    Invention and implementation of technologies for continuous flow synthesis

D., Massachusetts Institute of Technology, Department of Chemistry, May, 2019Cataloged from the PDF of thesis.Includes bibliographical references.In this thesis, I have optimized a synthesis of rufinamide an important epilepsy medication. This convergent synthesis generates two reactive intermediates in situ (aryl azide and propiolamide) and then combines them in a regioselective click reaction utilizing copper tubing as the catalyst. Next, I have optimized a synthesis of nicardipine which is prescribed to treat high blood pressure. The nature of the project required that the final product be relatively pure (>90 %) so that the final product could be crystallized from the reaction mixture. Nicardipine was synthesized in three steps, but in two flow reactors where one of the reactors induced two steps. The reaction mixture was then purified using two in-line aqueous extrations. First, the reaction stream was washed with HCl to produce the salt of nicardipine and wash away polar compounds. Then, the product is extracted into the aqueous layer by using a 1:1 water DMSO mixture.Finally, the synthesis's scale was increased and run in the system that was created in collaboration with the Jensen lab and Myerson lab. Next, a fully continuous synthesis of linezolid was optimized and run. The synthesis targeted the challenging intermediate amide epoxide that rapidly cyclizes into unwanted oxazolines. We were able to circumvent this side reactivity by masking the nucleophilic amide N-H by quenching the resulting nitrillium after Ritter type reaction with 2-propanol to produce the imidate. After accessing the masked amide epoxide, linezolid was produced by nucleophilic addition to the epoxide with the aniline made from a nucleophilic aromatic substitution (SNAr) reduction sequence. Finally, late stage oxazolidinone formation produces linezolid in a 73% yield in 27 minutes longest linear sequence. Next, I contributed to a system that automatically optimized and analyzed organic reactions in continuous flow.This system in collaboration with the Jensen lab fully integrated software, hardware that controlled the continuous platform, and in-line analytics. This system, after the chemist had provided the desired chemical space, could optimize a reaction without any manual intervention. Finally, I developed a monolithic cellular solid made of functionalized silica for catalyst support. This system could solve some of the problems associated with packed bed reactors including catalyst deactivation due to channeling or clogging of the reactor. This type of catalyst support could be applicable to a large number of catalysts by attaching the catalyst to silane side chains with appended functionality.     Interventions in the Delivery of Outpatient Diabetes Care: A systematic review of the literature 2001-2005

    CONTEXT: Current practices of outpatient care for type 2 diabetes are not producing adequate patient-oriented outcomes according to the targets established by clinical research trials. OBJECTIVE: To systematically review the best evidence available about the care of diabetes in the outpatient setting. METHODS: The author searched both the PubMed and CINAHL databases for reports of randomized controlled trials and systematic reviews published in English between 2001-2005 that matched the MeSH terms "diabetes" and "outpatient". Articles were then narrowed based on both relevance and quality. Included studies reported patient-oriented outcomes (blood pressure, percent hemoglobin A1c, or blood lipid levels). Interventions were divided into those affecting the setting of care, affecting the process of care, or involving primarily education. RESULTS: Of441 identified articles, 19 met all criteria and were included. Intensive process interventions were effective and generally involved multifaceted case management. Educational interventions were effective in the short term, but benefits did not extend beyond the intervention term for any study. Setting interventions showed no significant benefit. CONCLUSIONS: The literature on this topic is still inconclusive and heterogeneous. The few interventions that showed benefit are highly resource-intensive, and for this reason have significant threats to broad applicability. Successful process interventions require cost-benefit studies to determine their widespread feasibility in varied settings.     Instrumental Vibrato: An Annotated Bibliography of Historical Writings Before 1940

