    Biomechanical signals and the C-type natriuretic peptide counteract catabolic activities induced by IL-1? in chondrocyte/agarose constructs

    Introduction: The present study examined the effect of C-type natriuretic peptide (CNP) on the anabolic and catabolic activities in chondrocyte/agarose constructs subjected to dynamic compression. Methods: Constructs were cultured under free-swelling conditions or subjected to dynamic compression with low (0.1 to 100 pM) or high concentrations (1 to 1,000 nM) of CNP, interleukin-1? (IL-1?), and/or KT-5823 (inhibits cyclic GMP-dependent protein kinase II (PKGII)). Anabolic and catabolic activities were assessed as follows: nitric oxide (NO) and prostaglandin E2 (PGE2) release, and [3H]-thymidine and 35SO4 incorporation were quantified by using biochemical assays. Gene expression of inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), aggrecan, and collagen type II were assessed with real-time quantitative PCR (qPCR). Two-way ANOVA and the post hoc Bonferroni-corrected t tests were used to examine data. Results: CNP reduced NO and PGE2 release and partially restored [3H]-thymidine and 35SO4 incorporation in constructs cultured with IL-1?. The response was dependent on the concentration of CNP, such that 100 pM increased [3H]-thymidine incorporation (P &lt; 0.001). This is in contrast to 35SO4 incorporation, which was enhanced with 100 or 1000 nM CNP in the presence and absence of IL-1? (P &lt; 0.001). Stimulation by both dynamic compression and CNP and/or the PKGII inhibitor further reduced NO and PGE2 release and restored [3H]-thymidine and 35SO4 incorporation. In the presence and absence of IL-1?, the magnitude of stimulation for [3H]-thymidine and 35SO4 incorporation by dynamic compression was dependent on the concentration of CNP and the response was inhibited with the PKGII inhibitor. In addition, stimulation by CNP and/or dynamic compression reduced IL-1?-induced iNOS and COX-2 expression and restored aggrecan and collagen type II expression. The catabolic response was not further influenced with the PKGII inhibitor in IL-1?-treated constructs. Conclusions: Treatment with CNP and dynamic compression increased anabolic activities and blocked catabolic effects induced by IL-1?. The anabolic response was PKGII mediated and raises important questions about the molecular mechanisms of CNP with mechanical signals in cartilage. Therapeutic agents like CNP could be administered in conjunction with controlled exercise therapy to slow the OA disease progression and to repair damaged cartilage. The findings from this research provide the potential for developing novel agents to slow the pathophysiologic mechanisms and to treat OA in the young and old. <br/

    Improving scenario discovery by bagging random boxes

    AbstractScenario discovery is a model-based approach to scenario development under deep uncertainty. Scenario discovery relies on the use of statistical machine learning algorithms. The most frequently used algorithm is the Patient Rule Induction Method (PRIM). This algorithm identifies regions in an uncertain model input space that are highly predictive of model outcomes that are of interest. To identify these regions, PRIM uses a hill-climbing optimization procedure. This suggests that PRIM can suffer from the usual defects of hill climbing optimization algorithms, including local optima, plateaus, and ridges and valleys. In case of PRIM, these problems are even more pronounced when dealing with heterogeneously typed data. Drawing inspiration from machine learning research on random forests, we present an improved version of PRIM. This improved version is based on the idea of performing multiple PRIM analyses based on randomly selected features and combining these results using a bagging technique. The efficacy of the approach is demonstrated using three cases. Each of the cases has been published before and used PRIM. We compare the results found using PRIM with the results found using the improved version of PRIM. We find that the improved version is more robust to new data, can better cope with heterogeneously typed data, and is less prone to overfitting

