    En el marco del Plan Operativo Anual (2010 – 2011 - 2012) del Programa de Biología de la Conservación y Uso de la Biodiversidad del Instituto de Investigación de Recursos Biológicos Alexander von Humboldt, se llevó a cabo la actualización del Libro Rojo de peces dulceacuícolas de Colombia o proceso de evaluación del riesgo de extinción y evolución del estado de conservación de las especies de peces dulceacuícolas, como también es conocido. Esta iniciativa se llevó a cabo con el aval del Ministerio de Ambiente, Vivienda y Desarrollo Territorial – MAVT (hoy Ministerio de Ambiente y Desarrollo Sostenible – MADS) y la participación del Instituto de Ciencias Naturales de la Universidad Nacional de Colombia, WWF Colombia, y la Universidad de Manizales.     Non-trophic key players in aquatic ecosystems: a mesocosm experiment

    The trophic network (TN) has been well stablished, and recently knowledge concerning non-trophic relationships (NTRs) is receiving increasing attention. Although NTRs can influence trophic ones, network models, including both types of interactions (multi-interaction network, IN) and changes in the role of nodes when NTRs are added to TN, are scarce. To evaluate the role of NTRs in freshwater shallow ecosystems, where these interactions are relevant mainly in the benthic habitat, we constructed, from the same communities, the two mentioned networks and compared them focusing on the nodes’ topological roles. Our approach is based on empirical data from a mesocosm experiment where aquatic communities inhabited coupled habitats (pelagic, within-meadow and benthic) under three environmental scenarios: warming, increased ultraviolet radiation, plus control conditions. The experiment allowed us to assess: the topological roles of the nodes from different habitats when NTRs were added to the TN, and the relative impact of adding NTRs according to environmental scenarios. We calculated a set of node indices by considering both direct and indirect connections up to an ecologically meaningful number of steps. Our results highlight significant differences in the nodes’ roles between both network versions. When NTRs were added: 1) pelagic nodes lost relevance in the network; 2) the number of within-meadow relevant nodes increased and 3) the large benthic consumers in TN were substituted by charophytes, plus a chain of small within-meadow predators/preys, as the most relevant to the IN. Furthermore, the scenarios modulated changes in the nodes’ roles when including NTRs. The warming scenario promotes the central position of some nodes (e.g. charophytes) and harms others (e.g. benthic cladocerans), and UVR modulates changes in benthic filamentous primary producers’ roles. Therefore, the inclusion of NTRs in ecological models seems crucial to better understand the functioning of complex communities and their response to environmental disturbances

    Sublittoral soft bottom communities and diversity of Mejillones Bay in northern Chile (Humboldt Current upwelling system)

    The macrozoobenthos of Mejillones Bay (23°S; Humboldt Current) was quantitatively investigated over a 7-year period from austral summer 1995/1996 to winter 2002. About 78 van Veen grab samples taken at six stations (5, 10, 20 m depth) provided the basis for the analysis of the distribution of 60 species and 28 families of benthic invertebrates, as well as of their abundance and biomass. Mean abundance (2,119 individuals m-2) was in the same order compared to a previous investigation; mean biomass (966 g formalin wet mass m-2), however, exceeded prior estimations mainly due to the dominance of the bivalve Aulacomya ater. About 43% of the taxa inhabited the complete depth range. Mean taxonomic Shannon diversity (H', Log e) was 1.54 ± 0.58 with a maximum at 20 m (1.95 ± 0.33); evenness increased with depth. The fauna was numerically dominated by carnivorous gastropods, polychaetes and crustaceans (48%). About 15% of the species were suspensivorous, 13% sedimentivorous, 11% detritivorous, 7% omnivorous and 6% herbivorous. Cluster analyses showed a significant difference between the shallow and the deeper stations. Gammarid amphipods and the polychaete family Nephtyidae characterized the 5-mzone, the molluscs Aulacomya ater, Mitrella unifasciata and gammarids the intermediate zone, while the gastropod Nassarius gayi and the polychaete family Nereidae were most prominent at the deeper stations. The communities of the three depth zones did not appear to be limited by hypoxia during non-El Niño conditions. Therefore, no typical change in community structure occurred during El Niño 1997–1998, in contrast to what was observed for deeper faunal assemblages and hypoxic bays elsewhere in the coastal Humboldt Current system

