    Uppsats för avläggande av filosofie kandidatexamen i kulturvård, bygghantverk, 22,5 hp, 2017The purpose of this thesis has been to illuminate technical aspects associated with post-medieval dovetailed ledge and batten doors, i.e. dragspondører, in Norway. In order to answer questions regarding construction, design and material use, the method of investigation has been based on literature studies, field studies and practical craft experiments. Literature studies have provided basic knowledge of the subject, whereas field studies have further broadened the knowledge through observations of actual doors. 30 ledge and batten doors from 11 different locations in Norway have been documented. The complete documentation forms the basic source material for this survey. In the practical craft experiment, wherein a copy of a 18th century dovetailed ledge and batten door was made, further practical aspects on construction, design and materials use has been highlighted. Acquired knowledge from literature and field studies, as well as craft experiments, has led to a result that to a greater extent, highlights the technical aspects of dovetailed ledge and batten doors from the post-medieval period in Norway

    Morphologic and functional correlates of synaptic pathology in the cathepsin D knockout mouse model of congenital neuronal ceroid lipofuscinosis

    Mutations in the cathepsin D (CTSD) gene cause an aggressive neurodegenerative disease (congenital neuronal ceroid lipofuscinosis) that leads to early death. Recent evidence suggests that presynaptic abnormalities play a major role in the pathogenesis of CTSD deficiencies. To identify the early events that lead to synaptic alterations, we investigated synaptic ultrastructure and function in presymptomatic CTSD knockout (Ctsd) mice. Electron microscopy revealed that there were significantly greater numbers of readily releasable synaptic vesicles present in Ctsd mice than in wild-type control mice as early as postnatal day 16. The size of this synaptic vesicle pool continued to increase with disease progression in the hippocampus and thalamus of the Ctsd mice. Electrophysiology revealed a markedly decreased frequency of miniature excitatory postsynaptic currents (mEPSCs) with no effect on paired-pulse modulation of the evoked excitatory post synaptic potentials in the hippocampus of Ctsd mice. The reduced mEPSCs frequency was observed before the appearance of epilepsy or any morphologic sign of synaptic degeneration. Taken together, these data indicate that CTSD is required for normal synaptic function and that a failure in synaptic trafficking or recycling may bean early and important pathologic mechanism in Ctsd mice; these presynaptic abnormalities may initiate synaptic degeneration in advance of subsequent neuronal loss

    Cln3(Δex7/8) mice harbor the most common genetic defect causing juvenile neuronal ceroid lipofuscinosis (JNCL), an autosomal recessive disease involving seizures, visual, motor and cognitive decline, and premature death. Here, to more thoroughly investigate the manifestations of the common JNCL mutation, we performed a broad phenotyping study of Cln3(Δex7/8) mice. Homozygous Cln3(Δex7/8) mice, congenic on a C57BL/6N background, displayed subtle deficits in sensory and motor tasks at 10-14 weeks of age. Homozygous Cln3(Δex7/8) mice also displayed electroretinographic changes reflecting cone function deficits past 5 months of age and a progressive decline of retinal post-receptoral function. Metabolic analysis revealed increases in rectal body temperature and minimum oxygen consumption in 12-13 week old homozygous Cln3(Δex7/8) mice, which were also seen to a lesser extent in heterozygous Cln3(Δex7/8) mice. Heart weight was slightly increased at 20 weeks of age, but no significant differences were observed in cardiac function in young adults. In a comprehensive blood analysis at 15-16 weeks of age, serum ferritin concentrations, mean corpuscular volume of red blood cells (MCV), and reticulocyte counts were reproducibly increased in homozygous Cln3(Δ) (ex7/8) mice, and male homozygotes had a relative T-cell deficiency, suggesting alterations in hematopoiesis. Finally, consistent with findings in JNCL patients, vacuolated peripheral blood lymphocytes were observed in homozygous Cln3(Δ) (ex7/8) neonates, and to a greater extent in older animals. Early onset, severe vacuolation in clear cells of the epididymis of male homozygous Cln3(Δ) (ex7/8) mice was also observed. These data highlight additional organ systems in which to study CLN3 function, and early phenotypes have been established in homozygous Cln3(Δ) (ex7/8) mice that merit further study for JNCL biomarker development

