A corpus linguistic analysis of catalogue data: understanding curatorial practice over time
Legacies of Catalogue Descriptions and Curatorial Voice: Infographics
The project 'Legacies of Catalogue Descriptions and Curatorial Voice: Opportunities for Digital Scholarship' (2020-22, AHRC, Project Reference AH/T013036/1) sought to develop a platform for a transformational impact in digital scholarship within cultural institutions by opening up new and important directions for computational, critical, and curatorial analysis of collection catalogues. Our pilot research investigated the temporal and spatial legacy of a landmark catalogue: the ‘Catalogue of Political and Personal Satires Preserved in the Department of Prints and Drawings in the British Museum’, entries in which form the basis of related catalogue data at institutions including the Lewis Walpole Library and the British Library. Towards the end of the project we invited together members of our community to discuss shared agendas and actions: where we should focus our collective resources, what things we need to work differently, and the role of computational technologies in both shaping and constraining change. These infographics respond to those conversations. 'Legacy catalogue entries: actions and agendas' presents priorities that emerged from a longlist of project findings. And 'Tracing the transmission of legacy catalogue entries' visualises the journey of legacy catalogue data in our case study from conception, through cataloging infrastructures, and into the future. Both graphics were designed by Lucy Havens, with editorial support from James Baker and Rossitza Atanassova. The process is described at Lucy Havens, 'Designing Infographics for the “Legacies of Catalogue Descriptions” Project' (March 2022). Physiological functions of ASR proteins regarding sugar signaling, transport and metabolism in two cell culture models in grapevine
Les sucres, sont des signaux métaboliques, impliqués dans le développement des plantes et leurs réponses aux contraintes du milieu. Les transporteurs de sucres se révèlent à la fois acteurs de la répartition des sucres et cibles de leur signalisation. L'ASR (ABA, Stress and Ripening) de la Vigne, VvMSA, étant identifiée comme protéine régulatrice de l'expression génique du transporteur d’hexoses VvHT1, l'objectif de la thèse est d'appréhender ses rôles physiologiques dans une démarche de biologie intégrée.Le premier axe a été dédié à la mise en place des modèles biologiques, des cellules embryogènes et non embryogènes de Vigne, issues du même fond génétique mais cultivées sur deux sources de carbone différentes. La caractérisation des cinétiques de prolifération et l'analyse des métabolomes ont mis en évidence leur sensibilité/tolérance différentielle à la carence en sucres. Le deuxième axe a porté sur la régulation de VvHT1 dans les deux types cellulaires sauvages et leurs mutants de surexpression/répression de VvMSA. L'approche pharmacologique utilisant des analogues du glucose, l'analyse de l'expression génique, le transport du glucose et l'activité des enzymes de la glycolyse indiquent que VvMSA affecte l'expression de VvHT1 par la voie dépendante du métabolisme du glucose. Le troisième volet a été réalisé par une approche de protéomique quantitative et comparative des protéines nucléaires des cellules embryogènes sauvages et réprimées pour VvMSA. Les protéines à expression significativement affectée par l'absence de l'ASR, laissent entrevoir un nouveau rôle à l'interconnexion des réponses métaboliques aux stress et la régulation épigénétique de l'expression génique.Sugars are metabolic signals involved in plant development and responses to environmental cues. Sugar transporters are both actors of sugar partitioning and targets of sugar signaling. As Grape ASR (ABA, Stress, Ripening), VvMSA, is identified as a regulatory protein controlling gene expression of the hexose transporter VvHT1, the aim of the PhD thesis is to assess its physiological functions by an integrative biology approach. The first part of the study consisted in the establishment of biological models, embryogenic and non embryogenic grape cells, sharing the same genetic background but growing on distinct carbon sources. The characterization of the proliferation kinetics and metabolomes of both cell types revealed differences in their sensitivity/tolerance to sugar starvation.The second objective was focused on VvHT1 expression regulation in both cell types and their mutants overexpressing or silenced for VvMSA. The pharmacological approach using glucose analogues, coupled to the analysis of gene expression, glucose transport and glycolytic enzymes activity, suggest that VvMSA affects VvHT1 expression through a glucose metabolism dependent pathway.The third research axis was carried out through a quantitative and comparative proteomic analysis of nuclear proteins in embryogenic wild type and VvMSA silenced cells. Proteins whose expression is affected by ASR repression suggest a new functional role of VvMSA at the interplay between metabolic responses to stress and epigenetic regulation of gene expression