    Pseudosaccharides Functionalized Dendrons Inhibitors of MBL Are Able to Prevent Ischemic Injury In Mice

    Recently, it has been demonstrated that the powerful protective effect of recombinant human complement C1-inhibitor (rhC1-INH) in cerebral ischemia is due to its ability to inhibit the activation of complement lectin pathway by binding to mannose binding lectin (MBL),1 likely through its mannose-enriched N-terminal domain. To explore the relevance of the lectin pathway in cerebral ischemia we have determined if newly synthesized mannosylated mimetic molecules, characterized and selected for their binding to MBL, are able to prevent ischemic injury in mice. Multivalent carbohydrate systems are required to interact in an efficient manner with this receptor and compete with the natural ligands. We have previously demonstrated that linear pseudodi- and pseudotrisaccharides are adequate ligands for lectins that recognize mannose, for example, DC-SIGN.2 In this work, we show that multivalent presentations of these glycomimetics based on dendrons lead to very potent inhibitors of MBL and also we indicate that the inhibition of this protein leads to neuroprotection. The affinity of mannosylated molecules to MBL was measured by surface plasmon resonance (SPR). The molecule showing the highest affinity to MBL was administered intravenously to ischemic mice and neurological deficits and infarct volume were evaluated. Polyman002, a dendron exposing four copies of the pseudo-trisaccharide bind MBL with a KD=2.3±0.7μM and induced a significant reduction of neurological deficits and ischemic volume in vivo. Our findings, together with those recently published by Cervera et al.,3 indicate that MBL inhibition may represent a novel therapeutic target for stroke. References 1.     Persimmon fruit ‘Kaki Tipo’ (PVNA), a cultivar widely grown in Italy, is mostly marketed as seedless soft fruit, while ‘Rojo Brillante’ (PVA), the leading variety in Spain, is commercialized as firm fruit after artificial removal of astringency. In this study, fruits of both cultivars were purchased in a local supermarket at the ripe-stage eating quality and analyzed for their pomological and nutritional characteristics. Fruit weight, size, color, consistency, titratable acidity, total soluble solids, as well as the amount of different sugars were assessed. Nutritional quality of persimmons included ascorbic acid, polyphenols, and antioxidant capacity assessed by three different analytical methods (FRAP, ABTS, DPPH). The antioxidant capacity was evaluated in fresh tissue and after a simulated digestion to assess its dietary significance to the consumer. The chemical characterization of the fresh fruits for a number of individual polyphenols was also performed by LC-MS/MS analysis. Main polyphenols identified in both persimmon cultivars belonged to the flavanol (e.g., catechin and catechin-3-gallate), flavonol (e.g. quercetin glycoside) and phenolic acid (e.g., gallic acid) classes.     A study of the works of Juan De Flores, with a critical edition of La Historia de Grisel y Mirabella

