Hydroxylation of ARD-containing substrates by T. castaneum FIH.
(A) 1 μM MBP-tagged tcFIH was tested in triplicate in the presence of 40 μM Trx-6H-tagged mmNotch1 (1862–2104) (mmNotch1 ARD) or Trx-6H-tcNotch (1747–1989) (tcNotch ARD) and their Asn to Ala mutants mmNotch1 ARD NN1945/2012AA (mmNotch1 ARD dbl mut) and tcNotch ARD NN1830/1897AA (tcNotch ARD dbl mut) by in vitro hydroxylation assay (white bars). As a control, each substrate was also tested in duplicate with buffer in place of enzyme (black bars). Model Selection with PLANN-CR-ARD
This paper presents a new compensation mechanism to be used with a Partial Logistic Artificial Neural Network for Competing Risks with Automatic Relevance Determination (PLANN-CR-ARD) and tested comprehensibly on a real breast cancer dataset with excellent convergence properties and numerical stability for the non-linear model. The Model Selection is implemented for the PLANN-CR-ARD model, benefiting from a scaling of the prior error term which together with the data error term forms the total error function that is optimized. The PLANN-CR-ARD proves to be an excellent prognostic tool that can be used in regression analysis tasks such as the survival analysis of cancer datasets

Since 1997, the ARD-DeutschlandTREND has been conducted by Infratest dimap Gesellschaft für Trend- und Wahlforschung mbH on behalf of the Association of Public Broadcasting Corporations in the Federal Republic of Germany (ARD) and various print media. From 2021, the ARD-DeutschlandTREND will be surveyed on behalf of ARD-Tagesthemen and the daily newspaper DIE WELT. Up to and including February 2021, the monthly surveys were conducted in telephone interviews (CATI) with approx. 1000 respondents (for party preference approx. 1500) per wave. Starting in March 2021, both telephone interviews (CATI) and online interviews (CAWI) were conducted with approximately 1300 respondents per wave. The attitudes and assessments of the voting population regarding parties, politicians and current political issues were recorded. Respondents for the telephone interviews were selected using a multistage random sample from the ADM selection framework for telephone sampling, including landline and mobile phone numbers (dual-frame sampling). Respondents for the online interviews were selected using a stratified random sample from an online access panel. Some questions are repeated at each survey time and asked in an identical manner, while other topics are only surveyed in one or more survey months. Evaluation of tolerance to apple replant disease (ARD) in Malus germplasm
Apple replant disease (ARD), also described as soil decline, is known for centuries; however, up to now the ultimate causes are unknown. After replanting plants of the same species, the plants are characterized by a poor vegetative development, stunted growth and reduced yield in terms of quantity and quality. For tree nurseries and orchards, replant disease is increasingly problematic and a sustainable and economically feasible solution would be the use of ARD-tolerant rootstocks. Up to now, only a few Malus genotypes with tolerance to ARD have been described and hardly any information is available about the genetic and physiological mechanisms underlying these tolerances. The discrimination of ARD tolerant and susceptible genotypes is a prerequisite for future rootstock breeding, but also for molecular studies aiming at improving the breeding efficiency. For this reason, 48 genotypes of wild apple species and rootstock cultivars were evaluated for their tolerance/susceptibility to ARD in a greenhouse bio-test. The susceptibility to ARD was classified by an ARD susceptibility index (ASI) calculated based on biomass and the increase in shoot length, respectively. The development of genetic linkage maps enables the detection of markers linked to agronomical important traits. Such associated markers can be used for an early and rapid selection of eligible plants during the breeding process. In order to map quantitative trait loci (QTL) for ARD tolerance a cross between the genotype Malus × robusta 5 (tolerant to ARD) and the rootstock M.9 (susceptible to ARD) was established and used for the construction of a first genetic linkage map including 78 SSR anchor markers. In addition, a comparative transcriptome study using mRNA from roots of M.9 and M.×robusta 5, which were cultivated on both ARD and disinfected ARD soil will provide information about potential candidate genes, which are related to ARD-tolerance
Elucidating the genetic mechanisms underlying tolerance to apple replant disease (ARD)
Apple replant disease (ARD) is a soil-borne disease, which is increasingly problematic for tree nurseries and orchards worldwide. Although, ARD is known for centuries, the ultimate causes remain unknown. The phenomenon occurs after repeated replanting of the same plant species on the same soil, leading to the soil losing its capacity to support the growth of the species. As a result, the replanted plants are characterized by a poor vegetative development, and reduced yield in terms of quantity and quality. The use of ARD-tolerant rootstocks is a sustainable approach to control this phenomenon, however, only a few Malus ARD-tolerant genotypes have been described. Moreover, less is known about the genetic mechanisms underlying these tolerances. Identifying ARD-tolerant genotypes and understanding the molecular basis for tolerance are prerequisites for future rootstock breeding. Six Malus accessions of the genebank at JKI in Dresden were previously identified as ARD-tolerant in a greenhouse bio-test. Since the previous classification was based on only one soil origin, these accessions were tested in a greenhouse bio-test using three different soil types in this study - thus confirming their ARD-tolerant status. To identify potential candidate genes related to ARD-tolerance, a comparative transcriptome study was performed. For this, the gene expression between the susceptible rootstock M.9 and the tolerant wild apple accession M. × robusta 5 were compared by RNA sequencing after cultivation on ARD-soil and disinfected (γ) ARD-soil. As a result, 81 candidate genes with a significant response in expression were selected for further validation
