Enterocyte differentiation is compatible with SV40 large T expression and loss of p53 function in human colonic Caco-2 cells. Status of the pRb1 and pRb2 tumor suppressor gene products

Transfer of the SV40 large-T (LT) oncogene into isolated human and murine intestinal epithelial cells induced alterations of the ultrastructural organization and polarization of the resulting immortalized cell lines. We now demonstrate that the functional expression of the SV40 LT antigen in Caco-2 cells did not alter phenotypic markers of differentiation, including expression of villin, sucrase-isomaltase, brush border and dome formation. As compared to parental cells, the transfected Caco-2LT9 cells exhibited similar growth curves and no invasive properties in vitro. The major oncogenic function of the SV40 LT antigen in transfected Caco-2 cells is associated with reduced latency times necessary for the manifestation of tumors in athymic nude mice. The Caco-2 cell line contained deleted and mutant p53 alleles (stop codon in position 204) and has no detectable truncated p53 protein by Western blot. Molecular complexes between the SV40 LT antigen and the retinoblastoma-related proteins pRb1 and Rb2 were clearly identified at the different phases of the growth curve. When compared to normal human colonic crypts, Caco-2 cell differentiation is related to partial redistribution of pRb1 into hypophosphorylated, anti-proliferative forms. The pRb2 protein is found elevated in a subset of human colorectal tumors and their corresponding liver metastases. We conclude that: (1) Caco-2 cells exert a dominant control against the oncogenic functions of the LT antigen; (2) loss of p53 function is not restrictive for the establishment of polarity and differentiation of the enterocyte lineage; (3) the levels and phosphorylation status of the Rb1 and Rb2 proteins may play important roles in the proliferation and differentiation of normal and neoplastic human colonic mucosa

A critical look at the study of the relationships between disasters, media, knowledge, power and society

this paper constitutes a critical approach to i) the definition of a study object regarding the relationships between disasters, media, types of knowledge, power, and society and ii) some perspectives for the analysis of this study object. For this purpose, we used an excerpt from Michel Foucault’s work (and other works about this author), especially some texts published at the end of his life that revisit Immanuel Kant's question—raised two hundred years earlier—about what Enlightenment is. This process lead to a reflection that contributes to denaturalizing specific aspects of such study object and perspectives. The first section of the article is an introduction to the field of communication studies and their role in the relations between media, knowledge, and power, mainly in disaster situations. Second, emphasis is placed on the (sub)field of public communication of science and technology and its role in the relations between disasters, types of knowledge, power, and society. In sum, it is a question of identifying what Foucault's ideas can contribute to these standpoints and, if possible, to dialogue with them

ATT SKAPA EN AMBASSADÖR : EN UPPSATS OM VARUMÄRKESBYGGANDE INIFRÅN

    No full text
Title: To cement an ambassador - A case study on brand building from within Number of pages: 38 (40 including enclosures) Author: Alexandra Ekström Madrid, Fredrik Hermelin Tutor: Anne-Marie Morhed Course: Media and Communication Studies C Period: HT 2011 University: Division of Media and Communication, Department of Informatics and Media Uppsala University Purpose/Aim: The purpose of this case study is to examine how internal communication can be used to reinforce the external communication, that is, strengthening the brand and simplify branding. Material/Method: We have chosen to do a case study in order to get an image of the internal communication and its ability to create ambassadors for the organization. We have chosen to make the case study at S:t Eriks AB in order to determine the conditions for how to create brand ambassadors of employees. We’re comparing the empirical data we collect through the interviews at S:t Eriks with the interview with an expert on internal communication, along with our theories on the subject. By that defining the potential weaknesses of S:t Eriks internal communications and how to create brand ambassadors as well as the importance of internal communications. Main results: Our main result is that internal communication is a major part of brand building. By shaping the employees into a unified front and let them live the brand, you can avoid conflicting messages about the brand. Problems with internal communication in organizations is often based on that messages takes the wrong path. It is up to each organization to decide which brand promise is their own, and it is important that it’s clear and understood by the entire organization. The main issue in S:t Eriks internal communication is that management communicates an ambiguous picture of how the see their employees and their role, and they haven’t got a specific brand promise which they communicate. Keywords: Internal communication, brand ambassador, case study, leadership, organization

High "intelligence," low "IQ"? Speed of processing and measured IQ in children with autism

    No full text
The uneven profile of performance on standard assessments of intelligence and the high incidence of savant skills have prompted interest in the nature of intelligence in autism. The present paper reports the first group study of speed of processing in children with autism (IQ 1 SD below average) using an inspection time task. The children with autism showed inspection times as fast as an age-matched group of young normally developing children (IQ 1 SD above average). They were also significantly faster than mentally handicapped children without autism of the same age, even when these groups were pairwise matched on Wechsler IQ. To the extent that IT tasks tap individual differences in basic processing efficiency, children with autism in this study appear to have preserved information processing capacity despite poor measured IQ. These findings have implications for the role of general and specific cognitive systems in knowledge and skill acquisition: far from showing that children with autism are unimpaired, we suggest that our data may demonstrate the vital role of social insight in the development of manifest "intelligence"

Acute recurrent biliary pancreatitis associated with the ABCB4 gene mutation.

    No full text
International audienceThe ABCB4 gene codes for a protein involved in the transport of phosphatidylcholine across the canalicular membrane of the hepatocyte. ABCB4 gene defects have been associated with progressive familial intrahepatic cholestasis type 3, intrahepatic cholestasis of pregnancy, adult biliary cirrhosis and the more recently described low phospholipid associated cholelithiasis syndrome. The present paper describes 2 probands with a long history of recurrent pancreatitis and cholelithiasis and the same heterozygous, as yet undescribed del 3683>3688 within exon 28 of the ABCB4 gene resulting in a loss of function. This report shows that ABCB4 mutations may cause acute recurrent biliary pancreatitis

Studentdikt för tillfället 1660–1699 : Kvantitativa undersökningar av tryckta tillfällesdikter av Värmlandsstudenter vid Uppsala universitet

Early Modern students were prominent writers of occasional poetry. Of the 330 students matriculated 1660-1699 at Värmlands nation, Uppsala University, a total of 123 are listed as authors in the catalogue of occasional poetry at Uppsala University Library, with 373 poems attributed to them. Using quantitative methods, this study reveals the patterns of writing occasional poetry. The typical author was a reasonably successful student, intent on a career, neither poor nor of independent means. It is clearly shown that publishing occasional verse typically happens after matriculation at Uppsala but before taking up a position in society. The importance of local networks is highlighted. Typically students would return to the region where they grew up. Only a few ascended from a regional to a national context, a fact that is mirrored in the patterns of their production of occasional poetry. Four well-known Swedish authors can be found within the cohort – Petrus Lagerlöf, Olof Hermelin, Torsten Rudeen and Peter Warnmark. This quartet show an atypical publication pattern. Their output is greater, and they continue to write even after their studies are completed.

What did I say? versus What did I think?: Attributing false beliefs to self amongst children with and without autism.

    Get PDF
The task used most widely to assess recognition of false belief in self and others is the ‘Smarties’ unexpected contents task. Amongst individuals with and without autism, the Self and Other-person test questions of this task are of an equivalent level of difficulty. However, a potential confound with this task may allow the Self test question to be passed without false belief competence. Three groups of participants (with autism, developmental disability and typical development) undertook a new unexpected contents task which did not suffer from this confound. The main finding was that participants with autism performed significantly less well on the Self test question than the Other-person test question on this new task. Individuals with autism may have greater difficulty representing their own beliefs than the beliefs of other people
