Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. The role of regulatory B cells in milk oral immunotherapy in children with lgE-mediated cow's milk allergy
Regulatory B (Breg) cells are new players in the field of immune regulation. They have been implicated in venom immunotherapy and their role is being actively studied in non-IgE-mediated food allergies and autoimmune diseases. To date, no studies have examined the role of Breg cells in IgE-mediated milk allergy, nor has the action of Breg cells been examined in the treatment of food allergies with oral immunotherapy (OIT). Furthermore, there are currently no phenotypic, transcription factor, or lineage markers unique to Breg cells, making it a diverse and challenging focus of research. In my thesis I aimed to assess the change in peripheral Breg cells and milk-specific serum IgE, IgA, and IgG4 in children with IgE-mediated cow's milk allergy (CMA) undergoing milk OIT. Children enrolled in a clinical trial of oral immunotherapy for milk allergy were recruited at the Montreal Children's Hospital (MCH), Hôpital Ste-Justine (HSJ), BC Children's Hospital (BCCH), Chicoutimi Hospital, and the Hospital for Sick Children (SickKids). Seven out of 9 children showed an increase in PD-L1+ Breg cells from baseline to the end of escalation phase of OIT. Serum and salivary IgG4 increased significantly for all 3 milk proteins (α-lactalbumin, β-lactoglobulin, and casein) in 11 children from baseline to the end of escalation phase and further increased at 9 months post OIT (9M Post OIT), while casein-specific IgA decreased significantly at the end of escalation phase. Casein-specific IgE showed a trend to decrease at the end of escalation phase and further at 9M Post OIT. The IgG4 to IgE ratio increased significantly for β-lactoglobulin at the end of escalation phase and for casein and β-lactoglobulin at 9M Post OIT. My findings reveal that PD-L1+ Breg cells increase during milk OIT and may be part of the mechanism of successful desensitization in children. This population of Breg cells could play a role in other allergic diseases as well and future studies exploring Breg cells in such diseases are required.Les cellules B régulatoires (Breg) sont une découverte récente dans le domaine de la régulation du système immunitaire. Elles jouent un rôle important dans l'immunothérapie aux venins et leur fonction est un sujet de recherche courante des maladies autoimmunes et des allergies alimentaires non-IgE médiées. À date, aucune étude n'a examiné le rôle des cellules Breg dans les allergies alimentaires IgE, ni leurs fonctions dans le traitement des allergies alimentaires par voie d'immunothérapie orale (ITO). De plus, les cellules Breg sont difficiles à analyser puisqu'il n'y a pas de marqueurs ou phénotype uniques à ces cellules connus actuellement. L'objectif de ma thèse est d'évaluer le changement du niveau des cellules Breg et des IgE, IgA, et IgG4 spécifiques aux protéines de lait chez les enfants avec une allergie aux protéines de lait de vache qui reçoivent l'ITO au lait. Les participants étaient recrutés à l'hôpital de Montréal pour Enfants, l'Hôpital Ste-Justine, BC Children's Hospital, l'Hôpital de Chicoutimi, et SickKids. Sept des 9 enfants ont subi une haute de cellules PD-L1+ Breg à la fin de la phase d'augmentation de dose d'ITO au lait (200ml) comparé à leur point de base. L'IgG4 dans la salive et le sérum a augmenté significativement aux 3 protéines de lait (α-lactalbumine, β-lactoglobuline, et caséine) chez 11 enfants à la dose de 200ml et a continué d'augmenter significativement 9 mois plus tard pendant la phase d'entretien (9M ITO). L'IgA spécifique à la caséine a diminué significativement à la dose de 200ml et l'IgE spécifique à la caséine a eu une tendance à diminuer à la dose de 200ml, et a continué à diminuer à 9M ITO. Mes résultats montrent que les cellules PD-L1+ Breg augmentent pendant l'ITO au lait et peuvent faire partie du mécanisme de désensibilisation chez les enfants. Cette population de cellules Breg pourrait jouer un rôle important dans d'autres maladies allergiques, nécessitant plus d'études qui explorent leur rôle
