A Leishmania infantum FML-ELISA for the Detection of Symptomatic and Asymptomatic Canine Visceral Leishmaniasis in an Endemic Area of Iran
Background: Visceral leishmaniasis (VL) is caused by Leishmania infantum in
Mediterranean basin and is an endemic disease in some parts of Iran. Canines are the main
reservoirs of VL in most of the endemic areas. Different serological methods have been
introduced for diagnosis of canine visceral leishmaniasis (CVL). Objective: In this survey a
Fucose-Mannose Ligand (FML) ELISA, using native L. infantum antigen, was developed and
its validity for detection of infected dogs in comparison with direct agglutination test (DAT)
and PCR was evaluated. Methods: Blood samples of sixty ownership dogs (≤ 3 years old)
were collected from Meshkin-shahr district in Ardabil province, North-west of Iran. Sera
were separated for serological assays (DAT and FML-ELISA) and the buffy coats were
collected for molecular evaluation. Results: Two out of the 60 (3.33%) samples were found
to be positive (antibody titer of ≥ 1/320) in DAT while seven of the 60 (11.66%) samples
were positive by FML-ELISA. Nine out of 60 (15%) buffy coat samples showed a band about
680 bp indicative of L. infantum in PCR. Three out of 60 dogs had Kala-azar symptoms and
were positive by PCR and FML-ELISA, while two of these three dogs had antibody titers
>1/320 in their serum samples. The sensitivity and specificity of FML-ELISA for the
detection of CVL in both symptomatic and asymptomatic dogs were found to be 77.8% and
100%, respectively. Conclusion: Considering the acceptable sensitivity and high specificity
of FML-ELISA, use of this serological method can be recommended for epidemiological
surveys of CVL
Direct replacement of antibodies with molecularly imprinted polymer (MIP) nanoparticles in ELISA - development of a novel assay for vancomycin
A simple and straightforward technique for coating microplate wells with molecularly imprinted polymer nanoparticles (nanoMIPs) to develop ELISA type assays is presented here for the first time. NanoMIPs were synthesized by a solid phase approach with immobilized vancomycin (template) and characterized using Biacore 3000, dynamic light scattering and electron microscopy. Immobilization, blocking and washing conditions were optimized in microplate format. The detection of vancomycin was achieved in competitive binding experiments with a HRP-vancomycin conjugate. The assay was capable of measuring vancomycin in buffer and in blood plasma within the range 0.001-70 nM with a detection limit of 0.0025 nM (2.5 pM). The sensitivity of the assay was three orders of magnitude better than a previously described ELISA based on antibodies. In these experiments nanoMIPs have shown high affinity and minimal interference from blood plasma components. Immobilized nanoMIPs were stored for 1 month at room temperature without any detrimental effects to their binding properties. The high affinity of nanoMIPs and the lack of a requirement for cold chain logistics make them an attractive alternative to traditional antibodies used in ELIS

Food product authentication provides a means of monitoring and identifying products for consumer protection and regulatory compliance. There is a scarcity of analytical methods for confirming the identity of fruit pulp in products containing Soft Fruit. In the present work we have developed a very sensible qualitative and quantitative method to determine the presence of berry DNAs in different food matrices. To our knowledge, this is the first study that shows the applicability, to Soft Fruit traceability, of melting curve analysis and multiplexed fluorescent probes, in a Real-Time PCR platform. Trichoderma harzianum T39 induces resistance against downy mildew by priming for defense without costs for grapevine
Downy mildew caused by Plasmopara viticola is one of the most destructive grapevine diseases worldwide. The biocontrol agent Trichoderma harzianum T39 (T39) has previously been shown to be an inducer of grapevine resistance, and we aimed at characterizing the molecular mechanisms activated by T39 and the energy costs of the induced resistance in terms of plant growth. Here, we showed that T39 reduced downy mildew severity on susceptible grapevines under controlled greenhouse conditions by a direct modulation of defense-related genes and the activation of priming for enhanced expression of these genes after pathogen inoculation. The stronger local than systemic modulation of defense-related genes corresponded to an higher local than systemic disease control in T39-treated plants. The activation of a priming state was confirmed by the absence of any negative effect of T39 on grapevine growth, shoot and root weight, leaf dimension and chlorophyll content, in contrast to benzothiadiazole (BTH) applications. Priming of defense gene expression was greater in T. harzianum T39- than in BTH-treated plants. The modulation of marker genes suggested the involvement of jasmonic acid and ethylene signals in the defense processes induced by T39, in contrast to the salicylic acid pathway activated by BTH. These results offer a greater understanding of the mechanisms underlying the grapevine induced resistance and indicate that T. harzianum T39 can be used to control downy mildew without apparent costs for grapevine growth
Coaching early career teachers in urban elementary schools: A mixed method study
Coaching for urban early career teachers (ECTs) offers promise and aligns with features of effective professional development to support the implementation of evidence-based practices. However, the functional components and key elements of coaching and coach supervision are not well specified in the literature. The goal of the current study was to examine adherence and feasibility of a coaching intervention designed to provide urban ECTs with concentrated support in classroom management and engaging learners—two instructional domains that are robust predictors of attrition (Ingersoll and Strong in Rev Educ Res 81:201–233, 2011). Coaches (n = 6) worked with ECTs (n = 15) in three urban, high-poverty elementary schools during the 2-year intervention. A mixed-method design was employed, such that qualitative data (i.e., semi-structured interviews) and quantitative data (i.e., adherence measures) were collected concurrently, remained independent during analyses, and were integrated during interpretation (Creswell and Clark in Designing and conducting mixed methods research. Sage, Thousand Oaks, 2007). Findings revealed that ECTs generally received the intended frequency and duration of coaching but with fewer opportunities for post-conferences. Coach supervision, on average, was delivered with intended frequency, with variability across coaches. Thematic analyses highlighted coach provision of emotional and instrumental support, emphasized consistent coaching as critical, and that time was a significant barrier to ECT participation in coaching. Supervision promoted social support among coaches and provided opportunities to adapt the model to ECT needs.Peer reviewe