Doctrine, polemic and literary tradition in some hexameter poems of Prudentius
The thesis, the topic of which is restricted to the polemical didactic poems, Apotheosis, Hamartigenia and Contra Symmachum 1-2, aims to establish the attitudes of Prudentius to the literary tradition and argues for his relationship with the Latin classical poets. Its main argument is that the hexameter poems as a group can be profitably studied from a stylistic angle, since they show how Prudentius combined, and used with innovation, the styles of several poets, namely Lucretius, Virgil and Juvenal, and in many cases engaged with the literary tradition as a whole. Chapter I surveys, as reflected in the poems, Prudentius' awareness of the political, religious and literary milieu in the Christian Empire of the West in his day. Chapter II examines how Prudentius employed the style of argument and imagery in the D.R.N. to present Christian doctrines on the body and the soul, and to reject pagan superstition. Chapter III shows how with much imagination and respect Prudentius adapted Virgil's phraseology and techniques to give new Christian interpretations of some mythical and historical themes in the Aen., such as the 'Golden Age' and the battle of Actium, and of topics on agriculture from the Georg. Chapter IV argues that, like other fourth century Christian writers, the poet entered into the spirit of Satire and alluded to Juvenal's themes and language in his treatment of the topics of sin and sexuality. Finally, in Chapter V Prudentius' adaptations of the biblical accounts in Gen. 19 and of Ps. 136 are used to demonstrate how allegory, which is a main feature of his poetry, was combined successfully with different classical techniques. In conclusion, the hexameter poems demonstrate that Prudentius did not reject classical poetry on the basis of its content, but used both its themes and poetic techniques in order to merge the ancient with the Christian literary tradition

The monosaccharides ERD6-like (Early Response to Dehydration six-like) transporters family : emergence, evolution in terrestrial plants and responses to drought in Arabidopsis thaliana leaves
La caractérisation des réponses des plantes à la sécheresse est un enjeu actuel majeur. La gestion du carbone dans l’adaptation des végétaux aux contraintes environnementales requiert l’activité de transporteurs de monosaccharides ERD6-like (Early Response to Dehydration six-like), transporteurs impliqués dans la compartimentation subcellulaire des sucres et leur remobilisation à l’échelle de l’organisme. Le premier axe de la thèse a été dédié à l’analyse phylogénétique des ERD6-like par une approche de génomique évolutive. L’identification de 519 ERD6-like dans les protéomes de 47 Embryophytes a permis de reconstituer l’histoire évolutive de cette famille en mettant en exergue des événements majeurs de duplications. Le second axe a permis d’établir la cartographie de l’expression des ERD6-like dans les différents organes d’Arabidopsis thaliana et a mené à l’identification de gènes exprimés de façon constitutive dans tous les organes, et d’autres, manifestant une expression préférentielle. Le troisième axe a permis d’étudier la réponse des ERD6-like à la carence en eau, dans les feuilles d’A. thaliana. Pour cela, le phénotypage morphologique, biochimique et physiologique des rosettes de plantes cultivées en conditions normales d’alimentation hydrique ou de stress hydrique a été réalisé. Le profil d’expression des AtERD6-like au cours de la cinétique de carence en eau a mis en évidence l’induction de quatre AtERD6-like en réponse au déficit hydrique. L’analyse de simples mutants pour ces gènes, a permis d’initier l’étude de la diversification fonctionnelle des ERD6-like d’A. thaliana.Nowadays, characterizing plants responses to drought is a major goal for plant scientists. The carbon management in response to environmental cues involves the activity of ERD6-like (Early Response to Dehydration six-like) monosaccharides transporters, playing roles in sugar subcellular partitioning and re-allocation at the scale of the whole organism. The first purpose of this PhD thesis focused on the analysis of the ERD6-like evolution into the plant Kingdom, using a phylogeny approach. Identification of 519 ERD6-like in the proteome of 47 plants species lead to rebuild the evolutionary story of this gene family, and to provide evidence for the major duplication events. The second purpose was the mapping of ERD6-like expression levels in different organs of Arabidopsis thaliana and lead to the identification of almost constitutively expressed copies and others displaying organ-preferential expression patterns. The third purpose was to study the responses of ERD6-like genes to water starvation in A. thaliana leaves. This highlighted the expression induction of 4 AtERD6-like during water deficit. Finally, the characterization of erd6-like insertional mutants initiated the study of the functional diversity of this large family of monosaccharide transporters in Arabidopsis thaliana