Cow's milk specific lgE, lgA & lgG4 as a predictor of outcome in oral immunotherapy
Cow’s milk allergy (CMA) is one of the major food allergies among infants and young children in Western populations. CMA is defined as an immunologic adverse reaction to cow’s milk proteins. It affects 2-3% of infants. In 85% of the children, CMA resolves spontaneously within the first 5 years of life. Those with persistent CMA have a lifelong threat of anaphylaxis. 20 patients were recruited for a trial of cow’s milk (CM) oral immunotherapy (OIT). Blood samples were collected at baseline and at multiple time points during the trial. Specific IgE, IgA and IgG4 to casein, β-lactoglobulin (BLG) and α-lactalbumin (ALA) were measured in children treated with OIT using Enzyme-Linked Immunosorbent Assay (ELISA). We observed that cow’s milk specific IgE decreased by 5 to 10 times from baseline to maintenance therapy, where children are able to consume an appreciable amount of CMP. Importantly, in one patient there was no detectable sIgE 3 months post OIT. In some subjects, cow’s milk sIgE was variable. As the role of sIgA and sIgG4 is poorly understood, we measured sIgA and sIgG4 to three major CMP components in a subset of subjects. Specific-IgA increased significantly over time in 7 of 9 patients to casein, in 6 of 9 patients to BLG and ALA. Specific IgG4 to casein increased in 8 of 9 patients, and in all 9 patients in case of BLG and ALA. In summary, as patients underwent oral immunotherapy to milk, sIgA and sIgG4 increased in parallel to the decrease in IgE in most subjects tested. Our results support the observation that sIgA and sIgG4 are more important prognostic markers in specific oral tolerance induction as they are more consistent compared to sIgE. Thus, CMP specific IgA and IgG4 may help predict responses to CM OIT more accurately.L’allergie IgE-médiée au lait de vache se retrouve parmi les allergies alimentaires les plus fréquentes chez les jeunes enfants des pays industrialisés. Elle se définit par une réaction immunitaire inappropriée aux protéines retrouvées dans le lait de vache. Elle affecte 2-3% des enfants. Dans 85% des cas, l’allergie au lait de vache rentrera dans l’ordre spontanément avant l’âge de 5 ans. Par contre, ceux qui présentent une allergie persistante au lait de vache s’exposent à un risque à vie d’anaphylaxie. Nous avons recruté 20 patients dans le cadre d’un projet de recherche sur l’immunothérapie orale (ITO) au lait de vache. Un prélèvement sanguin a été effectué au début et à différents moments au cours de l’étude chez les sujets participants. Les IgE et les IgA spécifiques (IgEs/IgAs) pour la caséine, la β-lactoglobuline (BLG) et l’α-lactalbumine (ALA) ont été mesuré par technique ELISA, de même que les IgEs, les IgAs et les IgG4 spécifiques (IgG4s) pour le lait de vache chez les sujets ayant reçu l’ITO. Chez ceux ayant toléré une quantité suffisante de lait vache dans le cadre du processus d’ITO, nous avons observé une diminution de 5 à 10 fois des niveaux de base des IgEs aux protéines du lait de vache. En particulier, l’un des sujets présentait des niveaux indétectables d’IgEs après 3 mois de traitement. Il est à noter que certains sujets ont présenté des niveaux variables d’IgEs aux protéines du lait de vache au cours de l’étude. Nous avons ensuite mesuré chez un groupe de sujets les niveaux d’IgAs et d’IgG4s pour les 3 protéines allergènes principales du lait de vache afin de définir leur rôle lors du processus d’ITO. Les IgAs ont augmenté significativement au cours de l’étude chez 7 sujets sur 9 pour la caséine et 6 sujets sur 9 pour la BLG et l’ALA. En ce qui a trait aux IgG4s, les niveaux ont augmenté chez 8 sujets sur 9 pour la caséine et chez tous les 9 sujets pour la BLG et l’ALA. En