Caracterização do efeito da carbamazepina em modelo de neuralgia do trigêmeo em ratos machos e fêmeas

Estudos recentes demonstram que circuitos neurais específicos podem estar associados com a correlação entre dor e ansiedade, tal como a via dopaminérgica mesolímbica. Ademais, a maioria dos modelos animais para o estudo da dor envolve a aplicação de estímulos e avaliação das respostas comportamentais de dor evocada. Porém, sabese que a dor espontânea e o componente afetivo da dor são de grande relevância em condições dolorosas crônicas. Por fim, é importante ressaltar que a maioria dos estudos pré-clínicos que avaliam a eficácia de substâncias com potencial analgésico utilizam ratos machos, a despeito da maior prevalência de várias condições dolorosas crônicas no sexo feminino. No presente trabalho, avaliou-se a influência da carbamazepina na nocicepção evocada e espontânea, no aspecto afetivo/motivacional da dor e no comportamento do tipo ansioso em ratos machos e fêmeas submetidos à constrição do nervo infraorbital (CCI-ION), um modelo pré-clínico bem estabelecido de dor neuropática trigeminal. Na nocicepção evocada, avaliada pelo teste de Von Frey, a carbamazepina foi eficaz em reduzir a resposta de fêmeas e machos CCI-ION, também foi capaz de reduzir o comportamento de grooming, uma medida de nocicepção espontânea, sem influenciar a locomoção desses animais, avaliada no teste de campo aberto. Quanto à dor tônica, avaliada no teste de preferência condicionada ao lugar, apenas as fêmeas CCI-ION desenvolveram preferência à câmara pareada com a carbamazepina. As vocalizações ultrassônicas e o teste de labirinto em cruz elevado foram utilizados para avaliar o componente afetivo da dor e o comportamento do tipo-ansioso, respectivamente. Nossos resultados reforçam a teoria de que a presença de dor crônica provoca redução na emissão de vocalizações apetitivas (i.e. 50 kHz), porém o tratamento com a carbamazepina foi eficiente em restaurar esse comportamento apenas em machos. Também, no teste de labirinto em cruz elevado, apenas os machos CCI-ION apresentaram comportamento do tipo-ansioso, que não foi revertido pelo tratamento com a carbamazepina. Foram encontradas diferenças sexuais na concentração plasmática de carbamazepina, em que as fêmeas CCI-ION apresentaram uma concentração maior em relação aos machos do mesmo grupo. Por fim, a análise da concentração de dopamina no nucleo accumbens de machos e fêmeas submetidos à CCI-ION foi significativamente maior em relação aos animais falso-operados. As fêmeas apresentaram um aumento da dopamina na porção shell do nucleo accumbens, enquanto que os machos apresentaram esse aumento na porção core. Em ambos os casos o tratamento com a carbamazepina reverteu o aumento da dopamina. Esses resultados contribuem para ampliar a caracterização do modelo de CCI-ION em ratos machos e fêmeas, bem como fornecem evidências adicionais da eficácia da carbamazepina em parâmetros de dor que aumentam o valor translacional do modelo.Abstract: Trigeminal neuralgia represents a condition of severe and disabling trigeminal neuropathic pain, and in addition to living with severe pain, patients suffer from comorbidities, including anxiety and depression. Recent studies demonstrate that specific neural circuits, such as the mesolimbic dopaminergic pathway, may be associated with the correlation between pain and anxiety, Furthermore, most animal models for the study of pain involve the application of stimuli and evaluation of behavioral evoked pain responses. However, it is well known that spontaneous pain and the affective component of pain are of great relevance in chronic painful conditions. Finally, it is important to emphasize that most preclinical studies that evaluate the efficacy of substances with analgesic potential use male rats, despite the higher prevalence of several chronic painful conditions in females. In the present study, we evaluated the influence of carbamazepine on evoked and spontaneous nociception, on the affective/motivational aspect of pain and on anxiety-like behavior in male and female rats submitted to infraorbital nerve constriction (CCI-ION), a well-established experimental model of trigeminal neuropathic pain. In evoked nociception, evaluated by the Von Frey test, carbamazepine was effective in reducing mechanical hyperalgesia of females and males CCI-ION. Carbamazepine treatment also reduced the grooming behavior, a measure of spontaneous