Includes bibliographical references.This work describes the development of an integrated sensors system to measure concentrations of dissolved oxygen (DO), pH, glucose, and lactate concurrently at single cell level. DO was measured amperometrically using a three-electrode system of working (WE), counter (CE) and reference (RE) electrodes. pH was measured potentiometrically using two electrodes system of Indium Tin Oxide (ITO) WE and Ag/AgCl RE. Glucose and lactate were measured enzymatically by measuring the current generated from the oxidation of hydrogen peroxide generated from the catalysis of glucose or lactate at the WEs with their catalysis enzymes. A microfluidic chamber containing all four sensors was made using SU8 to investigate single oocytes/embryos immersed in up to 120 µL of respiration buffer. This work includes the results of using the integrated sensors system to measure the metabolic activities of real cells including single oocytes or embryos. The micro-chamber was completely sealed using top layer of ovoil and covered by top glass lid to avoid oxygen exchange between the inside of the chamber and the atmosphere, while being maintained at a temperature of 38.5 ºC to preserve cell viability. The oxygen consumption of cells, the lactate production and glucose consumptions were measured as a change in output current and converted to femto-mol (fmol) per second based on calibrations with buffer of known DO, lactate, and lactate concentrations. This integrated sensor system has some potential applications include evaluating effects of metabolic therapies on oocyte bioenergetics, study the effect of aging on embryos development and monitoring mitochondrial function throughout oocyte maturation and blastocyst development to predict embryo viability to compliment assisted reproductive technologies.Undergraduate Choice - Research Top Scholar

Preparation of anastrozole loaded PEG-PLA nanoparticles: evaluation of apoptotic response of breast cancer cell lines

Despite the wide use of ANS, it is associated with serious side effects due to uncontrolled delivery. In addition, ANS exhibits low solubility and short plasma half-life. Nanotechnology-based drug delivery has the potential to enhance the efficacy of drugs and overcome undesirable side effects. In this study, we aimed to prepare novel ANS-loaded PLA-PEG-PLA nanoparticles (ANS-NPs) and to compare the apoptotic response of MCF-7 cell line to both ANS and ANS-loaded NPs.Method: ANS-NPs were synthesized using double emulsion method and characterized using different methods. The apoptotic response was evaluated by assessing cell viability, morphology, and studying changes in the expression of MAPK3, MCL1, and c-MYC apoptotic genes in MCF-7 cell lines.Results: ANS was successfully encapsulated within PLA-PEG-PLA, forming monodisperse therapeutic NPs with an encapsulation efficiency of 67%, particle size of 186±27.13, and a polydispersity index of 0.26±0.11 with a sustained release profile extended over 144 hours. In addition, results for cell viability and for gene expression represent a similar apoptotic response between the free ANS and ANS-NPs.Conclusion: The synthesized ANS-NPs showed a similar therapeutic effect as the free ANS, which provides a rationale to pursue pre-clinical evaluation of ANS-NPs on animal models. SURGICAL SITE INFECTIONS: PREVALENCE, ASSOCIATED FACTORS AND ANTIMICROBIAL SUSCEPTIBILITY PATTERNS OF THE BACTERIAL ISOLATES AMONG POSTOPERATIVE PATIENTS IN SANA’A, YEMEN
Background: Surgical site infections (SSIs) are infections that occur one month after a surgical operation or one year after implant surgery and a surgical procedure, either at the injury site or near the injury site. Despite SSIs are still a major problem causing increased morbidity and mortality globally, especially in developing countries, there is a dearth of information in Yemen.
Objective: This study was aimed to assess the prevalence of SSIs, associated factors and antimicrobial susceptibility patterns of the bacterial isolates among post-operative patients in public and private hospitals in Sana’a, Yemen.
Methods: This is a cross-sectional study carried out on 309 postoperative patients, aged one year and over that underwent surgery in selected public and private hospitals between January 2019 and January 2020 in Sana’a. Patients’ demographic and clinical information were assessed using an interviewer-administered, pretested, structured questionnaire. Wound swabs and aspirates were collected, placed in Stuart transport media and transferred to the Bacteriology Department of the National Center for Public Health Laboratories in Sana’a for bacterial isolation, identification and antimicrobial susceptibility testing..
Results: The mean age of postoperative patients was 34.0±18.3 (SD) years, age group 25-34 years was 93 (30.1%), and females were 105 (33.8%). Total 309 postoperative patients, 98 (31.7%) presented with SSI. Having postoperative antibiotics (OR=7.0, 95%CI: 4.1-12.0), dirty surgical wound (OR=10.5, 95%CI: 5.0-21.9), emergency surgery (OR=3.1, 95%CI: 1.8-5.1), amputation (OR=2.5, 95%CI: 1.1-5.7), excision (OR=2.2, 95%CI: 1.2-4.0), hospital stay > two weeks (OR=5.2, 95%CI: 2.3-11.1) and having diabetes mellitus (OR=2.2, 95%CI: 1.1-4.7) were factors significantly associated with SSIs.
Conclusion: This study shows a significant proportion of SSIs among patients in Sana’a, Yemen. Being a female, having a dirty surgical wound, amputation, excision, long hospital stays, diabetes mellitus, and emergency surgery appears to be the major factors associated with SSIs. The most common SSI bacterial etiology is S. aureus, sensitive to vancomycin and rifampicin.

