Upregulation of YKL-40 Promotes Metastatic Phenotype and Correlates with Poor Prognosis and Therapy Response in Patients with Colorectal Cancer

YKL-40 is a heparin- and chitin-binding glycoprotein that belongs to the family of glycosyl hydrolases but lacks enzymatic properties. It affects different (patho)physiological processes, including cancer. In different tumors, YKL-40 gene overexpression has been linked to higher cell proliferation, angiogenesis, and vasculogenic mimicry, migration, and invasion. Because, in colorectal cancer (CRC), the serological YKL-40 level may serve as a risk predictor and prognostic biomarker, we investigated the underlying mechanisms by which it may contribute to tumor progression and the clinical significance of its tissue expression in metastatic CRC. We demonstrated that high-YKL-40-expressing HCT116 and Caco2 cells showed increased motility, invasion, and proliferation. YKL-40 upregulation was associated with EMT signaling activation. In the AOM/DSS mouse model, as well as in tumors and sera from CRC patients, elevated YKL-40 levels correlated with high-grade tumors. In retrospective analyses of six independent cohorts of CRC patients, elevated YKL-40 expression correlated with shorter survival in patients with advanced CRC. Strikingly, high YKL-40 tissue levels showed a predictive value for a better response to cetuximab, even in patients with stage IV CRC and mutant KRAS, and worse sensitivity to oxaliplatin. Taken together, our findings establish that tissue YKL-40 overexpression enhances CRC metastatic potential, highlighting this gene as a novel prognostic candidate, a predictive biomarker for therapy response, and an attractive target for future therapy in CRC

The Epoch and Personality of Peter the Great in Nikolay Charykov’s Research

    No full text
The article reviews the academic contribution of Russian historian and diplomat Nikolay Charykov (1855-1930) to our understanding of the reforms by Peter the Great. In his research, Nikolay Charykov argues that the reforms did not result in a radical break with previous Russia's historical path. He stresses that the cultivation of scientific knowledge and labor skills necessary for modernization started in Russia long before Peter the Great because Russia was part of a European system even before Peter the Great. For example, the author points to Ivan III's marriage to Zoe Palaiologina (Sophia Palaiologina); the employment of captive Livonians by Ivan IV; sending Russian youth to study at Western European universities; Boris Godunov's plans to form a European league of Christian sovereigns. Nikolay Charykov focuses in his studies on the reign of Tsar Alexei Mikhailovich, pointing to his plans to increase the involvement of Westerners in various spheres of life in Russia. Moreover, Nikolay Charykov shows that the personality of the future Emperor was formed under the influence of Russia's increasing interaction with European countries. He concludes that the development of Russia before Peter led to Peter's reforms. He saw the struggle between Sophia and Peter as an interpersonal struggle for power, which in no way violated the primary trend of Russia's development. Peter's reforms became a logical and natural consequence of the previous processes. The merit of Peter was that he successfully continued the previously started transformations by adding to them his character and thinking, which led to the birth of the Russian Empire

Nikolay Kirsanov in the System of Characters in the Novel “Fathers and Sons” by Ivan Turgenev

    No full text
The aim of the article is to study the character of Nikolay Kirsanov and its role in the novel “Fathers and Sons” since, until recently, this character has been rather neglected, and both national and foreign critics have focused on a standard, and often schematic, enumeration of some positive qualities of the character’s personality. In order to achieve the aim and to probably reconsider the ideas and thematic plot of the novel by I. Turgenev in general, the hermeneutic, biographical and cultural-historical methods are used in the article in the context of receptive aesthetics. The suggested approach has enabled the author to argue that the personality traits of Nikolay Petrovich are determined by his relationship with Fenechka because in this case they find themselves under pressure of social inequality, significant age difference, and their mutual illegitimate son. Such an unusual behavior of a basically positive character is motivated by his belonging to the infernal sphere, which is interpreted by means of the image of “le Diable Boiteux”, i.e. “a lame devil”, with all customs and traditions of the landowners and nobility represented in it. Moreover, the character of Nikolay Petrovich Kirsanov embodied Turgenev’s personal traumatic experience concerning the writer’s illegitimate daughter. All this has allowed the author to make a conclusion that it is possible to partially reconsider most of the novel characters due to their involvement into the ambiguous situation which formed because of one of the Kirsanov brothers. The role of the character of Nikolay Petrovich and its place in the system of characters in the novel “Fathers and Sons” consists in the fact that thanks to this contradictory character, the book obtains additional pier- cing human impulse, which ensures immortality of the genius Russian writer’s literary work

Mechanisms of Understanding: Nikolay A. Rubakin’s Mnema Theory

The study provides a brief overview of the Mnema Theory, a bibliopsychological theory based on the achievements in physiology and evolutionary biology formulated by Russian bibliographer, book critic, popularizer of science and writer Nikolai Alexandrovich Rubakin (Nicholas Rubakin), and the one which allows consistently and vividly reveal the structure of the human psyche, and show the mechanisms associated with understanding in general and understanding words and text, in particular. The Theory of Mnema, although presented to the scientific community at the beginning of the 20th century, is nevertheless highly relevant after a hundred years, and there continues to be more and more evidence pointing to the veracity and credibility of its provisions from modern humanitarian and natural sciences. Nikolay A. Rubakin’s Theory of Mnema is formulated on the basis of three laws of bibliopsychology: Zemon’s Law, Humboldt-Potebnya’s Law and Teng’s Law, and shows the structure of the human psyche in the form of mnema, which is a system of records received from external stimuli. According to Nikolay A. Rubakin’s theory, the mnema are a complex open system that is in a state of permanent development, and is always being added to and is continually changing. The complex composition of the reader’s mnema determines the perception and interpretation of a text, which is also understood as the external stimulus that generates a set of engrams and their activation leading to their subsequent ecphorias, as well as the projections that a reader receives when understanding texts. The validity of Nikolay A. Rubakin’s Theory of Mnema is confirmed by psychophysiological experiments aimed at measuring the semantic proximity of words (experiments of A.R. Luria and O.S. Vinogradova), psychological and psycholinguistic experiments (for example, Rorschach tests and various kinds of associative experiments), as well as the achievements of the philological science itself with regard to the study of the space of artistic and non-artistic text, the actual images of the author and the characteristics of the interpretations of the text by readers and researchers
