Product Development of Nutritious Rice Based Gluten-Free Snacks from Different Formulation of Rice Varieties by Extrusion and their Physical, Physicochemical and Sensory Evaluation
Flour from three high-nutritional rice varieties were used to produce gluten-free extruded rice snacks. This study investigated the optimal formulation of the flour mixes from brown jasmine rice (JR) and two other pigmented varieties brown black glutinous rice (BGR) and brown riceberry rice (BB) to produce the extruded snack. Chemical compositions, including dietary fiber and antioxidants of each rice variety were determined. Ten formulations of the flour mixes were evaluated using a Mixture Design and extruded via a single-screw extruder. The physical properties of the extruded snacks from each formulation such as expansion ratio, color, hardness and crispness, as well as their physicochemical properties, including water absorption and solubility indices were analyzed. Three flours showed high protein content (8.1–9.0%). Both pigmented rice (BGR and RR) indicated a higher DPPH scavenging activity at 0.51 ± 0.05 μmol Trolox/g dry weight (DW) and 0.25 μmol Trolox/g DW, respectively. BGR showed the highest % DPPH inhibition (31.63 ± 0.32%). The snack indicated the highest expansion ratio with brown riceberry rice, followed by jasmine rice and brown black glutinous rice. The use of flour mixes indicated higher overall liking scores than one specific flour, while the mixing ratio of jasmine rice, brown black glutinous rice and brown riceberry at 1: 1: 1 (Formulation 7) tended to achieve the highest overall liking (6.47 from 9 points hedonic scale) and the high scores for most of the sensory attributes. The total dietary fiber of the snacks from Formulation 7 was around 4.11% by weight. Nuclear intron sequence analysis and its implications for molecular systematics of Asian zoonotic blood flukes (Schistosoma spp.) (Trematoda: Schistosomatidae)
This study explored genetic variation among and within Schistosoma japonicum, S. mekongi, and S. malayensis populations using two intronic regions of the taurocyamine kinase (TK) gene, the bridge intron (TkBridInt) and intron 6 of domain 1 (TkD1Int6). Both introns were successfully amplified across all species and confirmed as true intronic sequences. Sequence analysis of S. japonicum revealed high nucleotide polymorphism in both introns, with the population from Sorsogon (Philippines) exhibiting the highest genetic diversity. Haplotype network analysis indicated several region-specific haplotypes. Pairwise FST estimates demonstrated significant genetic differentiation among geographical populations, reflecting limited gene flow. The observed low intraspecific but clear interspecific genetic distances further support distinct population structure. Notably, S. malayensis was genetically closer to S. mekongi than to S. japonicum. These findings highlight the potential of intron sequences in detecting intra- and interspecific variation. The findings underscore the importance of applying these markers to more recently collected and geographically comprehensive samples to better elucidate host-associated genetic structuring and transmission dynamics of these zoonotic schistosomes
Nuclear Intron Sequence Variation of the Bulinus globosus Complex (Mollusca: Planorbidae): Implications for Molecular Systematic Analyses
Urinary schistosomiasis is caused by the blood fluke Schistosoma haematobium, which is predominantly found in Africa. The freshwater snail Bulinus globosus is its main intermediate host. The species that make up the B. globosus group are genetically complex, and their taxonomic status remains controversial. Genetic variation, heterozygosity, and DNA recombination in B. globosus were examined using the mitochondrial cytochrome c oxidase subunit 1 (COI) and the intron 3 region of the arginine kinase gene (AkInt3). A total of 81 B. globosus snails were collected from three different localities in Kwale County, Kenya. Genomic diversity, heterozygosity, DNA recombination, and haplotype network were calculated using AkInt3 sequences. Low polymorphism in the COI sequence divided B. globosus into six haplotypes (C1–C6). However, AkInt3 sequencing studies showed high polymorphisms, classifying 81 B. globosus snails into 44 haplotypes (H1–H44). These haplotypes were separated into three haplogroups (I–III). AkInt3 sequence heterozygosity was also found. DNA recombination haplotypes between haplogroups were commonly found in heterozygous samples. AkInt3 sequence studies showed high levels of genetic polymorphism and heterozygosity, supporting its use as a genetic marker for elucidating the population genetics of B. globosus. Furthermore, our study showed that B. globosus populations in Kenya form a “species complex”
