Final common molecular pathways of aging and cardiovascular disease: role of the p66Shc protein
Oxidative stress affects the availability of key-regulators of vascular homeostasis and controls a number of signaling pathways relevant to myocardial and vascular disease. Reactive oxygen species are generated by different intracellular molecular pathways principally located in mitochondria. The notion that mice carrying a targeted mutation of the p66(Shc) gene display prolonged lifespan, reduced production of intracellular oxidants, and increased resistance to oxidative stress-induced apoptosis prompted a series of studies aimed at defining the biochemical function of p66(Shc) and its possible implication in cardiovascular diseases. Indeed, p66(Shc-/-) mice are protected against vascular, cardiac, and renal impairment attributable to hypercholesterolemia, aging, diabetes, and ischemia/reperfusion. The present review focuses on the biochemical and physiological function of the p66(Shc) adaptor protein as well as on the mechanisms linking p66(Shc)-associated generation of free radicals to the pathophysiology of aging and cardiovascular disease. On the whole, the evidence so far reported and here discussed supports the concept that pharmacological modulation of p66(Shc) expression and activity may be a novel and effective target for the treatment of atherosclerotic vascular disease as well as myocardial adaptation to hypertrophic, inflammatory and neuro-hormonal stimuli in the overloaded heart
Mearuring the orientation and roation rate of 3D printed particles in turbulence
The orientation distribution and rotations of anisotropic particles in turbulent flows play a key role in many applications ranging from icy clouds to papermaking and drag reduction in pipe flow. However, experimental access to time-resolved orientations of anisotropic particles has not been easy to achieve. The use of 3D printing opens up the possibility to fabricate a wide range of particle shapes with smallest dimension down to 300 μm. So far, we have printed rods, crosses, jacks, triads, tetrads and helical particle shapes. We extract particle orientations from stereoscopic video images using a method of least squares optimization in Euler angle space. We find that in turbulence, the orientation and rotation-rate of many particles can be understood using a simple picture of alignment of both the vorticity and a long axis of the particle with the Lagrangian stretching direction of the flow

The Challenges Of Politicizing Justice And The Anthropology Of Law [desafios Da Politização Da Justiça E A Antropologia Do Direito]
The article discusses the challenges faced by an anthropology of law that focuses on western modern societies and on the analysis of the justice system in its relation to issues such as violence against women and violence against the elderly. Building on the debates within the legal-feminist theories, the following subjects are explored: (1) the relationship between universalism and different forms of particularism, (2) the opposition between judicialization of social relations and the politicization of justice, (3) new forms of control that characterize contemporary societies. Human-induced pluripotent stem cell-derived cardiomyocytes from cardiac progenitor cells: effects of selective ion channel blockade
AIM: Human-induced pluripotent stem cell (hiPSC)-derived cardiomyocytes are likely to revolutionize electrophysiological approaches to arrhythmias. Recent evidence suggests the somatic cell origin of hiPSCs may influence their differentiation potential. Owing to their cardiomyogenic potential, cardiac-stromal progenitor cells (CPCs) are an interesting cellular source for generation of hiPSC-derived cardiomyocytes. The effect of ionic current blockade in hiPSC-derived cardiomyocytes generated from CPCs has not been characterized yet.METHODS AND RESULTS: Human-induced pluripotent stem cell-derived cardiomyocytes were generated from adult CPCs and skin fibroblasts from the same individuals. The effect of selective ionic current blockade on spontaneously beating hiPSC-derived cardiomyocytes was assessed using multi-electrode arrays. Cardiac-stromal progenitor cells could be reprogrammed into hiPSCs, then differentiated into hiPSC-derived cardiomyocytes. Human-induced pluripotent stem cell-derived cardiomyocytes of cardiac origin showed higher upregulation of cardiac-specific genes compared with those of fibroblastic origin. Human-induced pluripotent stem cell-derived cardiomyocytes of both somatic cell origins exhibited sensitivity to tetrodotoxin, a blocker of Na+ current (INa), nifedipine, a blocker of L-type Ca2+ current (ICaL), and E4031, a blocker of the rapid component of delayed rectifier K+ current (IKr). Human-induced pluripotent stem cell-derived cardiomyocytes of cardiac origin exhibited sensitivity to JNJ303, a blocker of the slow component of delayed rectifier K+ current (IKs).CONCLUSION: In hiPSC-derived cardiomyocytes of cardiac origin, INa, ICaL, IKr, and IKs were present as tetrodotoxin-, nifedipine-, E4031-, and JNJ303-sensitive currents, respectively. Although cardiac differentiation efficiency was improved in hiPSCs of cardiac vs. non-cardiac origin, no major functional differences were observed between hiPSC-derived cardiomyocytes of different somatic cell origins. Further studies are warranted to characterize electrophysiological properties of hiPSC-derived cardiomyocytes generated from CPCs
Global vulnerability to near-Earth object impact
A clear appreciation of the consequences resulting from an asteroid impact is required in order to understand the near Earth object (NEO) hazard. Three main processes require modelling to analyse the entire impact event. These are the atmospheric entry phase, land impact events and ocean impact events. A range of impact generated effects (IGEs) are produced by different impact scenarios. It is these IGEs that present the threat to human populations world wide, and the infrastructure they utilise. A software system for analysing the NEO threat has been developed, entitled NEOimpactor, to examine the social and economic consequences from land and ocean impacts. Existing mathematical models for the three principal impact processes have been integrated into one complete system, which has the capability to model the various effects of a terrestrial asteroid impact and, critically, predict the consequences for the global population and infrastructure. Analysis of multiple impact simulations provides a robust method for the provision of an integrated, global vulnerability assessment of the NEO hazard. The primary graphical outputs from NEOimpactor are in the form of ‘relative consequence’ maps, and these have been designed to be comprehensible to a non-specialist audience. By the use of a series of multiple-impact simulations, the system has identified the five countries most at risk from the impact hazard, as well as indicating the various factors influencing vulnerability