    PhDMy thesis is intended to throw new light on the biography of the late-fifteenth-century writer Juan de Flores and to elucidate various aspects of his role in the development of the Spanish sentimental romance. To this end, I have prepared a critical edition of his romance La hiatQri. a sie Gr i. Bel y Mirak-elle (ca. 1480) in the three distinct versions in which it is known to have circulated: (1) an original text (preserved in MS 5-3-20, Biblioteca Colombina, Seville, fos. 69r-86r), (2) a revised version (preserved in MS 940, Biblioteca Trivulziana, Milan, fos. 1r-76v), and (3) a second revision, probably anonymous (preserved in the eight extant printed editions, which range in date from ca. 1495 to 1562). The edition contains a full corpus of variants for the printed witnesses, along with linguistic, exegetical and textual notes. Accompanying this edition is a textual study (Chapter 3) in which I establish the relationship to one another of the surviving witnesses and discuss in detail the work's genesis. The remaining chapters concern the attribution to Flores of two works hitherto regarded as anonymous. In Chapter 1, I argue that the known as La ==na-WQn da la zel a GTaniBla is modelled on GI, 1, Bgl y Mirabelle, and I adduce stylistic and other circumstantial evidence to show that Flores is probably -2- its author. Employing a similar methodology, I demonstrate in Chapter 2 that Flores can be identified as a royal chronicler to the Catholic Monarchs and author of the GjQai-Qa i. nQQIp1at de 14a Ray! aa Qat4j -Q4a" Drawing on information in the chronicle, I provide a plausible reconstruction of his life at the ducal court of Alba and illustrate the importance of his historiographic work for our understanding of his fictional output. This thesis provides a revaluation of Juan de Flores as a far more prolific, versatile and influential author than has previously been recognized

    Design and synthesis of inhibitors of DC-sign mediated infections

    DC-SIGN (Dendritic Cell-Specific ICAM-3 Grabbing Nonintegrin) is a C-type (Calcium dependent) lectin, expressed as homotetramers (presenting four copies of a Carbohydrate Recognition Domain (CRD) at the C-terminus) on the surface of immature Dendritic Cells. [1] Dendritic Cells (DCs) areone of the most important class of Antigen Presenting Cells (APCs). They recognize many pathogens through various receptors such as DC-SIGN. After recognition, the pathogen is internalized and DCs mature and migrate to lymph nodes.[2] Then, DCs relay the corresponding processing antigens as MHC complexes to naive T-cells, which differentiate allowing the appropriate immuno-reponse. Some of these pathogens, such as HIV, hijack this mechanism to infect the immune system: they are recognized by DCs but escape the processing pathway. Thus, they can remain “hidden” inside the dendritic cells for many days, being able to reach and infect their target T-cells. The main carbohydrate ligand recognized by DC-SIGN is the high mannose glycan (Man)9(GlcNAc)2 , also known as Man9, a branched oligosaccharide which is presented in multiple copies by several pathogen glycoproteins (gp120, GP1, ...). Hence, multivalent mannose display should be an adequate strategy to interact with this lectin with high affinity. In vivo, mannosides are normally hydrolyzed by mannosidases: the use of a structural mimic in place of the natural sugar could avoid an easy degradation in a biological environment. The aim of this project is to design and prepare products that meet these requirements. So far we have demonstrated that the monovalent mimic 1 shown in Figure 1[3] interacts with DC-SIGN (using NMR) and inhibits the DC-SIGN mediated infection in a pseudo-typed Ebola virus model. Moreover, this molecule has been conjugated to a Boltorn-type, leading to neo-glycoconjugates that inhibit the binding of DC-SIGN to gp120 (envelope protein of HIV). In this communication we will report the synthesis of new monovalent inhibitors and the results of their binding assays by SPR. We will show also the synthesis of some multivalent compounds.     The AM Canum Venaticorum binary SDSS J173047.59+554518.5

    The AM Canum Venaticorum (AM CVn) binaries are a rare group of hydrogen-deficient, ultrashort period, mass-transferring white dwarf binaries and are possible progenitors of Type Ia supernovae. We present time-resolved spectroscopy of the recently discovered AM CVn binary SDSS J173047.59+554518.5. The average spectrum shows strong double-peaked helium emission lines, as well as a variety of metal lines, including neon; this is the second detection of neon in an AM CVn binary, after the much brighter system GP Com. We detect no calcium in the accretion disc, a puzzling feature that has been noted in many of the longer period AM CVn binaries. We measure an orbital period, from the radial velocities of the emission lines, of 35.2 ± 0.2 min, confirming the ultracompact binary nature of the system. The emission lines seen in SDSS J1730 are very narrow, although double-peaked, implying a low-inclination, face-on accretion disc; using the measured velocities of the line peaks, we estimate i ≤ 11°. This low inclination makes SDSS J1730 an excellent system for the identification of emission lines