Stochastic cost, reliability and maintenance optimization considering uncertain future usage scenarios
A new modeling approach has been developed to optimally design a degradable system structure and maintenance plans for applications exposed to distinct stresses under different environments and diverse usage conditions. When component stresses change or shift in the future and diverse operating conditions, equipment or components may not be operating with the same stress profile that was presented when the historical data was collected and analyzed. A more detailed mathematical perspective is described to analyze the uncertainty of actual system usages in future scenarios that take variations and uncertainties explicitly into consideration. The integrated development of cost-reliability optimization models and a system maintenance policy is an interesting and challenging problem. A new integrated system optimization planning model is developed and applied to provide an insightful mathematical representation of the process that more directly represents the system reliability design process. A four-stage optimization model is constructed to accommodate sequences of decisions over time with random future usage scenarios. In the first and second stage, a two-stage stochastic model with recourse is formulated with a system cost, reliability and maintenance combined objective function to determine an initial design structure and corrective or preventive maintenance intervals. In the third-stage, the system is fielded and data is collected and analyzed to update model parameters and coefficient estimates, and adaptive preventive maintenance optimization is performed. Bayesian posterior distributions are constructed to provide the model with timely and improved estimates to reflect the fielded data. In the fourth-stage, a cost saving strategy is implemented to decide whether the current system design or a new or revised system design can provide sufficient cost savings beyond a threshold to justify design changes. The optimization models are expressed as nonlinear stochastic integer programming models where several tools are used for solving problems. In particular, pattern-search (from MATLAB toolbox), iterative coordinate descent and neighborhood heuristic search are the methods used for searching the feasible region to obtain optimal or recommended model solutions. Breakthrough infections, hospital admissions, and mortality after major COVID-19 vaccination profiles: A prospective cohort study
BACKGROUND: Several COVID-19 vaccination rollout strategies are implemented. Real-world data from the large-scale, government-mandated Central Vaccination Center (CVC), Thailand, could be used for comparing the breakthrough infection, across all available COVID-19 vaccination profiles. METHODS: This prospective cohort study combined the vaccine profiles from the CVC registry with three nationally validated outcome datasets to assess the breakthrough COVID-19 infection, hospitalization, and death among Thais individuals who received at least one dose of the COVID-19 vaccine. The outcomes were analyzed by comparing vaccine profiles to investigate the shot effect and homologous effect. FINDINGS: Of 2,407,315 Thais who had at least one dose of COVID-19 vaccine, 63,469 (2.75%) had breakthrough infection, 42,001 (1.79%) had been hospitalized, and 431 (0.02%) died. Per one vaccination shot added, there was an 18% risk reduction of breakthrough infection (adjusted hazard ratio [HR] 0.82, 95% confidence interval [CI] 0.80–0.82), a 25% risk reduction of hospitalization (HR 0.75, 95% CI 0.73–0.76), and a 96% risk reduction of mortality (HR 0.04, 95% CI 0.03–0.06). The heterologous two-shot vaccine profiles had a higher protective effect against infection, hospitalization, and mortality compared to the homologous counterparts. INTERPRETATION: COVID-19 breakthrough infection, hospitalization, and death differ across vaccination profiles that had a different number of shots and types of vaccines. To analyze and synthesize the system and the process of administration management of an Islamic Vocational College; 2. to determine guidelines for the administrative management of the Islamic Vocational College. The results were as follows : system process in term of administration management of Islamic Vocational College consisted of four departments: (1) academic department consisting of curriculum and learning management. (2) the resources department consisted of General Management; (3) to develop the Student Activities Department, consisting of Student Activity Management; 