    Hepatic stem cells: from inside and outside the liver?

    The liver is normally proliferatively quiescent, but hepatocyte loss through partial hepatectomy, uncomplicated by virus infection or inflammation, invokes a rapid regenerative response from all cell types in the liver to perfectly restore liver mass. Moreover, hepatocyte transplants in animals have shown that a certain proportion of hepatocytes in foetal and adult liver can clonally expand, suggesting that hepatoblasts/hepatocytes are themselves the functional stem cells of the liver. More severe liver injury can activate a potential stem cell compartment located within the intrahepatic biliary tree, giving rise to cords of bipotential transit amplifying cells (oval cells), that can ultimately differentiate into hepatocytes and biliary epithelial cells. A third population of stem cells with hepatic potential resides in the bone marrow; these haematopoietic stem cells may contribute to the albeit low renewal rate of hepatocytes, but can make a more significant contribution to regeneration under a very strong positive selection pressure. In such instances, cell fusion rather than transdifferentiation appears to be the underlying mechanism by which the haematopoietic genome becomes reprogrammed

    Oral Migalastat HCl Leads to Greater Systemic Exposure and Tissue Levels of Active α-Galactosidase A in Fabry Patients when Co-Administered with Infused Agalsidase.

    Migalastat HCl (AT1001, 1-Deoxygalactonojirimycin) is an investigational pharmacological chaperone for the treatment of α-galactosidase A (α-Gal A) deficiency, which leads to Fabry disease, an X-linked, lysosomal storage disorder. The currently approved, biologics-based therapy for Fabry disease is enzyme replacement therapy (ERT) with either agalsidase alfa (Replagal) or agalsidase beta (Fabrazyme). Based on preclinical data, migalastat HCl in combination with agalsidase is expected to result in the pharmacokinetic (PK) enhancement of agalsidase in plasma by increasing the systemic exposure of active agalsidase, thereby leading to increased cellular levels in disease-relevant tissues. This Phase 2a study design consisted of an open-label, fixed-treatment sequence that evaluated the effects of single oral doses of 150 mg or 450 mg migalastat HCl on the PK and tissue levels of intravenously infused agalsidase (0.2, 0.5, or 1.0 mg/kg) in male Fabry patients. As expected, intravenous administration of agalsidase alone resulted in increased α-Gal A activity in plasma, skin, and peripheral blood mononuclear cells (PBMCs) compared to baseline. Following co-administration of migalastat HCl and agalsidase, α-Gal A activity in plasma was further significantly increased 1.2- to 5.1-fold compared to agalsidase administration alone, in 22 of 23 patients (95.6%). Importantly, similar increases in skin and PBMC α-Gal A activity were seen following co-administration of migalastat HCl and agalsidase. The effects were not related to the administered migalastat HCl dose, as the 150 mg dose of migalastat HCl increased α-Gal A activity to the same extent as the 450 mg dose. Conversely, agalsidase had no effect on the plasma PK of migalastat. No migalastat HCl-related adverse events or drug-related tolerability issues were identified.ClinicalTrials.gov NCT01196871