conclusion, chez la majorité des sujets ayant reçu le traitement d’ITO au lait de vache, l’augmentation des niveaux d’IgAs et d’IgG4s s’est associée à une baisse parallèle des niveaux d’IgEs. Ces résultats supportent le fait que les niveaux d’IgAs et d’IgG4s sont des marqueurs pronostiques importants des résultats de l’ITO et ce, de manière plus consistante que les niveaux d’IgEs pris isolément. C’est pourquoi, le dosage des IgAs et des IgG4s pour les protéines allergènes du lait de vache peut contribuer à prédire de façon plus adéquate la réponse clinique au traitement d’ITO par le lait de vache
Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more

Immune modulation by intravenous immune globulin: cross-talk between Fc gamma receptors and regulatory T cells in allergic inflammation
Intravenous immune globulin (IVIg), a preparation of pooled human antibodies, has potent immunomodulatory properties as a treatment for inflammatory pathologies. The specific mechanisms of action are not completely understood, especially in the context of allergen-driven disease. We sought to characterize several facets of IVIg regulatory biology using an established murine model of allergic airways disease. We first examined the requirement for activating and inhibitory Fc gamma receptors for the anti-inflammatory actions of IVIg, concluding that the inhibitory Fc gamma receptor IIb expression on dendritic cells is necessary and sufficient for IVIg to tolerize the immune system.Turning to the efferent end of the regulatory response, we studied the regulatory T cell compartment by selective depletion using cell-specific transgenic deletion mice. We demonstrated the absolute requirement for regulatory T cells in the IVIg-mediated anti-inflammatory response, and that IVIg generates a specific pool of regulatory T cells de novo from non-regulatory T cell precursors. We also characterized the gene expression profile of these IVIg-generated peripheral regulatory T cells, determining that they have a unique gene expression signature.In related work, we investigated the gene expression of activated B cells in a simulated regulatory context in vitro, profiling the transcriptome and reporting a novel marker for memory B cells that appears to play a role in B cell regulatory function.We complete this thesis with pertinent literature-review manuscripts and a supplemental review chapter on the roles of dendritic cells as activators of T cells.L'immunoglobuline intraveineuse (IgIV), une préparation d'anticorps humains, possède des propriétés immunomodulatrices qui sont utilisées comme traitement pour les pathologies inflammatoires. Les mécanismes spécifiques de son action ne sont pas entièrement connus, spécialement dans le contexte des pathologies contrôlées par des antigènes. Nous avons caractérisé plusieurs aspects de la biologie régulatrice d'IgIV en utilisant un modèle murin établi de pathologie des voies aériennes. Nous avons premièrement examiné les conditions d'activation des récepteurs Fc gamma activateurs et inhibiteurs des actions anti-inflammatoires d'IgIV, en concluant que l'expression du récepteur inhibiteur Fc gamma IIb est nécessaire et suffisante pour que l'IgIV rende le système immunitaire tolérant.Se concentrant sur le côté efférent de la réponse régulatrice, nous avons étudié le compartiment des cellules T régulatrices par déplétion ciblée, en utilisant des souris transgéniques de suppression cellulaire. Nous avons démontré le besoin absolu de la présence de cellules T régulatrices dans la réponse anti-inflammatoire médiée par l'IgIV, et que l'IgIV génère une population spécifique de cellules T régulatrices de novo à partir de cellules précurseures non-régulatrices. Nous avons aussi caractérisé le profil d'expression génique de ces cellules T régulatrices périphériques générées par l'IgIV, démontrant qu'elles ont une signature distincte d'expression génique.Dans un travail connexe, nous avons étudié l'expression génique des cellules B activées dans un contexte régulateur in vitro, décrivant le profil du transcriptome. Nous décrivons un nouveau marqueur pour les cellules B mémoire qui parait jouer un rôle dans les fonctions régulatrices des cellules B.Nous complétons cette thèse avec des manuscrits de revue de littérature pertinente et un chapitre de revue sur les rôles des cellules dendritiques comme activateurs des cellules T
