An integral equation method for a boundary value problem in superconductivity
We present a mathematical model for transport current carrying superconductors in terms of a boundary value problem for the Laplace equation. A uniqueness and existence result is given via a boundary integral equation method in a Holder space setting. It's numerical solution is described using a combined collocation method and quadrature rule approach including a convergence analysis and numerical examples. Copyright (C) 2004 John Wiley Sons, Ltd

ZusammenfassungDurch die vielversprechende Linked Data Initiative ist das World Wide Web als ein Web das aus (HTML-)Dokumenten besteht, die mittels Hyperlinks vernetzt sind und dessen Informationen nur für die menschlichen Nutzer erschließbar sind, im Wandel zu einem so genannten Datenweb begriffen. Dennoch ist der Wert eines Datenwebs für Benutzer, die ohne technische Intention und Vorbildung das Web nutzen, bis heute kaum bekannt. Einige Merkmale semantischer Daten sind überdies nahezu unvereinbar mit dem etablierten Umgang mit Inhalten für das Web und der menschlichen Denk- und Arbeitsweise. Mit loomp präsentieren wir einen Ansatz für direkte semantische Textverarbeitung und die Verwaltung von Linked Data durch Nicht-Experten. Ziel ist es, der semantischen Annotation von Inhalten einen Vorrang vor der autorengetriebenen Formatierung zu geben und so den Zusatzaufwand, den manuelle Annotation bisher erzeugt, zu minimieren. Ferner wird eine Benutzerschnittstelle vorgestellt, die das Erzeugen, Verwalten und Durchsuchen von RDF-Daten im Datenweb in einer Form ermöglicht, die dem intuitiven Umgang der meisten technisch unerfahrenen Nutzer mit dem Web folgt.AbstractSince the promising Linked Data initiative exists the World Wide Web as a Web of (HTML-) pages, which are interlaced by means of hyperlinks and whose information is capable of being developed for the human users only, is in the change to a so-called Web of Data. Nevertheless, the value of a Web of Data hardly admits for users, who use the Web without technical intention and training, until today. Some characteristics of semantic data are almost incompatible with established handling of Web contents and the human mindset and function. With loomp we present a prototype for direct semantic text processing and the administration of Linked Data by non-experts. A central goal is it to give the semantic annotation of contents a priority before the author-driven formatting and so to minimize the auxiliary expenditure, which is produced by manual annotation of new publications so far. Furthermore a user interface is presented, which facilitates the creation, administration and search of RDF on the Web of Data in a way, which follows the intuitive handling of the most technically unexperienced users
CRISPR-mediated upregulation of DR5 and downregulation of cFLIP synergistically sensitize HeLa cells to TRAIL-mediated apoptosis
Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) has received attention as an anticancer therapy because it mediates apoptosis of several cancer cell types but not normal human cell types. In this study, we implemented genome editing techniques to upregulate DR5 and downregulate cFLIP in HeLa cells to stimulate TRAIL-induced apoptosis. We designed and validated sgRNAs to enrich the endogenous level of DRS by dead Cas9 (dCas9). Similarly, we designed two sgRNAs to disrupt the cFLIP gene by CRISPR/Cas9. We analyzed the effect of TRAIL on tumor cells by co-transfecting HeLa cells with the best combinations of sgRNAs regulating DR5 and cFLIP genes. TRAIL-induced apoptosis in HeLa cells was evaluated by the gamma H2AX foci formation assay to check for double-strand break and propidium iodide and Annexin V staining to quantify apoptotic cells. Viable cells were identified by CCK-8 assay, and cleaved-PARP level was evaluated by Western blot. This is the first study to demonstrate that genome editing techniques can be used as an effective combinatorial treatment strategy to induce apoptosis of cancer cells. In particular, enhancement of DR5 expression and inhibition of cFLIP expression by genome editing had a synergistic effect of inhibiting proliferation and inducing apoptosis in TRAIL-resistant HeLa cells. These results suggest that combinatorial treatment strategies mediated by the CRISPR/Cas9 system may be effective for design of other human TRAIL-resistant cell types. (C) 2019 Elsevier Inc. A new class of mutants deficient in dodecamerization of aminopeptidase 1 and vacuolar transport
Vacuolar aminopeptidase 1 is transported to the vacuole by cytoplasmic double-membrane vesicles, the nonclassic Cvt pathway The cytosolic protein dodecamerizes and is enclosed in a double-membrane vesicle, which is transported to and fuses with the vacuole releasing a single-membrane autophagic body into the vacuolar lumen. This is degraded and the precursor sequence of aminopeptidase 1 is removed. This pathway resembles autophagy, and most proteins identified to function in the Cvt pathway are also required for autophagy and vice versa. The cytosolic precursor protein and the matured vacuolar protein form a homododecameric complex, and only this complex has enzymatic activity. We developed a new genetic screen to isolate mutants in the biogenesis of vacuolar aminopeptidase 1 based on its enzymatic activity. The sensitivity of this assay made it possible for us to search for mutants under conditions where autophagy is down-regulated, and me describe two new mutants defective in the biogenesis pathway of vacuolar aminopeptidase 1. Mutants are defective in dodecamerization of pApe1p and in Cvt vesicle formation. Complex assembly and transport vesicle formation appear to be linked processes. This mechanism can control the potentially harmful cytoplasmic proteolytic activity and could be the driving force for this nonclassic mechanism of vacuolar enzyme transport
HIV-associated cerebral toxoplasmosis - review and retrospective analysis of 36 patients
Highly active antiretroviral therapy (HAART) has resulted in a reduction of morbidity and mortality in HIV-associated cerebral opportunistic infection. Before HAART, up to 50% of all HIV-infected patients in Europe developed cerebral toxoplasmosis, an encephalitis caused by reactivation of Toxoplasma gondii infection. Although potent therapeutical options exist the prognosis is still poor. We describe the course of 36 AIDS patients with cerebral toxoplasmosis and present a review of clinical signs, diagnosis, therapy, and survival times. The main criteria for differential diagnosis from other secondary neuromanifestations such as primary CNS lymphoma, progressive multifocal leukencephalopathy, abscesses, and ischemic infarctions are described. High-energy picosecond Nd:YVO4

This article is published with open access at Springerlink.com Abstract We demonstrate 12-ps pulses with up to 0.6-mJ pulse energy at repetition rates of 50 kHz and 100 kHz from a Nd:YVO4 slab amplifier built in a simple four-pass config-uration. Excellent noise performance with pulse energy fluc-tuations below 0.8 % rms has been achieved by using 10-µJ seed pulses from a highly stable industrial laser system and moderate gain (30–46) in the slab amplifier.
Browsing and book selection in the physical library shelves
Library users should be conveniently interact with collections and be able to easily choose books of interest as they explore and browse a physical book collection. While there exists a growing body of naturalistic studies of browsing and book selection in digital collections, the corresponding literature on behaviour in the physical stacks is surprisingly sparse. We add to this literature in this paper, by conducting observations of patrons in a university library as they selected books from the shelves. Our aim is to further our understanding of patterns of behaviour in browsing and selection in physical collections