    Maintenance and Update Frequency: notPlannedStatement: This Batten Fault Zone Gravity Survey (P201780), gravity point data contains ground gravity point data for the NTGS Batten Fault Zone Gravity Survey (P201780) survey acquired for Northern Territory of Australia (Northern Territory Geological Survey). This dataset contains a total of 7475 point data values. The data is located in NT and were acquired in 2017. The point located data were collected in grids layout at a station spacing between 500 and 2000 metres. Terrain corrections were calculated using the INTREPID Geophysics software package. The processed data are checked by GA geophysicists using standard methods for assessing quality to ensure that the final data are fit-for-purpose. All data are provided in EPSG:4283 coordinates, Australian Height Datum (AHD) and gravity datum of AAGD07. The units are degrees, meters, and micrometres per second squared, respectively. Reference: Intrepid Geophysics, http://www.intrepid-geophysics.com.Gravity data measures small changes in gravity due to changes in the density of rocks beneath the Earth's surface. The data collected are processed via standard methods to ensure the response recorded is that due only to the rocks in the ground. The results produce datasets that can be interpreted to reveal the geological structure of the sub-surface. It is then anticipated that additional properties of Markov chains in general might come forth as a consequence of the tools presented here.     Wiley College and the Literacy Project

    In the years after the Civil War until the 1930s, blacks in America undertook a literacy project -- a vast effort, long-lived, undertaken not by the power, authority, or bureaucracy of government, but instead by blacks acting on their own, unaware of similar efforts in a thousand other places. In the project, Wiley College in Marshall, Texas, a Historically Black College, assumed the obligation to develop leaders, equip teachers, and contribute in any way possible to Negro literacy. Scholars of black colleges have lumped all black colleges together and assumed that what was true about one college was true of all. While this paper does not argue that Wiley College is a representative case study, its story is so different from widely accepted narratives about black colleges that the paper supports the need for historians to take a fresh look at accepted narratives. Commonly held narratives state that the two crucial elements of Negro college success were overall direction by northern church denominations in the 1800s and financial support from northern foundations in the early decades of the twentieth century; they deny agency to leaders and supporters of HBCs like Wiley College. The narrative I trace shows people of Wiley, its President, Matthew W.     Effectiveness of bony batten grafting for correction of caudal septal deviations

    Objective: The aim of this study was to retrospectively evaluate the efficacy of using the Batten septal bone graft intraoperatively to correct anterior septal deviations that covers Zone 1 and Zone 2 as proposed by Cottle using the NOSE scale. Methods: To this end, 30 patients with anterior septal deviations who underwent Functional Rhinoseptoplasty surgery from February 2019 to May 2023 and used septal bone grafting to correct the deviation were collected in a clinical study at the Otorhinolaryngology Department of a tertiary hospital in São Paulo. Results: There was a reduction in the Nose Scale from 75.5% to 17.3% in patients who underwent septal deviation correction using this graft (p < 0.001). Conclusion: The septal bone batten trought the endonasal approach proved to be an excellent alternative for correcting anterior deviations in patients where the simple removal of osteo-cartilaginous structures is not enough to resolve them. This graft is easy to obtain, has the ideal properties for its purpose and presented no complications related to its use in the study. Level of evidence: Level 2

    Tracking sex-dependent differences in a mouse model of CLN6-Batten disease

    Abstract Background CLN6-Batten disease is a rare neurodevelopmental disorder characterized pathologically by the accumulation of lysosomal storage material, glial activation and neurodegeneration, and phenotypically by loss of vision, motor coordination, and cognitive ability, with premature death occurring in the second decade of life. In this study, we investigate whether sex differences in a mouse model of CLN6-Batten disease impact disease onset and progression. Results A number of noteworthy differences were observed including elevated accumulation of mitochondrial ATP synthase subunit C in the thalamus and cortex of female Cln6 mutant mice at 2 months of age. Moreover, female mutant mice showed more severe behavioral deficits. Beginning at 9 months of age, female mice demonstrated learning and memory deficits and suffered a more severe decline in motor coordination. Further, compared to their male counterparts, female animals succumbed to the disease at a slightly younger age, indicating an accelerated disease progression. Conversely, males showed a marked increase in microglial activation at 6 months of age in the cortex relative to females. Conclusions Thus, as female Cln6 mutant mice exhibit cellular and behavioral deficits that precede similar pathologies in male mutant mice, our findings suggest the need for consideration of sex-based differences in CLN6 disease progression during development of preclinical and clinical studies