Role of autocrine IL-13 producing B cells in plasma cell development
Recent studies have determined that IgE-producing B lymphocytes are present in the respiratory mucosa of patients with asthma and rhinitis, but not in respiratory mucosa of normal individuals. Our laboratory has shown that B lymphocytes in human nasal mucosa stain strongly for interleukin 13 (IL-13), a crucial cytokine for airway hyper-responsiveness and remodelling, and that inhibiting IL-13 decreases IgE secretion significantly in B cells. We sought to determine the role of IL-13 in sustaining IgE-producing B lymphocytes in the absence of a lymph node-like environment. Human B lymphocytes were isolated from tonsils and purified by means of rosetting with sheep RBCs. Purified B cells were transfected by NucleofectionTM with IL-13- or IL-13Rα1-gene specific siRNA or anti-IL-13 antibodies. They were then stimulated with the LTK4A1 fibroblast line transfected with CD40-ligand or anti-CD40 antibodies, and recombinant IL-4. Total mRNA was extracted, and plasma cell related genes were measured by means of real-time PCR. Proliferation and colony formation were also observed to determine phenotypical differences. IL-13 siRNA and anti-IL-13 decreases IgE secretion in stimulated cultured B cells. Anti-IL-13 treatment also markedly increases prdm1 and april mRNA transcripts. Anti-IL-13 treatment caused stimulated B cells to produce more colonies that were smaller in size compared to control. Blocking production of IL-13 through anti-IL-13 may trigger B cells to mature into short-lived plasma cells by increasing prdm1 and april. IL-13 may be an important autocrine factor for long-lived IgE-producing B lymphocytes, and therefore be the reason for sustained IgE in the airways.Des études récentes ont déterminé que les lymphocytes B produisant l’IgE sont présents dans la muqueuse respiratoire des patients avec l'asthme et le rhinite, mais pas dans la muqueuse respiratoire d'individus normaux. Notre laboratoire a démontré que les lymphocytes B dans la muqueuse nasale humaine sont marqués fortement pour l’interleukine 13 (IL-13), une cytokine cruciale pour l’hypersensibilité des voies respiratoires et le remodelage, et en inhibant l’IL-13, la sécrétion d’IgE est diminuée de façon significative dans les cellules B. Nous avons cherché à déterminer le rôle d'IL-13 dans la production prolongée d'IgE par les lymphocytes B dans l’absence d’un environnement ressemblant à un environnement d’un ganglion lymphatique. Des lymphocytes B humaines ont été isolés d’amygdales et purifiés avec une méthode qui utilise le sang de mouton. Les cellules B purifiées ont été transfecté avec la technologie NucleofectionTM avec du ARN interférent spécifique pour l’IL-13 ou son récepteur ou avec des anticorps anti-IL-13. Ils ont été ensuite stimulés avec une lignée cellulaire de fibroblast (LTK4A1) transfectée avec le ligand CD40 ou stimulés avec des anticorps anti-CD40, et du IL-4 recombinante. L’ARNm total a été extrait, et les gènes reliés avec des cellules plasmocytaires ont été mesurés par la PCR temps réel. La prolifération et la formation des colonies ont été aussi observées pour déterminer les différences phénotypiques. L’ARN interférent d’IL-13 et l’anti-IL-13 diminue la sécrétion d'IgE dans les cellules B stimulées. De plus, le traitement d’anti-IL-13 augmente nettement les transcriptions de prdm1 et d'april. Le traitement d’anti-IL-13 a causé les cellules B stimulées à produire plus de colonies qui étaient plus petites comparée aux cellules contrôles.
