    The effect of competition between two spatially separated markets - An investigation of two interlinked Bak-Sneppen models

    This paper investigates the effect of competition in a market consisting of interlinked economic agents. In particular, the effect of increased competition from the surrounding markets is demonstrated. The presented work is an extension of the Bak-Sneppen model (Bak and Sneppen 1993). Here are two Bak-Sneppen models interlinked such that if the lowest fitness value of one market exceeds the fitness values of the other market minus transportation cost, all cells lower than this band will receive a new random value. The model shows that interdependency between markets has a strong effect on the competitiveness of the least competitive market.     Op 13 en 14 maart was weer het 2-jaarlijkse NVVK-congres op de inmiddels vertrouwde locatie van Papendal. Met 588 gasten was het andermaal ‘volle bak’. Achteraf hebben 314 van hen de moeite genomen om de evaluatie in te vullen (een respons van maar liefst 53 procent). Als vereniging en als congrescommissie mogen wij beslist tevreden zijn. Waarom?     Author response: Disordered clusters of Bak dimers rupture mitochondria during apoptosis

    During apoptosis, Bak and Bax undergo major conformational change and form symmetric dimers that coalesce to perforate the mitochondrial outer membrane via an unknown mechanism. We have employed cysteine labelling and linkage analysis to the full length of Bak in mitochondria. This comprehensive survey showed that in each Bak dimer the N-termini are fully solvent-exposed and mobile, the core is highly structured, and the C-termini are flexible but restrained by their contact with the membrane. Dimer-dimer interactions were more labile than the BH3:groove interaction within dimers, suggesting there is no extensive protein interface between dimers. In addition, linkage in the mobile Bak N-terminus (V61C) specifically quantified association between dimers, allowing mathematical simulations of dimer arrangement. Together, our data show that Bak dimers form disordered clusters to generate lipidic pores. However, the molecular architecture of the mitochondrial apoptotic pore remains a key open question and especially little is known about the contribution of lipids to MOMP. By performing a comparative lipidomics analysis of the proximal membrane environment of BAK isolated in lipid nanodiscs, we find a significant enrichment of unsaturated species nearby BAK and BAX in apoptotic conditions. We then demonstrate that unsaturated lipids promote BAX pore activity in model membranes, isolated mitochondria and cellular systems, which is further supported by molecular dynamics simulations. Accordingly, the fatty acid desaturase FADS2 not only enhances apoptosis sensitivity, but also the activation of the cGAS/STING pathway downstream mtDNA release. The correlation of FADS2 levels with the sensitization to apoptosis of different lung and kidney cancer cell lines by co-treatment with unsaturated fatty acids supports the relevance of our findings. Altogether, our work provides an insight on how local lipid environment affects BAX and BAK function during apoptosis.publishersversionpublishe

    The impact of propranolol on the predictive validity of estimated VO2 max by the Ekblom-Bak submaximal cycle test : a double-blind crossover study

    Purpose Usage of beta-blockers may influence heart rate response during exercise testing. The study aimed to investigate the impact of Propranolol, a non-selective beta-blocker, on the predictive validity of a commonly used submaximal cycle test, the Ekblom-Bak test (EB-test), for the estimation of maximal oxygen uptake (VO2 max). Methods The study was a double-blinded crossover study including 28 participants (14 women), aged 21-59 years. VO2 max was estimated by the EB-test and measured during a maximal incremental exercise test during placebo and 10 mg Propranolol beta-blockade condition. The EB-test estimates VO2 max based on the difference in heart rate, Delta HR, between two work rates (4 min cycling on each), a factor corresponding to the higher rate, heart rate at the lower rate, and age. Results Maximal heart rate (mean +/- SD; 165.5 +/- 16.5 vs. 181.4 +/- 9.8 bpm), Delta HR (36.4 +/- 13.2 vs. 43.0 +/- 11.4 bpm), and measured VO2 max (48.2 +/- 6.2 vs. 50.4 +/- 7.0 ml/kg/min, 3.55 +/- 0.74 vs. 3.67 +/- 0.71 L/min) were significantly lower in the beta-blockade condition compared to placebo (P = &lt; .001). This led to an overestimation of VO2 max by the EB-test during beta-blockade, + 0.377 L/min (95% CI 0.281-0.473 L/min, + 11.2%), with no overestimation seen in the placebo condition, + 0.030 L/min (95% CI (- 0.112)-0.172 L/min + 0.8%). The coefficient of variance, indicating variance of estimated VO2 max on an individual level, was lower during beta-block condition, 6.6%, compared to placebo, 9.9%. Conclusion The EB-test systematically overestimated VO2 max compared to measured VO2 max during beta-blockade condition.
