No full textComposto da fotografie appartenenti a tre progetti diversi – Lost in the K-Hole, Doppelgänger e Maelström – Human Surfaces è un diario visivo interpretativo che mette in luce, in modo originale, un’iconografia di varie e potenziali diversità individuali (l’alter ego, l’attore, il visionario) protagoniste di scene immaginarie ma non per questo meno obiettive. Di Donato si muove in un territorio non solo di osservazione ma di costruzione dell’immagine, a sancire che la fotografia, come tutte le forme visuali autoriali, è un’emanazione che contiene il suo artefice. Tessendo le fila delle sue visioni l’autore crea intersezioni, riconoscimenti, trasformazioni, vere e proprie descrizioni dense di ciò che immagina e ritrae, svelando le relazioni di senso affidate alle immagini e all’immaginario. Le sue Human Surfaces sono delle trasfigurazioni, metafore visive di esperienze interiori stagliate in uno spazio/vuoto esistenziale dove l’urlo angoscioso, la dissociazione e la proliferazione performativa si coniugano con il fascino della fotografia concettuale e creativa in uno stile surreale e brutale.Composed of photographs belonging to three different projects - Lost in the K-Hole, Doppelgänger and Maelström - Human Surfaces is an interpretative visual diary that highlights, in an original way, an iconography of various and potential individual diversities (the alter ego , the actor, the visionary) protagonists of imaginary scenes but no less objective. Di Donato moves in a territory not only of observation but of construction of the image, to establish that photography, like all authorial visual forms, is an emanation that contains its creator. By weaving the strings of his visions, the author creates intersections, acknowledgments, transformations, real dense descriptions of what he imagines and portrays, revealing the relationships of meaning entrusted to images and the imagination. His Human Surfaces are transfigurations, visual metaphors of inner experiences silhouetted in an existential space / void where the anguished scream, dissociation and performative proliferation combine with the charm of conceptual and creative photography in a surreal and brutal style

No full textBRAIN DAMAGE è formato da quattro progetti; quattro capitoli, “Craving”, “Doppelgänger”, “Lost in the K-Hole”, “Rorschach”, incentrati su differenti e autonome tematiche psicologiche, connesse l’una all’altra dalla presenza costante di tre elementi fondamentali: la mancanza, il desiderio, la sofferenza. Ciò che l’autore durante i tanti anni di ricerca e lavoro non ha potuto registrare istantaneamente, come nel caso di “Craving” e “Rorschach”, lo ha prima immaginato e poi ritratto alla maniera di “Doppelgänger” e “Lost in the K-Hole”. Non solo reportage, dunque, compongono il libro, ma anche regie. Per fotografare i deliri della mente, pur restando incredibilmente ancorati alla realtà, ci vuole, oltre a una profonda conoscenza dei temi affrontati, anche una potente e fervida immaginazione. Immaginazione che è, di fatto, visionarietà applicata a una logica dei sensi e dell’espressione. Queste doti artistiche Di Donato le possiede e le ha trasmesse evidentemente ai quattro progetti fotografici di Brain Damage dove le immagini delle pulsioni inconsce si sono trasformate in forme figurative inquietanti e perturbanti. Anche quando il corpo umano è nitido, integro e realistico (“Craving” e “Rorschach”), e non palesemente astratto, surreale e “defigurato” (“Doppelgänger” e “Lost in the K-Hole”), l’autore allude alla sua devastazione, distorsione ed evanescenza: il corpo e la sinestesia dei sensi sono immagini della mente e dei suoi tormenti, di alcuni traumi della condizione umana e dello stare al mondo. Nel panorama della fotografia e della cultura visuale contemporanea, Di Donato elabora immagini sinottiche e insieme narrative, storie visive riflessive che parlano in vario modo alla coscienza immaginale collettiva e individuale. Il carattere fortemente psicologico delle fotografie rende Brain Damage un’opera complessa, come complessa è la psiche umana così smarrita nei disagi dell’ipermodernità.BRAIN DAMAGE is made up of four projects; four chapters, "Craving", "Doppelgänger", "Lost in the K-Hole", "Rorschach", focusing on different and autonomous psychological themes, connected to each other by the constant presence of three fundamental elements: lack, desire, suffering. No full textActive cathepsin B has been found in cell extract and medium of human osteoblast-like cells and MG-63 cells. The released form is stable
at neutral and alkaline pH and, in both cell types, intracellular and extracellular cathepsin B activities are increased by interleukin-1 beta (IL-
1h) and parathyroid hormone (PTH). To evaluate the physiological role of cathepsin B in osteoblasts, we investigated the production and
secretion of this enzyme in normal human synovial fibroblasts and modulation by IL-1h and PTH. Lactate secretion concurrent with release
of cathepsin B and comparable responses in osteoblasts were also examined. Our data show that synovial fibroblasts respond differently to
treatment with the two agents, suggesting a cell-specific regulation of cathepsin B and possible involvement in osteoblast physiology.
Cathepsin B involvement was then evaluated in the activation of plasminogen activator (PA) in MG-63 cells using two specific inhibitors of
cathepsin B, CA074 and CA074-Me, in constitutive conditions and after treatment with IL-1h. As results of PA activity obtained in the
presence of IL-h were in contrast with previous reports, we examined the activities of PA, pro-PA activated with trypsin, and plasmin in cell
extract and media of MG-63 cells after 24-h treatment with IL-1h. Results show that in normal conditions and in the presence of IL-1h,
cathepsin B is involved in the activation of PA. Moreover, IL-1h stimulates PA, pro-PA activated by trypsin, and plasmin activity in medium,
whereas in cell extract it stimulates pro-PA activated by trypsin and plasmin activity. IL-1h has no effect on cell extract-associated PA
