PHYSICOCHEMICAL CHARACTERIZATION OF RAPAMYCIN-LOADED SOLID LIPID NANOPARTICLES FOR BRAIN TARGETING
Brain disorders such as Alzheimer’s and lysosomal storage diseases belong to the most debilitating and progressive lesions of affluence, which still remain without an effective remedy. Due to the presence of the blood-brain barrier (BBB), i.e tight cell-to-cell connections in brain capillary walls, the transport of pharmaceuticals to the central nervous system is strictly regulated and at times forbidden. One of the most crucial protein complexes, that regulates vital processes within signal transduction, proliferation and autophagy pathways in the brain cells, is the mammalian target of rapamycin complex (mTOR). Specific targeting of the mTOR by rapamycin in-feed administration has been recently demonstrated to repair the cognitative behavior in the Alzheimer’s mice model.
In light of an average size below 100 nm and lipophilic properties, P80-stabilized solid lipid nanoparticles (SLNs) have become a powerful, non-invasive strategy to overcome the BBB and release the drug directly into the brain tissue. We present a characterization of rapamycin loaded SLNs prepared using O/W emulsion - high-pressure homogenization technique and analyzed by means of Raman, UV-NIR and photon correlation spectroscopy. The effectiveness of the therapeutic application of SLN formulations will be validated in vitro on HEK293 and SH-SY5Y neuroblastoma cells and in vivo in normal and transgenic mice models
FORMULAZIONE E CARATTERIZZAZIONE DI NUOVI TIPI DI NANOPARTICELLE PER STRATEGIE DI “DRUG DELIVERY” A LIVELLO CEREBRALE
Attualmente le malattie neurodegenerative e cerebrovascolari hanno un impatto crescente sia dal punto di vista sociale che economico. Pertanto lo sviluppo di nuove strategie per una diagnosi precoce e per un trattamento efficace e sicuro di tali patologie sta diventando sempre più importante. Per queste ragioni, la ricerca di sistemi in grado di passare la barriera emato-encefalica per il trasporto di molecole terapeutiche a livello di sistema nervoso centrale (SNC) rappresenta una delle sfide più impegnative. La selettività della barriera emato-encefalica limita fortemente il numero di sostanze terapeutiche in grado di raggiungere il cervello e pertanto, negli ultimi anni, molti sforzi vengono rivolti allo sviluppo di sistemi che facilitino il passaggio dei farmaci verso il SNC. Tra questi, l’approccio basato sulle nanotecnologie sembra essere molto promettente. Tale approccio comporta l'uso di almeno due componenti, uno dei quali è costituito da una nanoparticella, che funge da vettore (nano-carrier), e l'altro è l'agente terapeutico (cargo).
La ricerca in atto ha lo scopo di indagare la potenziale efficacia di nanoparticelle solido-lipidiche coniugate con il farmaco Rapamicina (Rp-SLN) per la cura delle malattie neurodegenerative. Recentemente è stato dimostrato che l’inibizione specifica di mTOR, un complesso molecolare che riveste un ruolo centrale nel controllo della omeostasi cellulare, da parte della Rapamicina (Rp) è in grado di migliorare il comportamento cognitivo nel modello murino della Malattia di Alzheimer. mTOR è una serina / treonina chinasi implicata nella regolazione del metabolismo energetico, della proliferazione e motilità cellulare e dell'autofagia. mTOR forma due diversi complessi enzimatici,