Diversité fonctionnelle des transporteurs de sucres AtESL dans le développement et la réponse d'Arabidopsis thaliana aux contraintes abiotiques
Le déficit hydrique est l'un des facteurs abiotiques les plus importants, qui affecte gravement la croissance et le développement des plantes ainsi que le rendement des cultures. Chez Arabidopsis thaliana, les transporteurs de monosaccharides ESL (Early response to dehydration Six-Like) forment l'une des plus grandes sous-familles de transporteurs de sucre, cependant leurs fonctions biologiques ne sont pas encore élucidées. Notre analyse phylogénétique a révélé l'émergence de cette famille de gènes chez les Streptophytes, leur répartition en trois principaux groupes (ESL1, ESL2 et ESL3) ainsi que leur diversification qui est étroitement liée à l'évolution des plantes terrestres. Dans le but d'élucider la diversité fonctionnelle des AtESL dans le développement des plantes et leurs réponses aux stress abiotiques, le phénotypage de plantes sauvages Col-0 et mutantes esl a été effectué aux niveaux morphologique, physiologique, biochimique et moléculaire, dans des conditions d'arrosage et de carence en eau. Le profil d'expression des gènes ESL, en réponse au déficit en eau, a révélé l’induction de quatre d’entre eux et la répression de huit autres gènes. Afin de comprendre les rôles des transporteurs ESL induits par la carence en eau, quatre simples mutants d'insertion T-DNA ont été identifiés et caractérisés. L’absence de phénotypes marquants pour ces simples mutants, nous a conduit à créer par croisements génétiques dans différentes combinaisons, trois doubles et deux triples mutants esl. Grace à des tests de germination de graines des simples, doubles et triples mutants esl, nous avons fourni des preuves de leur sensibilité/résistance différentielle à la germination à l'ABA, au glucose, à la salinité et au froid. Pour mieux comprendre l'activité de transport de sucres de AtESL3.5 et de AtESL3.3, nous avons réalisé une expression hétérologue dans S. cerevisiae, ainsi que des essais de transport de sucres sur des vacuoles isolées de la plante. Finalement, en utilisant des constructions de fusion de protéines avec deux marqueurs fluorescents différents exprimées de façon transitoire dans des protoplastes isolés de feuilles de tabac agro-infiltrées, nous avons démontré que AtESL3.5 est localisé dans le tonoplaste.Water deficit is among the most important abiotic factors, which severely affects plant growth, development, and crop yield. In Arabidopsis thaliana, the monosaccharide transporters ESL (Early response to dehydration Six-Like) represent one of the largest sugar transporter subfamilies, but until now their biological functions remain not elucidated. Our phylogenetic analysis revealed the emergence of the ESL gene family in Streptophyta, while their diversification and distribution in three groups (ESL1, ESL2, and ESL3) are closely related to the evolution of land plants. In the aim to unravel the functional diversity of AtESLs in plant development and stress responses, phenotyping of wild-genotype Col0 and esl mutant plants was performed at morphological, physiological, biochemical, and molecular levels, under conditions of watering and water deprivation. The expression profiling of ESL genes revealed up-regulation of four genes and down-regulation of eight genes in response to water deficit. In order to decipher the roles of ESL transporters, four T-DNA insertional mutants corresponding to each of the four drought-responsive genes were identified. Given the lack of specific phenotypes for single mutants, we created through their genetic crossing in different combinations three double and two triple esl mutants. Using seed germination assays for esl single, double, and triple mutants we provided evidence for their differential germination sensitivity/resistance to ABA, glucose, salinity and cold treatments. To gain further insight into the sugar transport activity of AtESL3.5 and AtESL3.3, heterologous expression in S. cerevisiae, as well as sugar transport assays on plant isolated vacuoles were developed. Eventually, though using protein fusion constructs with two different fluorescent markers, and their transient expression in protoplasts isolated from agro-infiltrated tobacco leaves we demonstrated that AtESL3.5 is localized in the tonoplast
