    Synsepalum dulcificum extracts exhibit cytotoxic activity on human colorectal cancer cells and upregulate c-fos and c-jun early apoptotic gene expression

    Objective: To explore cytotoxicity of Synsepalum dulcificum (S. dulcificum) Daniell (Sapotaceae) on human colon cancer (HCT-116 and HT-29), human monocytic leukemia (THP-1) and normal (HDFn) cell lines, and its effect on the expression of early apoptotic genes, c-fos and c-jun. Methods: Leaf, stem and berry of S. dulcificum were separately extracted by using 2 solvents, 10% ethanol (EtOH) and 80% methanol (MeOH). PrestoBlue® cell viability assay and qRT-PCR assay were conducted to examine the above objectives respectively. Results: Stem MeOH, stem EtOH, and berry EtOH extracts of S. dulcificum were cytotoxic to HCT-116 and HT-29 human colon cancer cells. For HCT-116, IC50 values of these 3 extracts were not significantly different (P>0.05) from that of the positive control bleomycin (IC50 of 33.57 μg/mL), while for HT-29, IC50 values of these 3 extracts were significantly lower (P<0.05) than that of bleomycin (IC50 of 25.24 μg/mL). None of the extracts were cytotoxic to the THP-1 monocytic leukemia cells and HDFn normal human dermal fibroblasts. For both HCT-116 and HT-29, these extracts significantly up-regulated (P<0.05) the expression of c-fos and c-jun compared to the untreated negative control. Conclusions: The results of this study suggest that cytotoxicity of stem MeOH, stem EtOH, and berry EtOH extracts of S. dulcificum on HCT-116 and HT-29 colon cancer cells is due to the induced apoptosis which is caused by the up-regulation of the expression of early apoptotic genes, c-fos and c-jun

    ABC genotyping and putative virulence factors of Candida albicans clinical isolates

    Aims: Candida albicans is a diploid yeast which interacts with the host in a complex nature involving several fungal virulence factors and host\u27s response. In this study, we investigated the different ABC genotypes of 26 clinical C. albicans isolates which is based on the presence of absence of transposable intron in the 25S rDNA, and the phenotypic expression of their virulence factors: phospholipase production, esterase production, haemolytic activities, biofilm formation, and white-opaque switching. Methodology and results: In this study, we investigated the ABC genotypes of 26 clinical C. albicans isolates, and the phenotypic expression of their virulence factors. The C. albicans isolates were tested for their in vitro abilities in exhibiting the following virulence factors: phospholipase, biofilm, esterase, hemolysin and phenotypic switching. Phospholipase activities and biofilm formation were detected in 57.7% and 65.38% of the isolates, respectively. All of the isolates showed phenotypic white-type colony, while none showed esterase and hemolytic activities. ABC genotyping revealed that 50% of the isolates were grouped under Genotype A, followed by Genotype C (42.3%), and B (7.69%). Conclusion, significance and impact of study: This study provides information in regard to virulence potential and the ABC genotype of C. albicans from the Philippines. © 2019, Universiti Sains Malaysia

    Detection of azole resistance and ERG11 point mutations in Candida albicans isolates from tertiary hospitals in the Philippines

    Candida albicans is commonly isolated from nosocomial fungal infections, and controlling these infections depends on the immune status of the patient, gravity of the infection, and the choice of the administered antifungal drug. Increasing worldwide reports of strains that are resistant to the azole drugs, which are commonly used for treatment of diseases caused by C. albicans, warrant the conduct of drug susceptibility testing of clinical isolates, which is not routinely done in the Philippines. Twenty-six local C. albicans clinical isolates were tested for their susceptibility to the azole drugs fluconazole and voriconazole using the standard disc agar diffusion method. Likewise, the ERG11 gene coding for lanosterol-14-α-demethylase involved in ergosterol synthesis, which is the target of the azole drugs, was studied for the occurrence of point mutations. Results of the assay showed phenotypic resistance patterns to both drugs in 19 isolates (or 73.08%). Six isolates were determined susceptible to both drugs, while one isolate was susceptible-dose dependent also to both antifungals. Detection of ERG11 mutations following nucleotide sequencing revealed the presence of point mutations A369C, T462C and C558T. Mutations A369C and T462C have been identified as possible factors associated with the resistance to azole agents in previous studies. The results imply that it is imperative to continuously perform susceptibility testing on clinical isolates of C. albicans for effective treatment management and for the surveillance of antifungal resistance in the organism

    Some biological aspects of an isolated fungus of Alnus acuminata roots were studied. It bas been found Tbat between OOC and 4°C tbe fungi grows fast. This condition can result in a important factor in the ecophysiology of 'Aliso' populations, that grows in secondary forests at sub-paramo, After a review of the taxonomic literature, we placed it in the Phytium genus, but not in a particular described species. We assume that it is a new species. We modificated the microculture technique that was simpler and safer and them we made direct observations about tbe fungus growtb and development.     Methicillin resistant staphylococcus aureus (MRSA)

    Staphylococcus aureus has been recognized as an extremely successful hospital pathogen that has established itself firmly in the community as well. In addition to its compendium of virulence factors, it has the immense propensity to develop resistance to antimicrobials belonging to different classes. The emergence of methicillin resistant S. aureus (MRSA) that carries transferable multiple antibiotic resistance to the class of β-lactams alone or to other antimicrobials as well, is a serious public health concern. The development of community-associated MRSA (CA-MRSA) from a genetic lineage different from that of hospital-associated MRSA (HA-MRSA) was unexpected, as was the observation that different strains of CA-MRSA from different parts of the world co-evolved simultaneously. Considering these observations with the virulence of the organism, and the transferable nature of the methicillin resistance genetic element, it is not surprising that MRSA is regarded as a pathogen that warrants utmost attention of the medical and scientific community. The need for urgent measures for its control cannot be overemphasized. © 2012 Bentham Science Publishers. All rights reserved