mTORC1 e mTORC2. mTORC1 integra stimoli diversi provenienti da nutrienti, fattori di crescita, stato energetico e stress, interagendo strettamente con il complesso TSC1-TSC2 [Maiese et al., 2013 Trends Mol Med. 195, 1-60]. La Rp è un farmaco immunomodulatore approvato dalla US Food and Drug Administration per l'uso clinico come immunosoppressore. Attualmente viene usato per indurre tolleranza in pazienti che hanno subito trapianto d’ organo, ma anche in oncologia per il trattamento del carcinoma renale avanzato. Studi recenti dimostrano che il trattamento con Rp, o con il suo analogo Everolimus, apporta benefici neurologici nella Sclerosi Tuberosa (TS) [Krueger et al. 2010 N Engl J Med 363,1801-11], una malattia genetica associata allo sviluppo di tumori al cervello e nella Malattia di Alzheimer [Caccamo et al. J Biol Chem 2010, 285, 13.107-20]. La Rp ed il suo analogo sono in grado di passare la barriera emato-encefalica ed apportare significativi benefici nelle due patologie neurodegenerative sopra citate, ma la loro somministrazione sistemica produce una serie di effetti indesiderati che possono compromettere la loro efficacia terapeutica, soprattutto se si pensa ad una loro somministrazione cronica. D’altro canto una somministrazione in loco sarebbe troppo invasiva e non consona ad un trattamento prolungato e precoce, quando ancora la sintomatologia non è conclamata. Al fine di mettere a punto una strategia terapeutica a bassa invasività basata su un sistema di nano-carrier specifico in grado di superare la barriera emato-encefalica e rilasciare in modo efficace una dose terapeutica del farmaco, abbiamo sintetizzato nanoparticelle solido-lipidiche caricate con Rp e opportunamente funzionalizzate con polisorbato 80 (P80). Il P80 ha il duplice ruolo di stabilizzatore e modificatore di superficie, in grado di favorire l'adsorbimento delle apolipoproteine sulla superficie delle particelle, convogliando così le stesse nel pathway dei recettori delle LDL presenti sulla barriera emato-encefalica e consentendone così l’attraversamento ed il rilascio del loro contenuto (che in questo caso era rappresentato da un colorante) a livello di sistema nervoso centrale [Blasi 2007Advanced Drug Delivery Recensioni 59, 454-477]. Le Rp-SLN sono state da noi preparate con il metodo della omogeneizzazione a freddo ad alta pressione. Il Cetil-palmitato e la Rp (10 e 20% p/p) sono stati sciolti in cloroformio e adeguatamente miscelati con una soluzione acquosa di P80 all’1% (p/v) per ottenere una microemulsione olio-acqua. La produzione delle Rp-SLN consiste in 5 cicli di omogeneizzazione ad alta pressione. Il cloroformio è stato allontanato o over-night a temperatura ambiente oppure mediante evaporazione rapida sotto vuoto. Misure di light-scattering dinamico hanno evidenziato che la dimensione media delle particelle è tra 50 e 200 nm. Il potenziale zeta è risultato essere neutro per le preparazioni ottenute mediante evaporazione rapida, mentre con tempi di evaporazione lunghi le Rp-SLN risultano cariche negativamente. La differenza osservata è correlata alla quantità di farmaco presente sulla superficie delle particelle che aumenta con il tempo di caricamento. L’intrappolamento del farmaco è stato valutato mediante spettroscopia UV. Misure della Rp nel

sopranatante opportunamente centrifugato e filtrato hanno evidenziato una efficienza di incapsulamento compresa tra 84 e 90%. Inoltre è stata effettuata una analisi TEM per controllare la morfologia delle particelle ed escludere la presenza di nanocristalli di Rp.
La eventuale citotossicità delle Rp-SLN ed il grado di internalizzazione sono stati analizzati in vitro in cellule SH-SY5Y (modello cellulare neurale) ed in cellule staminali neuronali. In queste ultime, da noi prodotte a partire dal modello murino di Alzheimer o da quello di Sclerosi Tuberosa, è stato valutato il livello di inibizione di mTOR dopo esposizione alle Rp-SLN e quindi l’effetto terapeutico. Sulla base dei dati ottenuti abbiamo ora intrapreso la preparazione di Rp-SLN marcate con una sonda fluorescente per valutare in vivo la loro capacità di oltrepassare la barriera-ematoencefalica e di trasportare le dosi terapeutiche di Rp a livello di sistema nervoso centrale
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Dispelling the Myths Behind First-author Citation Counts
We conducted a full-scale evaluative citation analysis study of scholars in the XML research field to explore just how different from each other author rankings resulting from different citation counting methods actually are, and to demonstrate the capability of emerging data and tools on the Web in supporting more realistic citation counting methods. Our results contest some common arguments for the continued
use of first-author citation counts in the evaluation of scholars, such as high correlations between author rankings by first-author citation counts and other citation
counting methods, and high costs of using more realistic citation counting methods that are not well-supported by the ISI databases. It is argued that increasingly available digital full text research papers make it possible for citation analysis studies to go beyond what the ISI databases have directly supported and to employ more

We have done our best to complete the author checklist relating to the use of animals in the hut study. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
