Synthesis of non viral peptide vectors : targeting of cancer
Dans l’optique de développer de nouveaux agents bio-inspirés pour la détection et/ou le traitement des cellules cancéreuses, nos travaux se sont tournés vers la synthèse de macromolécules peptidiques complexes ayant la capacité de reconnaître les cellules tumorales. Ces travaux visent à développer des molécules permettant de cibler des particularités cellulaires présentes sur les cellules tumorales dans le but d’obtenir un traitement personnalisé via une vectorisation active permettant une augmentation de l'efficacité thérapeutique et une réduction intrinsèque de la toxicité du traitement. Pour cela, ces biomolécules doivent posséder à la fois un site de reconnaissance pour la liaison avec des protéines présentes à la surface de la cellule cible et un ou plusieurs éléments utilisés pour détecter et/ou détruire la cible. Ces systèmes ont été élaborés à partir d'un châssis moléculaire cyclodécapeptidique présentant des propriétés conformationnelles particulières. Plusieurs approches ont été envisagées. La première a consisté à rechercher de nouveaux ligands de récepteurs tumoraux en s'inspirant du domaine de reconnaissance d'un anticorps monoclonal thérapeutique. Dans ce contexte, nous avons proposé la conception de mimes du Rituximab ciblant l'antigène CD20 utilisé dans le traitement des lymphomes Non-Hodgkinien. Dans la seconde approche, nous avons développé des vecteurs destinés à des applications d'imagerie tumorale. Pour cela, des châssis multivalents présentant des ligands peptidiques RGD ciblant l'intégrine alpha-v-beta-3 ont été conjugués avec différents agents de détection puis évalués par des techniques d'imagerie telles que la TEP, la TEMP et l’imagerie optique. Toujours dans un but de diagnostic des cellules tumorales, nous nous sommes par la suite tournés vers l’application à la capture cellulaire. Pour cela, une surface d’or à été modifiée via la formation d’une monocouche organisée SAM (« Self-assembled monolayer ») présentant des cyclodextrines. Un gabarit peptidique adéquat a ainsi permis la capture et le relargage sélectif de cellules tumorales mesurées par la technique de microbalance à quartz. Ces mêmes vecteurs, validés pour le diagnostic ont par la suite été couplés à des peptides cytotoxiques issus d’une protéine pro-apoptotique « Bax ». Enfin, une dernière partie a été consacrée à la recherche de nouveaux composés comportant plusieurs éléments de ciblage tumoraux. Ces molécules présentent deux ligands de ciblage des récepteurs surexprimés sur la membrane et peuvent ainsi permettre une meilleure sélectivité vis-à-vis des tissus tumoraux.In order to develop new agents for cancer diagnosis and treatment, our work aims to synthesize complex peptide macromolecules that are able to specifically recognize cancer cells. Our goal is to increase the therapeutic efficiency and reduce the toxicity of currently available drugs using "targeted strategies". In this context, we designed sophisticated macromolecules encompassing a cell recognition domain and one or several components used to detect and/or destroy the target. This system was prepared starting from a cyclodecapeptidic scaffold presenting particular conformational properties. Different approaches were considered. First of all our work was to investigate new tumor receptor ligands based on the recognition domain of a therapeutics monoclonal antibody. We proposed the design of Rituximab mimics which targets the CD20 antigen used for the treatment of Non-Hodgkin lymphoma. In a second approach, we prepared new vectors for tumor imaging. For this purpose, multivalent scaffolds containing RGD peptide that targets alpha-v-beta-3 integrin were combined with several detection elements and evaluated by using PET, SPECT and optical imaging techniques. We also used this peptide vector for the selective cell capture and release from flowing suspensions, using a gold surface modified with a cyclodextrin-containing self-assembled monolayer (SAM). A scaffold containing ferrocenyl and -RGD- ligands permitted the selective capture and release of tumor cells. This experiment was monitored by QCM-D. This vector has been next grafted to a cytotoxic peptide that was discovered from a pro-apoptotic protein named “Bax”. Finally, we designed new molecules which include an additional ligand for the cell’s surface to increase the selectivity and the affinity of tumor tissue
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Development and industrialization of the synthesis of complex peptides
Les peptides montrent un potentiel thérapeutique important à ce jour relativement peu exploité. Dans ce contexte, les venins d’animaux représentent un sujet d’étude particulièrement intéressant puisque ceux-ci ont la particularité de contenir plusieurs centaines de toxines peptidiques structurées par un ou plusieurs ponts disulfures. Dans un premier temps isolées et identifiées à partir des venins, l’enjeu réside ensuite dans la capacité à synthétiser ces toxines par voie chimique. Cette thèse financée par l’ « Association Nationale Recherche Technologie »s’inscrit dans ce contexte et a pour objectif le développement de peptides complexes, en particulier des toxines, pour leur commercialisation. Deux problématiques principales ressortent : une limitation concernant la taille des peptides désirés, ainsi que leur structuration après synthèse chimique.La synthèse peptidique sur support solide, ou SPPS, trouve généralement sa limite dans les synthèses de peptides de plus d’une cinquantaine d’acides aminés, pour lesquels les rendements diminuent inexorablement avec l’augmentation du nombre de résidus. Afin d’accéder à des peptides et toxines plus longs, la première partie de cette thèse a été consacrée à l’étude de la native chemical ligation. Au travers de cette étude, divers peptides et toxines ne pouvant être obtenus directement par SPPS ont été synthétisés.La deuxième difficulté lors de la synthèse de toxines par voie chimique réside dans le repliement de la molécule dans sa conformation native. Les différents ponts disulfures des toxines doivent