4) the Planning and Cooperation Department consisted of Cooperation Management. The guidelines for Administrative Management of an Islamic Vocational College found the following 1) that the Academic Department Management had an integrated secular curriculum and an Islamic version which followed the five dairy prayers required by Islamic law; 2) Resource Department Management added an extra group of Muslim Education Management by a Muslim director and teachers who acted as role models for students, had a prayer room and Halal food Muslim canteen and teachers and students were allowed to pray at the mosque; 3) to develop the student activities department with an integrated secular curriculum and the Islamic model followed the Muslim Sanawee Study style, with students wearing a Muslim style uniform, morals training in every subject and support of a morals project, used computers and information technology for learning, had some scholarship for successful but under privileged students; 4) the Planning and Cooperation Department used multiple methods learning communities and supported local wisdom. Description, molecular characteristics and Wolbachia endosymbionts of Onchocerca borneensis Uni, Mat Udin & Takaoka n. sp. (Nematoda: Filarioidea) from the Bornean bearded pig Sus barbatus Müller (Cetartiodactyla: Suidae) of Sarawak, Malaysia
Background: The genus Onchocerca Diesing, 1841 includes species of medical importance, such as O. volvulus (Leuckart, 1893), which causes river blindness in the tropics. Recently, zoonotic onchocercosis has been reported in humans worldwide. In Japan, O. dewittei japonica Uni, Bain & Takaoka, 2001 from wild boars is a causative agent for this zoonosis. Many filarioid nematodes are infected with Wolbachia endosymbionts which exhibit various evolutionary relationships with their hosts. While investigating the filarial fauna of Borneo, we discovered an undescribed Onchocerca species in the bearded pig Sus barbatus Müller (Cetartiodactyla: Suidae). Methods: We isolated Onchocerca specimens from bearded pigs and examined their morphology. For comparative material, we collected fresh specimens of O. d. dewittei Bain, Ramachandran, Petter & Mak, 1977 from banded pigs (S. scrofa vittatus Boie) in Peninsular Malaysia. Partial sequences of three different genes (two mitochondrial genes, cox1 and 12S rRNA, and one nuclear ITS region) of these filarioids were analysed. By multi-locus sequence analyses based on six genes (16S rDNA, ftsZ, dnaA, coxA, fbpA and gatB) of Wolbachia, we determined the supergroups in the specimens from bearded pigs and those of O. d. dewittei. Results: Onchocerca borneensis Uni, Mat Udin & Takaoka n. sp. is described on the basis of morphological characteristics and its genetic divergence from congeners. Molecular characteristics of the new species revealed its close evolutionary relationship with O. d. dewittei. Calculated p-distance for the cox1 gene sequences between O. borneensis n. sp. and O. d. dewittei was 5.9%, while that between O. d. dewittei and O. d. japonica was 7.6%. No intraspecific genetic variation was found for the new species. Wolbachia strains identified in the new species and O. d. dewittei belonged to supergroup C and are closely related. Conclusions: Our molecular analyses of filarioids from Asian suids indicate that the new species is sister to O. d. dewittei. On the basis of its morphological and molecular characteristics, we propose to elevate O. d. japonica to species level as O. japonica Uni, Bain & Takaoka, 2001. Coevolutionary relationships exist between the Wolbachia strains and their filarial hosts in Borneo and Peninsular Malaysia.[Figure not available: see fulltext.] © 2020 The Author(s)
Zoonotic infection with Onchocerca dewittei japonica in an 11-year-old boy in Kansai Region, Western Honshu, Japan
An 11-year-old boy living in Otsu City, Shiga Prefecture, Kansai Region, Western Honshu, Japan had zoonotic onchocercosis. The patient developed a painful swelling on the little finger of his left hand. The worm detected in the excised mass had external transverse ridges but did not have inner striae in the cuticle. On the basis of the parasite's histopathological characteristics, the causative agent was identified as a female Onchocerca dewittei japonica (Spirurida: Onchocercidae). The species of the filarial parasite was confirmed by sequencing the cox1 gene of the parasite. The Japanese wild boar Sus scrofa leucomystax is a definitive host for O. dewittei japonica, which is then transmitted by blackflies as the vector to humans. The current case described occurred in the Kansai Region, Western Honshu, where such infections were previously not reported