nociception, without influencing the locomotion of the animals, assessed in the open field test. As for tonic pain, evaluated in the conditioned place preference test, only CCI-ION females developed a preference for the chamber paired with carbamazepine. Ultrasonic vocalizations and the elevated plus maze test were used to assess the affective component of pain and anxiety-like behavior, respectively. Our results reinforce the idea that the presence of chronic pain causes a reduction in the emission of appetitive vocalizations (i.e. 50 kHz). Still, the treatment with carbamazepine was efficient in restoring this behavior, but only in males. As well as in the elevated plus maze test, in which only CCI-ION males showed anxious-like behavior, which was not reversed by treatment with carbamazepine. Sex differences were also found in the plasma concentration of carbamazepine, in which CCI-ION females had a higher concentration than males that received the same dosage. Finally, the analysis of dopamine concentration in the nucleus accumbens of males and females submitted to CCI-ION was significantly higher in relation to sham-operated animals. Females showed an increase in dopamine in the shell portion of the nucleus accumbens, while males showed this increase in the core portion of the nucleus accumbens. In both cases, treatment with carbamazepine reversed this increase in dopamine in rats' nucleus accumbens submitted to CCI-ION. These results contribute to broadening the characterization of the CCI-ION model in male and female rats, as well as to provide additional evidence of the effectiveness of carbamazepine in parameters that enhance the translational value of the model

Contribuição dos canais de cálcio do tipo T para os mecanismos de sinalização ganglionar relevantes na fisiopatologia da migrânea

Defesa : Curitiba, 31/10/2025Inclui referênciasResumo: A migrânea é uma condição neurológica altamente incapacitante, caracterizada por episódios de dor de cabeça pulsátil, de intensidade moderada a severa, frequentemente acompanhados por sintomas como fotofobia, fonofobia, náusea e vômito. Afeta aproximadamente 15% da população mundial, apresentando prevalência duas a três vezes maior em mulheres. A ativação do sistema trigeminovascular desempenha papel crucial na fisiopatologia da migrânea, sendo o peptídeo relacionado ao gene da calcitonina (CGRP) um dos principais mediadores envolvidos. Avanços recentes resultaram no desenvolvimento de terapias direcionadas ao CGRP, reforçando sua relevância clínica. O gânglio trigêmeo (TG) é considerado um sítio-chave na ação do CGRP, exercendo papel relevante na comunicação neuro–glia e na sensibilização dos aferentes trigeminais. Além de contribuir para a perpetuação e intensificação dos sinais dolorosos, esses mecanismos favorecem a sensibilização central, processo associado à cronificação da migrânea. Apesar desses avanços, os mecanismos moleculares e as vias de sinalização ativadas pelo CGRP no TG ainda não estão completamente elucidados. Considerando que a sinalização mediada pelo CGRP e a excitabilidade neuronal dependem criticamente do influxo de cálcio, os canais de cálcio voltagemdependentes do tipo T emergem como candidatos particularmente relevantes nesse contexto. Diferentemente de outros subtipos de canais de cálcio, os canais tipo T apresentam baixo limiar de ativação e atuam como amplificadores de sinais sublimiares, desempenhando papel fundamental na modulação da excitabilidade basal, no disparo em rajadas e na facilitação da transmissão nociceptiva. Nesse contexto, este trabalho investigou a contribuição dos canais de cálcio do tipo T, com foco nos canais CaV3.2, para os mecanismos ganglionares associados à fisiopatologia da migrânea. Ensaios de coimunoprecipitação em células tsA-201 transfectadas demonstraram que os canais CaV3.2 formam um complexo molecular com o receptor de CGRP, indicando uma interação entre essas proteínas. Nesse mesmo modelo, a expressão do receptor de CGRP promoveu inibição da corrente de cálcio, acompanhada por um desvio despolarizante nas curvas de ativação e inativação dos canais. Em contraste, a incubação de neurônios primários do TG com CGRP resultou em um aumento significativo da densidade da corrente do tipo T, aproximadamente 3,6 vezes superior aos valores basais, efeito completamente ausente em neurônios nocaute para CaV3.2. Adicionalmente, o bloqueio farmacológico dos canais de cálcio do tipo T com Z944 reduziu cerca