être reproduits à l’identique pour obtenir l’activité biologique attendue. Une des méthodes pour y parvenir consiste à utiliser un jeu de groupements protecteurs orthogonaux de la cystéine permettant de former les ponts disulfures un par un à travers une succession d’oxydations et de déprotections : c’est la formation dirigée de ponts disulfures. La deuxième partie de cette thèse a été consacrée à l’étude de cette méthode, son optimisation et son adaptation à différentes toxines d’intérêt. Pour ce faire, une nouvelle combinaison de groupements protecteurs a été utilisée afin de pallier aux différentes difficultés rencontrées au cours des travaux effectués.To date, Peptides present an important therapeutic potential that is poorly exploited. In this context, animal venoms are particularly interesting since they contain several hundreds of peptide toxins that are structured by one or more disulfide bridges. Initially isolated and identified from venoms, the challenge then lies in the ability to synthesize these toxins via chemical means. This thesis, supported by the “Association Nationale Recherche Technologie” aims to develop complex peptides, in particular toxins, for commercialization. Two main problems emerge: a limitation concerning the size of the desired peptides and the folding of these synthetic peptides.Solid phase peptide synthesis, or SPPS, generally finds its limit in the synthesis of peptides containing more than fifty amino acids, for which the yields decrease dramatically with the increase in the number of residues. In order to access the longest peptides and toxins, the first part of this thesis was devoted to the study of the Native Chemical Ligation. Through this study, various peptides and toxins that cannot be obtained directly by SPPS have been synthesized.The second difficulty during the synthesis of toxins by chemical means lies in the folding of the molecule into its native conformation. The natural disulfide bridges of toxins must be reproduced to obtain the expected biological activity. A method consists in the use of orthogonal protecting groups of cysteine, allowing successive disulfide bridge formations through a sequence of oxidation and deprotection steps, named directed disulfide bond formation. The second part of this thesis was therefore devoted to the study of this method, its optimization and its adaptation to various toxins of interest. For this purpose, a new combination of protecting groups was used to overcome the drawbacks encountered in these studies
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Design, synthesis and characterization of monoclonal antibody mimics
Depuis une 20aine d'années, les anticorps monoclonaux (mAb) sont apparus comme une classe prometteuse de produits pharmaceutiques pour le traitement de nombreuses pathologies dont le cancer. Cependant, peu de mAbs sont utilisés en thérapie un quart de siècle après leur découverte. Le coût de la thérapie est un facteur limitatif majeur avec des coûts annuels qui peuvent avoisiner les 100 000 euros par patient, ce qui soulève de graves préoccupations quant à leur pertinence économique. Par ailleurs, comme la plupart des protéines, de nombreux facteurs, tels que l'agrégation, leur grande taille (150 kDa), et leur immunogénicité, limitent leur utilisation en oncologie. Il y a donc un besoin urgent en systèmes moléculaires qui intègrent les propriétés des mAbs tout en s'affranchissant de ces obstacles.A partir des expertises de deux laboratoires partenaires (DCM et DPM, Grenoble), nous proposons de concevoir des mimes du mAb rituximab qui est utilisé pour traiter certains lymphomes, et dont l'épitope CD20 est parfaitement connu. Pour cela, des aptamères sélectifs de l'épitope CD20 ont été sélectionnés en utilisant l'électrophorèse capillaire (méthode de CE-SELEX). Après caractérisation in vitro notamment par ITC, les séquences ont été optimisées afin de limiter les poids moléculaires ainsi que les coûts de production. L'objectif est ensuite de greffer ces candidats sur des châssis cyclopeptidiques sous une forme multimérique via l'utilisation de réactions chimiosélectives du type ligation « oxime » et CuAAC (copper(I)-catalyzed azide alkyne cycloaddition. Les composés les plus affins sont finalement étudiés lors d'expériences biologiques préliminaires permettant de vérifier la spécificité des mimes du rituximab.In the late 90’s, the monoclonal antibodies (mAbs) became a major tool for therapeutic purposes. However, some restrictions1 provided strong arguments for the development of alternative agents like aptamers, that integrate the benefits of mAbs while circumventing these limits. As a proof of concept, we propose to design small synthetic antibody mimics of the Rituximab, uses to treat some lymphomas by targeting the CD20 antigen. In order to prepare efficient and selective mimics, a selection of recognition elements must be achieved. Aptamers were chosen because of their very low immunogenicity, their high affinity generally for their target, their structural and chemical stability and the fact that their production is carried out by organic synthesis2. Few DNA sequences were thus selected from a random ssDNA library using the systematic evolution of ligands by exponential enrichment (SELEX) method, coupled to capillary electrophoresis (CE). Among the modified SELEX methods, CE-SELEX was preferred due to its multiple benefits such as less consumption of samples, a natural binding environment, and a higher screening efficiency. The affinity and specificity of the best candidates for their target, were then characterized in solution by CE and isothermal titration calorimetry (ITC) and in cellulo using flow cytometry. Thus, the first part of this project led to the selection of specific recognition elements, selected against a short fragment of the CD20 antigen. Our main goal now, is to design dimeric or tetrameric compounds on macromolecular scaffolds using chemical ligations such as oxime or CuAAC (copper(I)-catalyzed azide alkyne cycloaddition) to assess the multivalent effects
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more
