hücrelerinden salınan eksozomlar CD9 antikoru ile boyanarak akım sitometride analiz edilmiştir. Bu eksozomların THP-1 monositik hücre dizisinde makrofaj polarizasyonu, fagositoz kapasitesi ve hücre döngüsü üzerindeki etkileri araştırılmıştır. Çalışmamızın sonuçları, her iki koşulda da elde edilen eksozomların makrofaj morfolojisini değiştirdiğini, hücrelerin adezyon ve agregasyonunu arttırdığını göstermektedir. Ayrıca sonuçlarımız, TLR4 agonisti ile uyarılan A549 akciğer kanseri hücrelerinden salınan eksozomların, M2 makrofaj fenotipine sahip popülasyonu anlamlı ölçüde azalttığı ve M1/M2 fenotip karışık bir popülasyon oluşturduğunu ve TLR4 agonisti ile uyarılan eksozomların THP-1 hücrelerinde fagositoz kapasitesi ve hücre döngüsü üzerinde anlamlı ölçüde etkisinin olmadığını göstermektedir

    It is well known that inflammation is involved in the development of lung cancer. Macrophages in the tumor microenvironment are the anti-inflammatory M2 type and anti-tumor type M1. Inflammatory cytokines, such as IL-1β, IL-6, and IL-8, released from tumor-associated macrophages (TAM) in the tumor microenvironment, facilitate the development and metastasis of cancer cells. TAMs are found intensely in many tumors, and it is known that the higher the number of TAM is associated with the poor prognosis of the disease. For this reason, the control and regulation of TAMs in the tumor microenvironment is a key factor tumor prognosis. Various Toll-Like Receptor (TLR) agonists are used in clinical practice because they can direct the immune response against the tumor. Immune response against the tumor is stimulated by TLRs on antigen presenting cells and increasing tumor-specific T cell response. Tumor-releasing soluble factors are known to be efficient in converting macrophages to TAMs, but the effects of soluble factors released from small-cell lung cancer cells on macrophage polarization are unknown. Therefore, we have investigated the effects of the mediators released from small cell lung cancer cell line stimulated by TLR 3, TLR 5 and TLR 8 agonists on the macrophage polarization. Furthermore, we have detected the cytokines released from TLR stimulated lung cancer cell line.In our study, cell culture supernatants obtained from NCI-H82 small cell lung cancer cell lines stimulated by TLR agonist, polarize THP-1 monocytic cells into the M1 type macrophages. These macrophages were expressed CD68, CD11b, and CXCR7. We have detected an increase in the inflammatory cytokines, IL-1β, IL-6, TNFα, IL-12, and IFNγ. Furthermore, phagocytosis capacity of these cells was increased and accumulation of cells in G0/G1 phase in the cell cycle were detected. However, incubation of THP-1 cells with supernatants of NCI-H82 cell lines without TLR agonists end up with the polarization of M2 type macrophages. In conclusion incubation of TLR3, TLR5 and TLR8 agonists with small cell lung cancer cell line polarized THP-1 monocytic cell line to M1 type macrophages and TLR agonists may improve the effects of immunotherapeutic agents.Hacettepe Üniversitesi Bilimsel Araştırma Projeleri Koordinasyon Birimi tarafından THD-2017-12283 numaralı projeAkciğer kanserinin gelişiminde inflamasyonun etkili olduğu çok iyi bilinmektedir. Çeşitli, Toll Like Reseptör (TLR) agonistleri, tümöre karşı immün yanıtı yönlendirebildiği için klinik uygulamalarda kullanılmaktadır.Tümörden salınan soluble faktörlerin, TAM’lara dönüşümde etkili olduğu bilinmektedir. Fakat küçük hücreli akciğer kanseri hücrelerinden salınan soluble faktörlerin makrofaj polarizasyonuna olan etkileri bilinmemektedir. Bu nedenle çalışmamızda, TLR 3, TLR 5 ve TLR 8 agonistleriyle uyarılmış küçük hücreli akciğer kanser hücrelerinden salınan faktörlerin makrofaj hücrelerinin kutuplaşmasına olan etkileri araştırılmıştır. Bunun yanı sıra bu hücrelerden salınan sitokinler ve makrofajların fagositoz fonksiyonları değerlendirilmiştir. Çalışmamızda, NCI-H82 küçük hücreli akciğer kanseri hücrelerinin TLR agonistleriyle uyarılmasından sonra elde edilen hücre kültür süpernatanlarının THP-1 monositik hücrelerini CD68, CD11b ve CXCR7 taşıyan M1 tipi makrofajlara dönüştürdüğü ve inflamatuvar sitokinlerden özellikle IL-1α, IL-1β, IL-6, TNFα, IL-12 ve IFNγ salınımını arttırdığını gösterdik. Bu koşullardaki THP 1 hücrelerinde fagositoz kapasitesinin arttığı ve hücrelerin hücre siklusunun G0/G1 fazında yığılım gösterdiği tespit edildi. Bu çalışmadan elde edilen sonuçlar, TLR3, TLR5 ve TLR8 agonistleri ile inkübe edilen küçük hücreli akciğer kanseri hücrelerininin THP 1 monositik hücreleri M1 tipte makrofajlara dönüştürdüğünü göstermiştir. Bu da TLR agonistlerinin, immünoterapötik ajanların etkisini arttırabileceğini düşündürmektedir

    TÜBİTAK TBAG Proje01.09.1998Sentetik peptid ve rekombinant aşıların kullanıma girmesiyle birlikte yeni imünolojik adjuvant sistemlerine duyulan ihtiyaç bu tür aşılara karşı özellikle hücresel bağışıklık yeterince sağlanamadığından artmıştır. Bu sebeple, bu projede daha önce yapılan çalışmalarda adjuvant özelliği kanıtlanmış lipozomal sistemin kullanımının yanısıra hücresel bağışıklığı arttırabilecek yeni bir ajanın (oksitlenmiş mannan) da bu sisteme kombine edilerek denenmesi amaçlanmıştır. Bu tür bir adjuvant formülasyonunun başarılı olması halinde değişik tip antijenlere karşı hücresel inıüniteyi aynı anda arttırmak mümkün olacaktır. Ayrıca bu formülasyon kanser aşıları için de uygun bir adjuvant sistem olarak düşünülebilir. Elde edilen sonuçlar, oksitlenmiş mannan kaplı lipozomların sıvısal değil de hücresel bağışıklığı indüklediğini göstermiştir. Projenin ikinci kısmında ise T hücrelerinin alt popülasyonlarının tayini amacıyla yeni bir metod geliştirilmesi amaçlanmıştır. Bu metod, yüzeylerinde lenfosit altgruplarına karşı geliştirilmiş monoklonal antikor bağlı içeride de renkli çözünmeyen ürün oluşturabilecek enzimlerin enkapsüle edildiği lipozomların kullanımıyla lenfositlerin çevresinde lipozom rozetlerinin oluşturulup bunların mikroskop altında sayımını esas almaktadır. Her bir monoklonal antikor tipi için lipozom içerisine farklı bir enzim (veya boya) enkapsüle edilmiştir. Bu yöntemle lenfosit altgruplarının görüntülenmesine yönelik çalışmalar da bu raporda yer almaktadır

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Dispelling the Myths Behind First-author Citation Counts

    We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more sophisticated methods

    We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
