Role of polysialic acid in the innate immune response : a focus on macrophages, monocytes and eosinophils
Toutes les cellules de notre corps sont recouvertes d’un réseau dense de sucres complexes ou glycannes. Ces derniers participent à de nombreux processus biologiques clés incluant la communication cellulaire, la migration et l'adhésion des cellules, le trafic et l'élimination des protéines, l'activation et l'endocytose des récepteurs ainsi que la transduction des signaux. L’implication des glycannes dans ces divers processus biologiques s’explique par leur remarquable complexité et diversité et par le fait que les informations portées par les glycannes peuvent être décodées par des familles de protéines appelées lectines. Mes travaux de thèse s’inscrivent dans un ensemble de recherches en glycoimmunologie visant à comprendre le rôle des glycannes dans la régulation du développement et des fonctions effectrices des cellules de l’immunité. Au sein de mon laboratoire, nous étudions plus particulièrement la réponse immunitaire induite suite à une infection par Mycobacterium tuberculosis (Mtb), l’agent étiologique de la tuberculose. Cette dernière est la maladie causée par un agent pathogène unique la plus meurtrière dans le monde. Au cours de ma thèse, je me suis intéressé au rôle d’un polymère de sucre particulier nommé acide polysialique (PolySia) qui est synthétisé par la polysialyltransférase ST8SIA4 dans les cellules de l’immunité innée. Dans un premier temps, j’ai exploré in vitro le rôle de ST8SIA4 et du PolySia dans la régulation des fonctions effectrices du macrophage, un acteur de premier plan dans la réponse immunitaire antituberculeuse. Cependant, mes résultats n’ont pas permis de mettre en évidence un impact de l’expression de ST8SIA4 et du PolySia sur la différenciation des macrophages, sur la production de médiateurs inflammatoires suite à une stimulation par des composés bactériens, ou encore sur la capacité de ces cellules à répondre à l’infection par Mtb. Dans un deuxième temps, j’ai étudié in vivo le rôle de ST8SIA4 et du PolySia dans le développement, l'homéostasie et les fonctions effectrices des cellules immunitaires innées en comparant le phénotype de souris sauvages ou déficientes en ST8SIA4 (St8sia4-KO), à l’état basal et au cours d’une infection par Mtb. Ainsi, j’ai pu montrer que les souris St8sia4-KO présentent de nombreuses perturbations dans leur réponse immunitaire en comparaison avec les souris sauvages. En particulier, deux phénotypes majeurs ont été observés dans les souris St8sia4-KO, à savoir un nombre anormalement faible de monocytes classiques dans le sang et une accumulation d’éosinophiles au niveau de la moelle osseuse. Ces deux phénotypes présents à l’état basal se traduisent au cours de l’infection par Mtb, par un défaut de recrutement des monocytes classiques et une augmentation du nombre d’éosinophiles au niveau du tissu pulmonaire. De plus, les souris St8sia4-KO semblent mieux contenir Mtb durant les premières semaines de l’infection. D’un point de vue mécanistique, mes résultats indiquent que ST8SIA4 et le PolySia jouent un rôle essentiel pour i) la migration dépendante des chimiokines des monocytes classiques de la moelle osseuse vers le sang et le maintien d’un nombre physiologique de monocytes dans la circulation sanguine et ii) le développement des éosinophiles dans la moelle osseuse des souris. Cette étude montre pour la première fois un rôle majeur de ST8SIA4 et du PolySia dans la régulation de la réponse immunitaire innée, notamment au cours d’une infection, et ouvre la voie au développement de nouvelles thérapies ciblant le PolySia afin de moduler l’immunité et lutter contre des maladies infectieuses dévastatrices telles que la tuberculose.Every cell in our body is covered with a dense network of complex sugars or “glycans”. Tuberculosis is the deadliest disease worldwide caused by a single pathogen. During my thesis, I focused on the role of a particular sugar polymer named polysialic acid (PolySia) which is synthesized by the polysialyltransferase ST8SIA4 in innate immunity cells. First, I explored in vitro the role of ST8SIA4 and PolySia in the regulation of the effector functions of the macrophage, a key player in the anti-tuberculosis immune response. However, my results failed to demonstrate an impact of ST8SIA4 and PolySia on macrophage differentiation, on the production of inflammatory mediators following stimulation by bacterial compounds, or on their ability of these cells to respond to Mtb infection. Second, I studied in vivo the role of ST8SIA4 and PolySia in the development, homeostasis and effector functions of innate immune cells by comparing the phenotype of wild-type (WT) and St8sia4-KO mice at steady state and during Mtb infection. I found that St8sia4-KO mice have many alterations in their immune response compared to WT mice. In particular, two major phenotypes were observed in St8sia4-KO mice, namely an abnormally low number of classical monocytes in the blood and an accumulation of eosinophils in the bone marrow. Both of these phenotypes, present under basal conditions, are reflected during Mtb infection by a defect in the recruitment of classical monocytes and an increase in the number of eosinophils in the lung tissue. In addition, St8sia4-KO mice appear to better contain Mtb during the first weeks of infection. Mechanistically, my results indicate that ST8SIA4 and PolySia play a critical role in (i) chemokine-dependent migration of classical monocytes from the bone marrow into the blood and maintenance of physiological numbers of circulating classical monocytes and (ii) eosinopoiesis in the bone marrow of mice. This study shows for the first time a major role of ST8SIA4 and PolySia in the regulation of the innate immune response, especially during infection, and paves the way for the development of new therapies targeting PolySia to modulate immunity and fight devastating infectious diseases such as tuberculosis

Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more

We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
