Stepping stones in the electron transport from cells to electrodes in Geobacter sulfurreducens biofilms
Geobacter sulfurreducens bacteria grow on biofilms and have the particular ability of using polarized electrodes as the final electron acceptor of their respiratory chain. In these biofilms, electrons are transported through distances of more than 50 μm before reaching the electrode. The way in which electrons are transported across the biofilm matrix through such large distances remains under intense discussion. None of the two mechanisms proposed for explaining the process, electron hopping through outer membrane cytochromes and metallic like conduction through conductive PilA filaments, can account for all the experimental evidence collected so far. Aiming at providing new elements for understanding the basis for electron transport, in this perspective article we present a modelled structure of Geobacter pilus. Its analysis in combination with already existing experimental evidence gives support to the proposal of the "stepping stone" mechanism, in which the combined action of pili and cytochromes allows long range electron transport through the biofilm.Fil: Bonanni, Pablo Sebastian. Open circuit potentiometry reports on internal redox states of cells in G. Sulfurreducens biofilms
The outstanding capacity of Geobacter sulfurreducens cells to directly connect their inner electron transport chain to a polarized electrode have prompted the application of a wide arrange of electrochemical techniques to explore their metabolism and current production possibilities. In this work we use very basic electrochemical assays as open circuit potential, voltammetric and chronopotentiometric measurements to obtain fundamental information on bacterial electrochemical characteristics of importance to interpret bacterial functioning. Specifically, we obtained information on G. sulfurreducens biofilm capacity to store charge in three major molecular reservoirs bridging the gap between NADH and the electrode, demonstrating, at the same time, the value of OCP measurements as a reporter of internal redox state of cells.Fil: Schrott, Germán David. A long way to the electrode: how do Geobacter cells transport their electrons?
The mechanism of electron transport in Geobacter sulfurreducensbiofilms is a topic under intense study and debate. Although some proteins were found to be essential for current production, the specific role that each one plays in electron transport to the electrode remains to be elucidated and a consensus on the mechanism of electron transport has not been reached. In the present paper, to understand the state of the art in the topic, electron transport from inside of the cell to the electrode in Geobacter sulfurreducens biofilms is analysed, reviewing genetic studies, biofilm conductivity assays and electrochemical and spectro- electrochemical experiments. Limitations for current production in Geobacter sulfurreducens biofilms
Devices that exploit electricity produced by electroactive bacteria such as Geobacter sulfurreducens have not yet been demonstrated beyond the laboratory scale. The current densities are far from the maximum that the bacteria can produce because fundamental properties such as the mechanism of extracellular electron transport and factors limiting cell respiration remain unclear. In this work, a strategy for the investigation of electroactive biofilms is presented. Numerical modeling of the response of G. sulfurreducens biofilms cultured on a rotating disk electrode has allowed for the discrimination of different limiting steps in the process of current production within a biofilm. The model outputs reveal that extracellular electron transport limits the respiration rate of the cells furthest from the electrode to the extent that cell division is not possible. The mathematical model also demonstrates that recent findings such as the existence of a redox gradient in actively respiring biofilms can be explained by an electron hopping mechanism but not when considering metallic-like conductivities.Fil: Bonanni, Pablo Sebastian. Reduced depth stacked constructed wetlands for enhanced urban wastewater treatment
This work presents a new configuration for constructed wetlands (CW) for treating urban and domestic wastewater with enhanced chemical oxygen demand (COD) and nitrogen removal efficiencies. In the new configuration three stacked stages of shallow depth, operated under partial saturation replace a traditional single stage vertical down flow CW. A single pulse of wastewater produces sequential aeration steps on each stage, improving convective oxygen transfer and therefore boosting the aerobic degradation of COD and nitrogen removal via nitrification denitrification reactions. When a conductive filling was used in the systems, the treatment performance was further improved due to direct interspecies electron transfer (DIET) among bacteria in the bed. As a result, systems operated under a high organic load rate (75 g m−2 d−1), removed 92% of the COD, 80% of the ammonia nitrogen (NH4-N) and 55% of the total nitrogen (TN). Besides, under a higher loading rate of 126 g m−2 d−1, COD degradation reached 85%. Traditional single-stage conductive units showed a poorer performance, indicating that high oxygen input may be a requirement for achieving high nitrogen and COD degradation efficiencies in treatment wetlands filled with conductive materials.Fil: Cabred, Santiago. Charge accumulation and electron transfer kinetics in Geobacter sulfurreducens biofilms
Electroactive bacteria can use a polarized electrode as final electron acceptor, allowing the use of electrochemical techniques for a very accurate quantification of its respiration rate. Biofilm cell respiration has been recently demonstrated to continue after the interruption of electrode polarization since these bacteria can store electrons in the haem groups of exocytoplasmic cytochromes. Interestingly, it has been shown that when the electrode is connected again, stored electrons can be recovered as a current superimposed to the basal steady state current produced by biofilm respiration. This work presents a model for the biofilm-catalysed electron transfer mechanism that reproduces the current profile obtained upon electrode reconnection. The model allows the estimation of kinetic parameters for internalization of the reduced substrate by the cells and the subsequent reduction of cell internal cytochromes, the electron transfer to mediators in the exterior of the cell, charge transport across the biofilm matrix to the electrode through fixed mediators and, finally, the oxidation of cytochromes at the biofilm/electrode interface. Based on these estimates, the distribution of stored charge within the biofilm can also be calculated. The results indicate that the processes involved in electron transfer from acetate to internal cytochromes represent the main limitation to current production, showing that both electron transport through the matrix of cytochromes and interfacial electron transfer are orders of magnitude faster than this process. Stored charge, on the other hand, is an order of magnitude higher inside the cells compared with that in the conductive matrix, suggesting that internal cytochromes are approximately ten times more abundant inside the cells than in the conductive matrix.Fil: Bonanni, Pablo Sebastian. Energetics, electron uptake mechanisms and limitations of electroautotrophs growing on biocathodes – A review
Electroautotrophs are microorganisms that can take the electrons needed for energy generation, CO2 fixation and other metabolic reactions from a polarized electrode. They have been the focus of intense research for its application in wastewater treatment, bioelectrosynthetic processes and hydrogen generation. As a general trend, current densities produced by the electron uptake of these microorganisms are low, limiting their applicability at large scale. In this work, the electron uptake mechanisms that may operate in electroautotrophs are reviewed, aiming at finding possible causes for this low performance. Biomass yields, growth rates and electron uptake rates observed when these microorganisms use chemical electron donors are compared with those typically obtained with electrodes, to explore limitations and advantages inherent to the electroautotrophic metabolism. Also, the factors affecting biofilm development are analysed to show how interfacial interactions condition bacterial adhesion, biofilm growth and electrons uptake. Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