    The awakening of Miss Prim is a novel that tells the story of Prudencia Prim in the small town of San Ireneo de Arnois. In the novel, Prudencia is surprised by the way of life of the irenites, who seem to despise the modern world. The novel serves as a framework for ideas that go far beyond the plot. It is a reflection on the value of truth, good and beauty and how they crystallized in Western civilization. This paper aims to shed some of the main ideas that give substance to the novel and gauge its value. In particular, the relationship between the present and the past, between tradition and progress and the option of accommodation or rebellion against current mainstream dynamics, will be valued. Finally, this paper will try to show how the whole novel revolves around the notion that there is a truth and that it is beautiful and what role that truth has in human life. Chesterton, G.K.     INDONESIA: Kajian MCMST merupakan pengembangan dari masalah optimasi Minimum Spanning Tree (MST) dengan memuat dua kriteria atau lebih. Salah satu algoritma yang mampu untuk menyelesaikan masalah MCMST adalah EDPA. EPDA memiliki tiga tahapan. Sebagai fondasi awal, pada tahap pertama dibangun dari Algoritma Kruskal atau Algoritma Prim dengan memperhatikan kriteria yang bersesuaian satu per satu. Kemudian pada tahap kedua dan ketiga dilakukan proses mutasi sampai akhirnya didapatkan spanning tree baru yang menjadi solusi efisien atau Pareto Front. Dengan perbedaan karakteristik yang dimiliki Algoritma Kruskal dan Algoritma Prim, penulis ingin menjelaskan perbandingan antara EPDA yang dibangun dari Algoritma Kruskal dan EPDA yang dibangun dari Algoritma Prim. Secara umum, baik EPDA dengan Algoritma Kruskal maupun EPDA dengan Algoritma Prim menghasilkan solusi yang sama. Adapun perbedaan yang dihasilkan terdapat pada indeks yang digunakan. Kemudian untuk memperkecil banyaknya kemungkinan solusi yang diberikan, maka pada saat pemilihan sisi baik untuk Algoritma Kruskal maupun Algoritma Prim tidak hanya memperhatikan kriteria yang dikerjakan, namun sekaligus memperhatikan pertimbangan bobot yang termuat dalam tabel Edge List. Oleh karena itu, dengan diperolehnya banyaknya kemungkinan solusi yang lebih sedikit, maka proses penyelesaian yang dilakukan menjadi lebih singkat. ENGLISH: The study of MCMST is the development of Minimum Spanning Tree (MST) optimization problem by containing two or more criteria. One of the algorithms that can solve the MCMST problems is EDPA. EPDA has three stages. As the initial foundation, the first stage is built from Kruskal’s Algorithm or Prim’s Algorithm by considering the corresponding criteria one by one. Then on the second and third stages the process of mutations are carried out until the new spanning trees that is the efficient solution or Pareto Front are obtained. By the difference between the characteristic of Kruskal's Algorithm and Prim's Algorithm, the author will explain a comparison between EPDA constructed by Kruskal's Algorithm and EPDA constructed by Prim's Algorithm. In general, either EPDA constructed by Kruskal’s Algorithm or EPDA constructed by Prim's Algorithm produces the same solution. As for the difference found is the use of indexes. Then, to minimize the amount of the given solution, the better sides choosen by Kruskal's Algorithm or Prim's Algorithm not only consider to the criteria, but also consider to the weights contained in the table of Edge List. Therefore, by getting the amount of the given solution that is less, then the process of solving can be shorter

    Improving scenario discovery for handling heterogeneous uncertainties and multinomial classified outcomes

    AbstractScenario discovery is a novel model-based approach to scenario development in the presence of deep uncertainty. Scenario discovery frequently relies on the Patient Rule Induction Method (PRIM). PRIM identifies regions in the model input space that are highly predictive of producing model outcomes that are of interest. To identify these, PRIM uses a lenient hill climbing optimization procedure. PRIM struggles when confronted with cases where the uncertain factors are a mix of data types, and can be used only for binary classifications. We compare two more lenient objective functions which both address the first problem, and an alternative objective function using Gini impurity which addresses the second problem. We assess the efficacy of the modification using previously published cases. Both modifications are effective. The more lenient objective functions produce better descriptions of the data, while the Gini impurity objective function allows PRIM to be used when handling multinomial classified data