“Trust your gut”: exploring the connection between gut microbiome dysbiosis and the advancement of Metabolic Associated Steatosis Liver Disease (MASLD)/Metabolic Associated Steatohepatitis (MASH): a systematic review of animal and human studies
Metabolic Associated Steatosis Liver Disease (MASLD) and its advanced form, Metabolic Associated Steatohepatitis (MASH), represent growing global health concerns closely linked to obesity, type 2 diabetes mellitus (T2DM), and metabolic syndrome. The gut microbiome has emerged as a key modulator in MASLD pathogenesis through the gut–liver axis, influencing hepatic fat accumulation, inflammation, and fibrosis via microbial metabolites and immune responses. Dysbiosis–characterized by altered microbial diversity and composition–contributes to hepatic lipid dysregulation, systemic inflammation, and impaired bile acid signaling. Metabolites such as short-chain fatty acids (SCFAs), trimethylamine-N-oxide (TMAO), and ethanol play critical roles in disease progression. Recent innovations in precision medicine, including microbiome profiling, metabolomics, and genomics, offer promising diagnostic and therapeutic strategies. Targeted probiotics, fecal microbiota transplantation (FMT), and personalized dietary interventions are under investigation for modulating the gut microbiome. This systematic review, conducted in accordance with PRISMA 2020 guidelines, is the first to comprehensively integrate both animal and human studies on MASLD/MASH-related gut microbiome alterations. It uniquely synthesizes microbial taxa, functional metabolites, and region-specific patterns–including data from underrepresented MENA populations. Eligible studies from PubMed, Scopus, and Web of Science evaluated microbial composition, metabolite profiles, and associations with steatosis, inflammation, and fibrosis. The findings underscore the diagnostic and therapeutic potential of microbiome modulation and emphasize the need for longitudinal, mechanistically driven studies. This systematic review is the first to integrate both animal and human studies on MASLD/MASH-related gut microbiome alterations. Unlike previous reviews, it uniquely emphasizes microbial taxa, functional metabolites, and region-specific patterns, including underrepresented MENA populations. In recent years, several genes have been implicated in the variable disease presentation of global developmental delay (GDD) and intellectual disability (ID). The endoplasmic reticulum membrane protein complex (EMC) family is known to be involved in GDD and ID. Homozygous variants of EMC1 are associated with GDD, scoliosis, and cerebellar atrophy, indicating the relevance of this pathway for neurogenetic disorders. EMC10 is a bone marrow-derived angiogenic growth factor that plays an important role in infarct vascularization and promoting tissue repair. However, this gene has not been previously associated with human disease. Herein, we describe a Saudi family with two individuals segregating a recessive neurodevelopmental disorder. Both of the affected individuals showed mild ID, speech delay, and GDD. Whole-exome sequencing (WES) and Sanger sequencing were performed to identify candidate genes. Further, to elucidate the functional effects of the variant, quantitative real-time PCR (RT-qPCR)-based expression analysis was performed. WES revealed a homozygous splice acceptor site variant (c.679-1G > A) in EMC10 (chromosome 19q13.33) that segregated perfectly within the family. RT-qPCR showed a substantial decrease in the relative EMC10 gene expression in the patients, indicating the pathogenicity of the identified variant. For the first time in the literature, the EMC10 gene variant was associated with mild ID, speech delay, and GDD. Thus, this gene plays a key role in developmental milestones, with the potential to cause neurodevelopmental disorders in humans. This article is protected by copyright. Alterations in Antioxidant Defense System in the Plasma of Female Khat Chewers of Thamar City, Yemen

Unfortunately, farmers use to apply pesticide for the better product of Khat. The present study has been designed to investigate the activity of plasma butyrylcholinesterase (BChE) and to assess the antioxidant defense system in the plasma of female Khat chewers in Thamar city, Yemen. Plasma of twenty female Khat chewers and twenty controls (non Khat chewers) were prepared and the activities of BChE and catalase (CAT) were estimated along with the measuring levels of reduced glutathione, total thiols and cholesterol. At biochemical level a significant decrease in the activities of BChE and CAT were observed in the plasma of female Khat chewers (P < 0.05) concomitant with reductions in the levels of reduced glutathione and total thiols (P < 0.05) as compared to non Khat chewers. This alterations on the antioxidants resulted in decrease of plasma cholesterol in Khat chewers group (P < 0.05). The present data show that the production of oxidants which are responsible for reduction in antioxidant defense system might be due to chewing Khat plant with more attention to the pesticide applied to the plant
The comparison between four PLCC packages and eight PLCC packages in personal computer (PC) using computational fluid dynamic (CFD), fluent software™ using epoxy moulding compound material (EMC)
Link to publisher's homepage at http://www.ttp.net/The paper present the three dimensional numerical analysis of heat and fluid flow through Plastic Leaded Chip Carrier (PLCC) packages in inline orientation horizontally mounted on a printed circuit board in a wind tunnel is carried out using a commercial CFD code, FLUENT™ by using Epoxy Moulding Compound (EMC) as a main material. The study was made for four and eight packages with different Reynolds Number and package chip powers. The results are presented in term of junction temperature for four and eight PLCC package under different conditions. It is observed the chip temperatures of eight PLCC packages have higher junction temperature compare to four PLCC packages due to effect of other PLCC because of space and gap between PLCC that have more number of PLCC is smaller. Hence it makes junction temperature of eight PLCC higher compare to four PLCC packages. Moreover, the junction temperature of the packages decreases with increase in Reynolds Number