de 80% das correntes tipo T. No nível comportamental, utilizando o modelo de injeção intraganglionar de CGRP no TG de camundongos, o tratamento com Z944 aboliu o desenvolvimento de alodinia mecânica periorbital, tanto em camundongos machos quanto fêmeas. De forma consistente, animais CaV3.2 nocaute não desenvolveram alodinia após a administração intraganglionar de CGRP. Experimentos farmacológicos adicionais sugerem que esses efeitos dependem da ativação do receptor canônico de CGRP e da subsequente ativação da proteína quinase A, culminando na modulação funcional dos canais CaV3.2. Em conjunto, os resultados indicam que o CGRP modula a atividade dos canais CaV3.2 no TG, promovendo aumento da excitabilidade neuronal e contribuindo para a sensibilização trigeminal. Esses mecanismos podem desempenhar papel relevante na gênese e na manutenção da dor durante as crises de migrânea, apontando os canais Cav3.2 como potenciais alvos terapêuticos complementares no tratamento dessa condiçãoAbstract: Migraine is a highly disabling neurological disorder characterized by recurrent episodes of pulsating headache of moderate to severe intensity, frequently accompanied by symptoms such as photophobia, phonophobia, nausea, and vomiting. It affects approximately 15% of the global population and is two to three times more prevalent in women. Activation of the trigeminovascular system plays a crucial role in migraine pathophysiology, with the calcitonin gene-related peptide (CGRP) acting as a key mediator. Recent advances have led to the development of CGRP-targeted therapies, reinforcing its clinical relevance. The trigeminal ganglion (TG) is considered a key site of CGRP action, playing an important role in neuron–glia communication and in the sensitization of trigeminal afferents. In addition to perpetuating and intensifying nociceptive signaling, these mechanisms promote central sensitization, a process associated with migraine chronification. Despite these advances, the molecular mechanisms and signaling pathways activated by CGRP in the TG remain incompletely understood. Given that CGRP-mediated signaling and neuronal excitability critically depend on calcium influx, voltage-gated T-type calcium channels emerge as particularly relevant candidates in this context. Unlike other calcium channel subtypes, T-type channels exhibit a low activation threshold and act as amplifiers of subthreshold signals, playing a fundamental role in the modulation of basal excitability, burst firing, and facilitation of nociceptive transmission. Accordingly, this study investigated the contribution of T-type calcium channels, with a specific focus on CaV3.2 channels, to ganglionic mechanisms associated with migraine pathophysiology. Co-immunoprecipitation assays performed in transfected tsA-201 cells demonstrated that CaV3.2 channels form a molecular complex with the CGRP receptor, indicating an interaction between these proteins. In the same model, expression of the CGRP receptor inhibited T-type calcium currents and induced a depolarizing shift in the voltage dependence of channel activation and inactivation. In contrast, incubation of primary TG neurons with CGRP resulted in a marked increase in T-type current density, approximately 3.6-fold above basal levels, an effect that was completely absent in neurons derived from CaV3.2 knockout mice. In addition, pharmacological blockade of T-type calcium channels with Z944 reduced approximately 80% of T-type currents. At the behavioral level, using the intraganglionic CGRP injection model in mice, treatment with Z944 abolished the development of CGRP-induced periorbital mechanical allodynia in both male and female animals. Consistently, CaV3.2 knockout mice did not develop allodynia following intraganglionic CGRP administration. Additional pharmacological experiments suggest that these effects depend on activation of the canonical CGRP receptor and subsequent activation of protein kinase A, culminating in functional modulation of CaV3.2 channels. Collectively, these findings indicate that CGRP modulates CaV3.2 channel activity in the trigeminal ganglion, promoting increased neuronal excitability and contributing to trigeminal sensitization. These mechanisms may play a relevant role in the initiation and maintenance of migraine pain, identifying CaV3.2 channels as potential complementary therapeutic targets for migraine treatmen
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more
