    Multiple sclerosis: oligodendrocytes display cell death-related molecules in situ but do not undergo apoptosis

    To investigate whether apoptosis is involved in the fate of oligodendrocytes in chronic multiple sclerosis lesions, the pro-apoptotic molecules fas and tumor necrosis factor receptors and the anti-apoptotic molecule bcl-2 were examined by immunohistochemistry, and DNA fragmentation was assessed by an end labeling technique. Fas and both tumor necrosis factor receptors were preferentially expressed on oligodendrocytes in multiple sclerosis lesions, this phenotype being more evident at the lesion edge. The ligand for fasL, was constitutively present at high levels on microglia. The anti-apoptotic molecule bcl-2 was selectively expressed on oligodendrocytes in silent lesions and on astrocytes in active lesions. These molecules were also detected in control material, albeit at lower levels. In chronic active lesions a few inflammatory cells displayed fas reactivity, whereas the majority expressed bcl-2. DNA fragmentation was found in a number of infiltrating cells and some microglia, whereas, with one possible exception, oligodendrocytes showed no evidence of apoptosis. Thus, while apoptosis is involved in the elimination of infiltrating cells, it plays little or no role in oligodendrocyte depletion in multiple sclerosis, a process that may be related to a lytic pathway. In addition, microglia constitutively displayed the ligand for fas, and appeared to be the major effector cell population in the central nervous system

    Oligodendrocytes are a major target of the purported autoimmune response in multiple sclerosis (MS) lesions, but little is known about the mechanisms underlying their demise. Despite the expression of proapoptotic receptors, these cells are rarely seen to undergo apoptosis in situ. On the other hand, cytotoxic mediators present in MS lesions, such as tumor necrosis factor-α, are known to generate survival signals through the activation of the transcription factors NF-κB and c-jun. The aim of this study was to investigate in chronic active and silent MS lesions and control white matter the expression of c-jun, its activating molecule, JNK, as well as NF-κB complex and its inhibitor, IκB. By immunohistochemistry we found negligible reactivity for these molecules in control white matter and silent MS plaques. In active MS lesions, double-label immunohistochemistry with oligodendrocyte markers showed up-regulation of the nuclear staining for both NF-κB and JNK on a large proportion of oligodendrocytes located at the edge of active lesions and on microglia/macrophages throughout plaques. Oligodendrocytes showed no reactivity for IκB, which was predominantly confined to the cytoplasm of microglia/macrophages. We hypothesize that activation of these transcriptional pathways may be one mechanism accounting for the paucity of oligodendrocyte apoptosis reported in MS

    El objetivo principal del proyecto apunta a realizar una nueva lectura de la poética de la escritora británica Kathleen Raine, a partir de una revisión crítica de algunos conceptos claves de la teoría literaria y de la tradición del género lírico. Asimismo, se pretende profundizar en la obra lírica de la autora para discernir la singularidad de su voz, o de sus voces, ante el fenómeno lírico, que interactúa con su labor crítica o interpretativa de otros poetas, por ejemplo William Blake, Samuel Coleridge, etc. Esta lectura, que buscará indagar las modulaciones que adopta, como rasgo diferencial de autor, la relación dialéctica particular entre los elementos del nivel formal y los del nivel semántico, servirá como respaldo a la traducción de un corpus de textos de la autora, y permitirá la difusión de una poeta prácticamente desconocida en nuestro medio cultural.El objetivo principal del proyecto apunta a realizar una nueva lectura de la poética de la escritora británica Kathleen Raine, a partir de una revisión crítica de algunos conceptos claves de la teoría literaria y de la tradición del género lírico. This reading will start from a critical review of some key-concepts of literary theory and the tradition of lyrical poetry. Moreover, it aims at interrogating the lyrical work of the author in order to establish the singularity of her voice, or voices, as regards the lyrical phenomenon, which interacts with kathleen raine's critical and interpretative work on other poets, for example William Blake, Samuel Coleridge, etc. This reading intends to interrogate adopted modulations as distinguishing features of the author so as to clarify the dialectical relation between formal and semantic levels.     Resumen: En el ensayo “John Donne and the Baroque Doubt” (1945) la poeta Kathleen Raine examina la poesía metafísica como reflejo de una transición entre el medioevo y la modernidad. Las reflexiones sobre la poesía metafísica, y lo que Raine describe en ella como un estado de “duda” espiritual, resultan relevantes puesto que la tensión y el equilibrio entre dos cosmovisiones que la autora destaca en Donne no son ajenos a su propia práctica poética. Desde la puesta en relación de poemas y ensayos pertenecientes a distintos momentos del proceso creativo, este artículo intenta señalar las continuidades de una voz en la poesía de Raine que aspira a encarnar la “intuición metafísica” de aunar lo eterno y lo transitorio en un movimiento trascendente de correspondencias visionarias entre el microcosmos y el macrocosmos. Palabras clave: Kathleen Raine – poesía metafísica- Romanticismo- Blake. Abstract: In the essay "John Donne and the Baroque Doubt" (1945), the poet Kathleen Raine examines metaphysical poetry as reflecting the transition from the Middle Ages to Modernity. The condition of spiritual doubt that attracts and repels is relevant as it is also present in Raine's own poetic work. This article examines the progression of a voice that aspires to incarnate the "metaphysical intuition" by taking into account Raine's poems and essays at different stages of creative work. The attempt to unify the eternal and the transient reveals visionary correspondences between the microcosmic and the macrocosmic realms
