Untersuchungen zur biologischen Funktion der „Processing“-α1,2-Mannosidase IA mittels „knock out“-Mäusen
Nach heutigen Kenntnissen existieren zwei ER- und drei Golgi-residente α1,2-Mannosidasen, die am „frühen“ Oligosaccharid-Processing von N-Glykoproteinen beteiligt sind. Im Gegensatz zu den beiden ER-Mannosidasen, die Glc(X)Man9GlcNAc2 zu spezifischen Man8lGlcNAc(X)Isomeren abbauen, hydrolysieren die drei Isoemzyme des Golgi (α1,2-Mannosidasen IA, IB und IC) alle α1,2-mannosidischen Bindungen in Man9-Man8GlcNAc2-Intermediaten unter Bildung von Man5GlcNAc2. Die zelluläre Bedeutung der verschiedenen α1,2-Mannosidasen ist in vielen Punkten noch unverstanden. Um die spezifische Funktion, der in unserem Arbeitskreis seit längerem untersuchten Golgi- α1,2-Mannosidase IA aufzuklären, wurde in der vorliegenden Arbeit eine „knock out“-Mauslinie hergestellt, bei der das α1,2-Mannosidase IA-Gen spezifisch ausgeschaltet war. Anschließend wurden die Auswirkungen dieses Gendefekts auf zell- und entwicklungsspezifische Prozesse in der Maus analysiert. Für die Herstellung der α1,2-Mannosidase IA-Nullmutante wurden molekular- und zellbiologische Standardtechniken verwendet. Die heterozygote und homozygot defiziente Mauslinie zeigte weder im juvenilen noch im adulten Stadium einen unmittelbar erkennbaren Phänotyp. Kreuzungsexperimente zeigten dann, dass die Man-/--Männchen normal fortpflanzungsfähig, aber die weiblichen Mannosidase IA-defizienten Mäuse infertil waren. Um den Effekt der Mannosidase IA-Defizienz näher einzugrenzen, wurden zunächst verschiedene Stadien des weiblichen Vaginalzyklus und der ovariellen Follikulogenese näher betrachtet. Hierbei konnten keine morphologischen Auffälligkeiten bzw. Unterschiede zwischen ManIA-/-- und gesunden Weibchen festgestellt werden. Untersuchungen mit postovulativen Eizellen, die aus homozygoten (ManIA-/-) Weibchen isoliert wurden, zeigten dann, dass die Man IA-/-_Eizellen nach 3-5 Stunden unter Kulturbedingungen wesentlich instabiler waren, als Eizellen aus Wildtyp-Weibchen. Wurden die postovulativen Oozyten unter Kulturbedingungen mit Spermien aus gesunden Männchen zusammengebracht war eine Zellteilung, wie sie bei Eizellen aus Wildtyp-Weibchen beobachtet weden konnte, bei den Man IA-/--Eizellen nicht nachweisbar. Es zeigte sich, dass die Spermien zwar an die ManIA-/--Eizellen andockten, aber offenbar nicht die Zona pellucida durchdringen konnten. Die Zona pellucida (ZP) ist essentiell für die Oogenese, Fertilisation und frühe Embryogenese und umgibt als extrazelluläre Matrix die Oozyte. Die Ergebnisse sprechen dafür, dass offensichtlich im Bereich der Zona pellucida bestimmte N-Glykoproteinstrukturen durch den Verlust der Enzymaktivität nicht korrekt modifiziert werden können, so dass diese funktionell wirksam sind, d.h. dass die Zona pellucida-Matrix den Bereich darstellt an dem die Mannosidase IA eingreift. Insgesamt gesehen zeigen die beschriebene Ergebnisse, dass der Verlust der Mannosidase IA-Aktivität zwar nicht die Enstehung einer ZP-Matrix verhindert, aber dass durch den Eingriff in die Biosynthesesequenz der N-gebundenen Oligosaccharidstrukturen auf den ZP-Glykoproteinen im frühen Golgi (durch das Aussschalten des Mannosidase IA-Gens) die Funktionsfähigkeit der ZP-Matrix in erheblichen Maße beeinflusst wird bzw., dass keine voll funktionsfähige Zona pellucida gebildet werden kann.</p
Charakterisierung und Funktionsanalyse der Untereinheiten des hetero-oligomeren Oligosaccharyltransferase-Komplexes aus Schweineleber