    Recently, the so-called sub-6GHz band of the 5G new radio (NR) has been extended to 7.125GHz to address the relentless customer demand for higher data-rate communication. This demands a new design approach for the local area base-station (LA-BS) receivers (RXs) to cover a wide operating frequency range of 0.41 to 7.125GHz. Moreover, for NR bands above 3GHz, the maximum RF bandwidth (BW) is as high as 400MHz, in which a -35dBm modulated in-band (IB) blocker can be present. These impose stringent BW and IB linearity requirements for the baseband amplifiers in the LA-BS receivers. In addition to IB interferences, a -15dBm continuous-wave (CW) out-of-band (OOB) close-in blocker can also be present at 60MHz offset frequency from the passband edges, thus demanding a highly selective RX.     A 4 GHz third-order continuous-time ΔΣ ADC is presented with a loop filter topology that absorbs the pole caused by the input capacitance of its 4-bit quantizer and also compensates for the excess delay caused by the quantizer's latency. The ADC was implemented in 45 nm-LP CMOS and achieves 70 dB DR and -74 dBFS THD in a 125 MHz BW, while dissipating 260 mW from 1.1/1.8 V supply.     Motivet til denne avhandlingen er å verdsette selskapet BW LPG den 31.12.2023. Våre beregninger av verdien til selskapet er basert på en fundamental verdsettelse på grunnlag av deres kontantstrøm. Analyser av både strategi og regnskap har vært med på å danne grunnlaget for verdsettelsen. Vi har også valgt å supplere vår analyse med en komparativ verdsettelse, der vi har blant annet brukt P/E, P/B og EV/EBITDA. BW LPG med sin flåte på 47 skip, har siden sin børsnotering i 2013, blitt til et av verdens ledende aktører i LPG shipping markedet. Bransjen selskapet opererer i, er sterkt preget av internasjonale faktorer, konkurranse, og sykliske perioder. Den strategiske analysen forteller oss at LPG markedet er i vekst, med et økt behov for LPG shipping fremover. Gjennom PESTEL-analysen ser vi hvordan mulighetene for utvikling er i markedet, og da særlig i Asia. Samtidig er det økte fokuset på alternative energikilder som vindkraft og solceller truende for selskapets fremtid. VRIO-analysen uttrykte hvordan BW LPG ligger godt posisjonert i markedet med en bred og moderne flåte, sunn finansiell struktur, og en innovativ teknologi. Videre i VRIO-analysen ser vi at selskapet ikke har noen varige konkurransefortrinn, men holder en ledende posisjon samlet sett. Gjennom regnskapsanalysen gikk vi dypere inn i selskapets lønnsomhet, likviditet, soliditet og finansiering. I denne analysen så vi at BW LPG stiller sterkt økonomisk, med en stor egenkapitalandel og god avkastning på både egen- og totalkapital. Samtidig så vi at selskapet har tilfredsstillende likviditet, men ligger litt under bransjesnittet. Dette kan også skyldes vektingen av kortsiktig gjeld satt opp mot langsiktig gjeld. Alt i alt, virker BW LPG som et godt drevet selskap, fra et økonomisk perspektiv. Med beregning i form av multipler, oppnådde vi et betydelig lavere estimat på aksjekursen.The purpose of this assignment is to value the equity of BW LPG Limited as of 31.12.2022. The valuation is based on a fundamental valuation developed from a cash flow model of the company’s future revenue and costs. The future cash flow has been calculated from the foundation of a strategic and accounting-based analysis of the company’s operation and market. The DCF valuation is supplemented by a comparative valuation, with price/earnings, price/book value and EV/EBITDA. BW LPG has an assembled fleet of 47 Very Large Gas Carriers, which makes them the world leading LPG fleet. The company went public on Oslo Stock Exchange in 2013 and have had a steady growth where they have gained market share throughout the years. The shipping industry highly affected by international factor, competition and cyclical periods, which are considered in the valuation. The strategic analysis of the company indicates that the LPG market is growing, which will increase the demand for LPG shipping in the future. The PESTEL-analysis provided an overview of the market’s opportunities, especially in Asia. On the other hand, the market may encounter difficulties due to the increasing focus on green energy such as wind power and solar cells. The VRIO-analysis expressed that the company is good positioned due to their modern fleet, good financial structure and innovative technology. Furthermore, the VRIO implied that BW LPG doesn’t have a persistent competitive advantage, however they are collectively holding a leading position. In the account-based analysis moves deeper into the company’s profitability, liquidity, solidity and financing. The analysis communicates that BW LPG has a strong financial position, because of the majority of their capital is financed by equity, and a reasonably high return on equity and capital. They have a satisfying liquidity, but it is beneath the average of the industry. The explanation can be justified by the part of current debt is bigger than their long-term debt. In summary, BW LPG seems like a well operated company from a financial view. With the use of the fundamental valuation the company achieved a stock price on 15,84. Through a sensitivity analysis we discovered that the growth and weighted average cost of capital has the most influence on the stock price.     A 280μW dynamic-zoom ADC with 120dB DR and 118dB SNDR in 1kHz BW

    Micro-power ADCs with high linearity and dynamic range (DR) are required in several applications, such as smart sensors, biomedical imaging, and portable instrumentation. Since the signals of interest are then often small (tens of μν) and slow (<1kHz BW), such ADCs should also exhibit low offset and flicker noise. Noise-shaping SAR [1] and incremental ADCs [2] have been proposed for such applications, but their DR is limited to about 100dB. Although the ΔΣ modulator (ΔΣM) proposed in [3] achieves 136dB DR, it is at the expense of high power consumption (12.7mW). The incremental zoom ADC proposed in [4] combines a coarse SAR ADC and a fine ΔΣ ADC to efficiently achieve 119.8dB DR, but is limited to DC signals. The dynamic zoom ADC in [5] solves this problem, but requires external filtering to cope with out-of-band interference. This paper describes an interferer-robust dynamic zoom ADC that consumes 280μW while achieving 120.3dB DR and 118.1dB SNDR in 1kHz BW, resulting in a Schreier FoM of 185.8dB. It also achieves a maximum offset of 30μν and a 1/f corner of 7Hz.     Using bacterial biomarkers to identify early indicators of cystic fibrosis pulmonary exacerbation onset

    Acute periods of pulmonary exacerbation are the single most important cause of morbidity in cystic fibrosis patients, and may be associated with a loss of lung function. Intervening prior to the onset of a substantially increased inflammatory response may limit the associated damage to the airways. While a number of biomarker assays based on inflammatory markers have been developed, providing useful and important measures of disease during these periods, such factors are typically only elevated once the process of exacerbation has been initiated. Identifying biomarkers that can predict the onset of pulmonary exacerbation at an early stage would provide an opportunity to intervene before the establishment of a substantial immune response, with major implications for the advancement of cystic fibrosis care. The precise triggers of pulmonary exacerbation remain to be determined; however, the majority of models relate to the activity of microbes present in the patient's lower airways of cystic fibrosis. Advances in diagnostic microbiology now allow for the examination of these complex systems at a level likely to identify factors on which biomarker assays can be based.     The apsidal motion analysis of the eccentric eclipsing binaries: V397 Cep, V493 Car and BW Aqr have been presented. The method described by Lacy (1992) [Lacy, C.H S.. 1992. AJ 104, 2213] has been used for the apsidal motion analysis. The apsidal motion periods have been found to be 174.2 +/- 1.4, 277.3 +/- 21.3 and 7195 +/- 174 years for V397 Cep, V493 Car and BW Aqr, respectively. (C) 2009 Elsevier B.V.
