    Data on in vivo phenotypes of GFRα1-positive spermatogonia stimulated by interstitial GDNF signals in mouse testes

    AbstractThis article contains the data related to the research article “in vivo dynamics of GFRα1-positive spermatogonia stimulated by GDNF signals using a bead transplantation assay” (Uchida et al., 2016) [1]. A novel transplantation assay of growth factor-soaked beads into the mammalian testicular interstitium was developed, in order to examine the effects of various soluble factors on in vivo dynamics of the spermatogonia including spermatogonial stem cells (SSC). Here we provide the image data of GFRα1-positive stem/progenitor spermatogonia in mouse seminiferous tubules near the beads soaked in GDNF (glial cell-derived neurotrophic factor), one of the SSC niche factors. The data provide various phenotypes of GFRα1-positive spermatogonia induced by bead-derived GDNF signals, which are useful to understand the active state of GFRα1-positive stem/progenitor spermatogonia in vivo

    On September 14, 2000, a brackish-water hydromedusa, Moerisia horii (Uchida and Uchida) (Hydrozoa, Anthomedusa, Moerisiidae) was found in Enoshima-Shonan Bay, Fujisawa City, Kanagawa Prefecture. The medusa is the biggest specimen of this species from Japan (6mm high of bell), with the largest number of tentacles (32). It has never been caught from the sea, therefore as a representative of a very rare specimen its morphology is described with photographs.2000年9月14日, 神奈川県藤沢市江の島湘南港にて, 汽水産のヒドロクラゲであるヒルムシロヒドラMoerisia horii(U chida and Uchida) のクラゲが1個体採集されたので報告する.この成熟クラゲはこれまでの日本産の記録中の最大個体で, 傘高が6mm あり, 触手も最多の32本を有していた.本種が海で採集されたのは初めてでたいへん希少な例である

    The effectiveness of the Uchida-Kraepelin test for psychological stress: an analysis of plasma and salivary stress substances

    Abstract Background The hypothalamic-pituitary-adrenocortical (HPA) axis and sympathetic adrenomedullary (SAM) system are the major stress-response pathways. Plasma adrenocorticotropic hormone (ACTH) represents HPA axis activity, while plasma catecholamines are used as markers of the SAM system. Salivary alpha amylase (AA), chromogranin A (CgA), and immunoglobulin A (IgA) are candidate markers of stress activation, although their role has not been established. The Uchida-Kraepelin (U-K) test is a questionnaire that requires intense concentration and effort, and has been used as a tool to induce mental stress. However, it is not clear whether or not the test is effective as a psychological/mental stressor. Methods In this study, normal young women took the U-K test and serial measurements of plasma ACTH and catecholamines (dopamine, noradrenaline, and adrenaline) (n = 10), as well as salivary AA, CgA, and IgA (n = 16) before, during and after the test. Results We found no changes in any of these parameters at any time point during or after the U-K test. Conclusion Our findings indicate that the U-K test is not a suitable for measuring the psychological/mental stress of young women because the plasma data showed that it did not affect the HPA axis and SAM system. The U-K test should be employed carefully as a psychological/mental stressor due to insufficient scientific evidence of its effectiveness. In addition, salivary AA, CgA, and IgA should not simply be compared with previous reports, because the mechanism of secretion and normal range of each salivary parameter remain unknown. Salivary AA, CgA, and IgA may not be suitable candidate markers of psychological/mental stress.</p

    Global Solvably Closed Anabelian Geometry

    In this paper, we study the pro-&#931; anabelian geometry of hyperbolic curves, where &#931; is a nonempty set of prime numbers, over Galois groups of “solvably closed extensions” of number fields — i.e., infinite extensions of number fields which have no nontrivial abelian extensions. The main results of this paper are, in essence, immediate corollaries of the following three ingredients: (a) classical results concerning the structure of Galois groups of number fields; (b) an anabelian result of Uchida concerning Galois groups of solvably closed extensions of number fields; (c) a previous result of the author concerning the pro-&#931; anabelian geometry of hyperbolic curves over nonarchimedean local fields.</p

    The role of Hic1 in testicular smooth muscle cells

    Testicular smooth muscle cells, including peritubular myoid cells and vasculature smooth muscle cells, regulate spermatogonial stem cell function in the mammalian testis. However, the mechanisms and signaling pathways that affect germ cell function largely remain to be elucidated. Here we report the expression of Hypermethylated in cancer 1 (Hic1) in testicular smooth muscle cells by using the Hic1CreERT2:RosatdTomato mice. Linage tracing experiments with Hic1CreERT2:RosatdTomato mice revealed that Hic1 expressing peritubular myoid expanded their population throughout testicular development. We hypothesized that Hic1 expression in testicular smooth muscle cells is involved in maintenance of spermatogenesis and tested this hypothesis by using aSMACreERT2:Hic1flox:RosatdTomato mice that allow conditional knock out (cKO) of Hic1 in smooth muscle cells. Deletion of Hic1 in smooth muscle cells resulted in aberrant spermatogenesis with degenerate seminiferous epithelium and dilated seminiferous tubules in mouse testis. The Hic1 cKO mice had heavier testes with more undifferentiated spermatogonia and fewer differentiated germ cells compared to controls. The peritubular myoid cells surrounding the seminiferous tubules were distributed at similar density in cKO and control mice, yet the number of Sertoli cells per seminiferous tubule in cKO mice was decreased compared to the control mice. Deposition of fibronectin, an extracellular matrix protein secreted from peritubular myoid cells, was disrupted in Hic1 cKO mice, suggesting a functional defect in Hic1 cKO peritubular myoid cells. Additionally, a tight junction protein, occludin, showed a disruption in cKO mice, indicating an indirect effect of cKO peritubular myoid cells on the germ cells, through Sertoli cells. Taken together, we demonstrated that Hic1 signaling in testicular myoid cells is playing an important role to regulate spermatogenesis. This study demonstrated the expression of Hic1 in testicular smooth muscle cells and provided insights into a signaling pathway in testicular smooth muscle cells to directly/indirectly affect germ cells