Die Oligosaccharyltransferase (OST), ein Schlusselenzym der N-Glykoproteinbiosynthese, katalysiert die Übertragung Dol-PP-aktivierter Oligosaccharide auf spezifische Asparaginreste wachsender Polypeptidketten. Die OST aus Schweineleber wurde als hetero-oligomerer Komplex gereinigt, der OST48, Ribophorin I, Ribophorin II und ein weiteres 40 kDa Protein enthielt. Daneben scheinen DAD1 und STT3 weitere Untereinheiten des OST-Komplexes darzustellen. Die spezifischen Funktionen der Untereinheiten sowie die tatsächliche Zusammensetzung des Enzym-Komplexes sind in wesentlichen Punkten noch unverstanden. Mit dem Ziel, Aminosäuren im aktiven Zentrum der OST zu identifizieren, wurden in der vorliegenden Arbeit Messungen mit verschiedenen Inhibitoren durchgeführt. Die Ergebnisse der Hemm-Messungen mit TLCK, einem Reagenz, das mit Histidinseitenketten im aktiven Zentrum von Enzymen reagieren kann, sprechen für das Vorhandensein eines Histidinrestes am aktiven Zentrum der OST und für dessen Beteiligung an der Katalyse. Untersuchungen mit einem Hexapeptid, in das zwei 5-Azido-2-nitrobenzoylgruppen eingeführt worden waren, zeigten, dass dieses Peptid ein gutes Substrat der OST darstellt und die OST in einer zeitabhängigen Reaktion zu hemmen vermag, die Hemmung war jedoch insgesamt nur mässig. Um spezifische Funktionen der Untereinheiten im OST-Komplex zu untersuchen, wurde die cDNA von DAD1 kloniert. Aktivitätsmessungen mit den verschiedenen Untereinheiten (OST48, Ribophorin I, Ribophorin II und DAD1), die in COS 1-Zellen überexprimiert worden waren, zeigten, dass die Untereinheiten für sich alleine als katalytisch inaktive Proteine synthetisiert werden, wohingegen die Koexpression von OST48 und Ribophorin I zu einem reproduzierbaren Aktivitätsanstieg von ca. 80 % -bezogen auf Kontrollzellen- führte. Dies lässt vermuten, daß OST48 und Ribophorin I direkt an der Katalyse beteiligt sind. Aktivitätsmessungen mit OST48 und Ribophorin I, die unter zellfreien Bedingungen in einem gekoppelten Transkriptions-/Translationssystem synthetisiert worden waren, haben gezeigt, daß die Synthese beider Untereinheiten zusammen auch hier zu einer zwar mässigen (~25 %igen) aber reproduzierbaren Erhoehung der OST-Aktivität fuehrte.Characterisation and functional analysis of subunits of the hetero-oligomeric oligosaccharyltransferase-complex of pig liver The oligosaccharyltransferase (OST) a key enzyme of the N-linked biosynthesis, catalyses the transfer of Dol-PP-activated oligosaccharides to selected asparagine side chains of nascent polypeptides. OST from pig liver was purified as a hetero-oligomeric complex composed of four subunits (OST48, Ribophorin I, Ribophorin II and a 40 kDa protein). DAD1 and STT3 seem to be further subunits of the mammalian OST-complex. The specific functions of the different subunits and the structure of the enzyme remain unknown at present. In order to identify amino acids in the active site of the complex OST activity was measured in the presence of different inhibitors. Studies with N-tosyl-L-lysine-chlormethylketone (TLCK), a reagent that reacts with histidine side chains at the active site of enzymes, indicate that histidine is located at the active site and is involved in catalysis. Investigations with a hexapeptide containing two photoactivatable azido groups led to an inactivation of the OST in a time-dependent manner, which was however moderate. In order to investigate specific functions of the different subunits the cDNA of DAD1 was cloned. Expression studies with the different subunits (OST48, Ribophorin I, Ribophorin II, DAD1) showed that the single subunits when over expressed in COS 1-cells were without catalytic activity. Co-expression of OST48 and Ribophorin I resulted in a reproducible increase (~80%) in OST activity, suggesting that OST48/Ribophorin I are directly involved in catalysis. Investigations with OST48 and Ribophorin I, which were expressed in vitro in a coupled transcription-/translation system, have shown that the co-expression of both proteins results in a modest (~25 %) but reproducible increase in OST activity
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more

We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