    The paper presents professional and life path of Primarius Doctor of Medicine Ivo Glavan, the founder of the Department of Neuropsychiatry in Osijek. The paper is based on existing references about life and work of this well-known physician from Osijek, as well as review of his professional and scientific papers. The author tended to highlight Primarius Dr. Ivo Glavan as a person, because his personality was very important for establishment, as well as development of the Department of Neuropsychiatry in Osijek. The paper presents information on his heritage, childhood and youth referring to the period in which he had been both psychologically and biologically formed as a person with broad insight, persistence and structure in achieving his goals, ability of having deeper perspective, who was ahead of time. Despite extremely adverse circumstances and with significant personal sacrifice, Primarius Dr. Ivo Glavan was not only the founder of the Department of Neuropsychiatry in Osijek, but neuropsychiatrist of broad culture who contributed to development of profession and medical science thorough his professional and scientific papers in our region. His best known work was a textbook in neurology written in Croatian language called "Živčane bolesti ("Nervous Diseases"). He altruistically propagated professional and scientific thought on local level participating in work of Croatian Medical Association in Osijek.U članku je prikazan životni i stručni put prim. dr. Ive Glavana, osnivača neuropsihijatrijskoga odjela u Osijeku. Članak je napisan na temelju postojećih zapisa u literaturi o životu i radu ovoga poznatoga osječkog liječnika te na temelju pregleda njegovih stručnih i znanstvenih radova. Autor je nastojao posebno osvijetliti prim. dr. Ivu Glavana kao osobu, jer je njegova osobnost bila od presudnoga značenja za osnutak i razvoj neuropsihijatrijskoga odjela u Osijeku. Stoga je u članak uvrstio podatke o njegovu podrijetlu, djetinjstvu i mladosti tj. podatke o razdoblju u kojem se biološki i psihološki razvio u osobu koja je svojom širinom pogleda, dubinom pronicanja u bit stvari, sustavnošću i ustrajnošću u postizanju postavljenoga si cilja, nadilazila prostor i vrijeme u kojem je djelovala. Unatoč krajnje nepovoljnim okolnostima, uz velika osobna odricanja, prim. dr. Ivo Glavan nije samo osnovao Neuropsihijatrijski odjel u Osijeku, nego je kao neuropsihijatar i čovjek široke kulture, pridonosio razvoju struke i medicinske znanosti kod nas i u svijetu pisanjem stručnih i znanstvenih članaka, a njegovo najpoznatije djelo jest prvi udžbenik iz neurologije napisan na hrvatskom jeziku pod naslovom "Živčane bolesti".     Do good health and material circumstances protect older people from the increased risk of death after bereavement?

    This is an open access Article. Copyright @ 2012 The AuthorsAn increased risk of death in persons who have suffered spousal bereavement has been described in many populations. The impact of modifying factors, such as chronic disease and material circumstances, is less well understood. The authors followed 171,120 couples 60 years of age or older in a United Kingdom primary care database between 2005 and 2010 for an average of 4 years. A total of 26,646 (15.5%) couples experienced bereavement, with mean follow up after bereavement of 2 years. In a model adjusted for age, sex, comorbid conditions at baseline, material deprivation based on area of residence, season, and smoking status, the hazard ratio for mortality in the first year after bereavement was 1.25 (95% confidence interval: 1.18, 1.33). Further adjustment for changes in comorbid conditions throughout follow up did not alter the hazard ratio for bereavement (hazard ratio = 1.27, 95% confidence interval: 1.19, 1.35). The association was strongest in individuals with no significant chronic comorbid conditions throughout follow up (hazard ratio = 1.50, 95% confidence interval: 1.28, 1.77) and in more affluent couples (P = 0.035). In the first year after bereavement, the association between bereavement and death is not primarily mediated through worsening or new onset of chronic disease. Good health and material circumstances do not protect individuals from increased mortality rates after bereavement.This study was funded by a grant from the Dunhill Medical Trust

    S’appuyant sur les récents travaux d’I. Malkin, l’auteur soutient qu'au moment de sa fondation, le sanctuaire de Diane sur l’Aventin assuma aussi bien la fonction d’asile que celle de sanctuaire fédéral rival d’Aricie. Dès l'origine, ces deux fonctions de protection et de médiation impliquaient deux niveaux d’identité concomitants de la Diana Aventinensis : un horizon grec / maritime et un horizon latin / terrestre. À la lumière de ces éléments, l’auteur revient également sur la question de la localisation du temple de Diane. Les récentes données obtenues par la recherche archéologique et l’étude des fragments 21 et 22 de la FUM sont confrontées aux informations topographiques tirées d’une nouvelle analyse des sources littéraires. Ces éléments, de même que les données sur les fonctions originelles du temple dégagées au fil de cette enquête, conduisent l'auteur à privilégier une localisation du sanctuaire de Diane dans la partie septentrionale de l'Aventin.Relying on the recent works of I. Malkin, the author supports the view that at the time of its foundation, the Aventine sanctuary of Diana fulfilled the function of an asylum as well as that of a federal sanctuary, conceived as a rival to that at Aricia. From the beginning, these two functions of protection and mediation, involved two concomitant levels of its identity: Greek-maritime / Latin-terrestrial. In the light of these elements, the author reconsiders the question of the location of the sanctuary. The recent data generated by archaeological research and the study of fragments 21 and 22 of the FUM are compared with topographical information provided by a new analysis of literary sources. The objective is to determine the best hypothesis considering the functional and topographical evidences obtained from that survey