    abstract: The use of instrumental vibrato in certain periods of classical music performances has become a highly debated and often fiery topic. The scholars of yesterday had only a few sources with which to gain a better understanding of the definition, mechanics, employment, and prevalent attitudes of those coming before them. This project aims to develop the foundation to a better understanding of instrumental vibrato by compiling primary source material written before 1940 and secondary source material relevant to that period into an annotated bibliography. The source materials in this study were mainly comprised of treatises, tutors, method books, newspaper articles, and dictionaries. The instruments covered in this study included the violin family and relatives (viols, etc...), woodwinds (including recorder), members of the brass family, organ, other keyboard instruments, guitar/banjo/lute, theremin, and prototype/niche instruments (such as player pianos). This project investigated 309 historical documents, finding 258 contained writings about instrumental vibrato. Of those, 157 were presented as bibliographic annotations. The author found no consensus at any time in the history of Western art music between 1550-1940 that vibrato is wholly acceptable or wholly unacceptable.Dissertation/ThesisD.M.A. Music 201

    Fossil fuels, global warming and democracy: a report from a scene of the collision

    What happens to democracy when the fossil fuel industry collides with global warming? Introduction Democracy is caught in a collision between two forces: the need to respond to global warming by cutting carbon emissions, and the demands of the fossil fuel industry to increase carbon use and production. This is a slow motion collision that will take decades to conclude, though its ending seems inevitable: coal, and then oil and natural gas, will be replaced by more sustainable energy sources, but only after great damage to the environment. In this paper I explore the question, What happens to democracy when the fossil fuel industry collides with global warming? This collision is already making its marks on democratic practices. The fossil fuel industry is using every tool it can to preserve its wealth and power by pressuring governments, political parties, universities, regulators, courts, and voters. It is a process of tough, aggressive, and sophisticated politics that ultimately depends on denying the evidence that global warming poses a danger that needs to be urgently confronted. Without a theoretical framework to focus this inquiry, it could easily produce little more than a list of anecdotes about politics and influence. The value of good theory is that it reveals the patterns in the evidence, showing how the disparate pieces are connected to one another, and to larger historical, social, and economic factors. In this paper, I drew theory from (among others) Valerie Bunce, Timothy Mitchell, and most importantly Terry Lynn Karl. I use the work of these scholars to focus on the Canadian province of Alberta. Alberta provides an example of what can happen to democracy in places where fossil fuel production predominates. From time-to-time I link the paper to Australia, which depends even more than Canada on mineral extraction, and which is on the burning edge of global warming. This paper should be read as a warning to people everywhere who are concerned about fossil fuel dependence, global warming, and democracy.     PhDThis thesis considers the discrete, historically-specific theatrical and musical sub-genre of ‘Rock Opera’ as a lens through which to examine the cultural, political and social changes that are widely assumed to have characterised ‘The Sixties’ in Britain. The musical and dramatic texts, creation and production of Hair (1967), Tommy (1969), Godspell (1971), Jesus Christ Superstar (1970) and other neglected ‘Rock Operas’ of the period are analysed. Their great popularity with ‘mainstream’ audiences is considered and contrasted with the overwhelmingly negative and often internally contradictory reaction towards them from the English ‘counter-culture’. This examination offers new insights into both the ‘counter-culture’ and the ‘mainstream’ against which it claimed to define and differentiate itself. The four ‘Rock Operas’, two of which are based upon Christian scriptures, are considered as narratives of spiritual quest. The relationship between the often controversial quests for re-defined forms of faith and the apparently precipitous ‘secularization’ and ‘de-Christianization’ of British society during the 1960s and 1970s is considered. The thesis therefore analyses the ‘Rock Operas’ as significant, enlightening prisms through which to view many of the profound societal debates – over ‘faith’ and ‘belief’ in the widest senses, sexuality, the Vietnam war, generational conflict, drugs and ‘spiritual enlightenment’, and race – which were, to some considerable extent, elevated onto the national, political agenda by the activities of the broadly-defined ‘counter-culture’. It considers subsequent representations of the ‘counter-culture’ as the root of a contested but enduring popular legacy of ‘The Sixties' as a period of profound cultural change
