    A un anno dalle elezioni che il 20 novembre 2011 hanno portato al governo il Partito popolare di Mariano Rajoy, una ricerca ideata e coordinata da Bonnie N. Field e Alfonso Botti sfocia in un volume che offre una ricostruzione della seconda legislatura di Rodríguez Zapatero (2008-2011), del tramonto della sua stella e della prima fase della nuova stagione politica.     Variability and functional role of chromosomal sequences in 16SrI-B subgroup phytoplasmas including aster yellows and related strains

    Aims: Partial genetic characterization of several chromosomal regions on 35 16SrI-B phytoplasma strains maintained in periwinkle and collected in different geographical areas from plants of diverse species. Methods and Results: Genes coding for ribosomal protein rpL22, elongation factor EF-Tu and random cloned sequences amplified with primers AY19p/m, G35p/m and BB88F1/R1 after RFLP analyses showed a high degree of polymorphism among the strains studied. The ribosomal protein (rp) subgroups B and K, and an undescribed subgroup designated N, were identified. Amplicons obtained with primers AY19p/m and BB88F1/R1, revealed a high and a low degree of polymorphism, respectively. Conclusions: A probable spacer role could be attributed to the AY19p/m sequence and a possible coding function to the BB88F1/R1 sequence. No relationship was found among genetic polymorphisms, identified by statistical analyses, and epidemiological or biological parameters. Significance and Impact of the Study: The analyses of five different genomic sequences of the 35 strains belonging to subgroup 16SrI-B allowed a finer distinction among them, confirming that the polymorphism level of 16S rDNA is too low to be adopted as unique parameter for classification

    Targeting systems to the brain obtained by merging prodrugs, nanoparticles, and nasal administration

    About 40 years ago the lipidization of hydrophilic drugs was proposed to induce their brain targeting by transforming them into lipophilic prodrugs. Unfortunately, lipidization often transforms a hydrophilic neuroactive agent into an active efflux transporter (AET) substrate, with consequent rejection from the brain after permeation across the blood brain barrier (BBB). Currently, the prodrug approach has greatly evolved in comparison to lipidization. This review describes the evolution of the prodrug approach for brain targeting considering the design of prodrugs as active influx substrates or molecules able to inhibit or elude AETs. Moreover, the prodrug approach appears strategic in optimization of the encapsulation of neuroactive drugs in nanoparticulate systems that can be designed to induce their receptor-mediated transport (RMT) across the BBB by appropriate decorations on their surface. Nasal administration is described as a valuable alternative to obtain the brain targeting of drugs, evidencing that the prodrug approach can allow the optimization of micro or nanoparticulate nasal formulations of neuroactive agents in order to obtain this goal. Furthermore, nasal administration is also proposed for prodrugs characterized by peripheral instability but potentially able to induce their targeting inside cells of the brain

    Identification of phytoplasmas associated with grapevine yellows in Serbia.

    The molecular identification and characterization of phytoplasmas infecting grapevine in four locations of different grape-growing regions of Serbia is reported. Samples from symptomatic and asymptomatic grapevine plants were collected during 2002 and 2003. Detection and identification of phytoplasmas were done by the restriction fragment length polymorphism analysis (RFLP) of polymerase chain reaction (PCR) amplified 16S rDNA. Grapevine yellows were associated with three molecularly distinguishable phytoplasmas. Flavescence dorée phytoplasmas (elm yellows group - 16SrV-C subgroup) were present only in the Župa Aleksandrovac region; in other surveyed regions Bois noir phytoplasmas (stolbur group - 16SrXII-A subgroup) were detected; a mixed infection of European stone fruit yellows (apple proliferation group - 16SrX-B subgroup) and Bois noir phytoplasmas was identified in one sample. A finer molecular characterization by the RFLP analysis on rpS3 and SecY genes of Flavescence dorée phytoplasmas from Župa Aleksandrovac allowed to verify that the Serbian genotype is indistinguishable from a strain from the Veneto region, Italy. Characterization performed on tuf gene of Bois noir phytoplasmas showed lack of amplification of samples from Erdevik. The identity of HpaII profiles on the tuf gene PCR products was observed between samples from Palić and Radmilovac, this pattern appears indistinguishable from one of the two profiles obtained in samples from the Italian grapevines used as reference strains
