    Cadmium accumulation and physiological response of sunflower plants to Cd during the vegetative growing cycle

    The effects of soil Cd contamination on Cd accumulation and distribution, growth and physiological responses of sunflower plants were investigated. Plants were subject to six levels of soil contamination (from 2.5 to 15 mg Cd kg/soil) with an untreated control, from the emergence of the cotyledon leaves until the harvest, when plants were at the flower bud stage. An overall increase of Cd concentration was found in all tissues of the plants (roots, stem, young, mature and old leaves) by increasing the Cd contamination in the soil. Regardless of treatments, Cd concentration in roots always exceeded those in the aboveground dry matter with a low translocation from roots to shoots. At early stage of growth, Cd concentration in plants was higher than at the flower bud stage. Soil Cd contamination did not affect plant growth, relative water content and gas exchange parameters. Negative and significant correlation was only found between Cd concentration in the young leaves and chlorophyll concentration at the end of vegetative growing stage. Roots and old leaves are the main metal sinks suggesting a defense or tolerance mechanism of the plants to avoid toxic levels in physiologically most active apical tissues. These results should be tested in open field to verify the suitability of sunflower in the area of phytotechnologies

    Variation of glucosinolates concentration and root growth of horseradish as affected by nitrogen and sulphur supply

    The aim of the study was to evaluate total and individual glucosinolates concentration, root weight and diameters of horseradish accessions (Cor and Mon) grown by adding nitrogen (N) and sulphur (S) and harvested at the beginning (late autumn) and towards the end (early spring) of the harvest period. The concentration of the 9 glucosinolates (GLS) quantified in roots varied greatly between accessions (6-fold higher in Cor) and with fertilization (N alone led to an increase of 64% of GLS with an additional enhancement of 65% when S was added to N). Root GLS concentration was significantly higher in early spring compared to the previous harvest in late autumn (12.5 vs 8.9 μmol/g DW (dry weight)); a drastic decline of GLS (2.7 μmol/g DW) occurred in root harvested in the following autumn from plants left ad hoc in the field. The aliphatic sinigrin was the most abundant (80% of total GLS), followed by the aromatic gluconasturtiin and the indolic glucobrassicin (5.8% and 4.7%, respectively). Root diameter and weight increased in plants treated with fertilizers (19% and 61%, respectively) and throughout the harvests with Cor showing values double with respect to Mon (925 vs 476 g/plant). Relationships between GLS concentration and root weight (R2 = 0.61) and diameter (R2 = 0.69) were improved by excluding the roots harvested from plants left ad hoc in the field that showed the worst performances

    Lysimetric determination of muskmelon crop coefficients cultiveted under plastic mulches

    A trial was carried out at the lysimeter station in southern Italy on muskmelon crop cultivated with and without plastic mulch during spring–summer in 2001 and 2003. The objective of the experiment was to verify the reliability of the crop evapotranspiration (ETc) estimate by means of the most recent update of the FAO Irrigation and Drainage Paper 56, in comparison with ETc directly measured by two mechanical weighing lysimeters. Crop coefficients (Kc) were determined during different development stages based on lysimetric measures of ETc and of the reference evapotranspiration (ET0) estimated through the Penman Monteith and the Hargreaves methods. On melon crop cultivated without plastic mulch, corrected crop coefficients (Kc) following the last FAO Irrigation and Drainage Paper 56 procedures were well correlated with those measured from lysimeter and were as reliable as the ETc estimate. In contrast, values of Kc proposed by FAO Irrigation and Drainage Paper 56 for crops grown with plastic mulch were meaningfully lowers than those measured from lysimeter, loading to an underestimation of water consumption. On muskmelon, cultivated with and without plastic mulch, it is necessary to adapt development phase duration, suggested by the FAO Irrigation and Drainage Paper 56, to the real phenology of the employed cultivar. An adaptation of the phenology to the real duration of the single phases is essential to avoid error in the estimate of ETc

    Culturable bacteria from Zn- and Cd-accumulating Salix caprea with differential effects on plant growth and heavy metal availability.

    Aims: To characterize bacteria associated with Zn ⁄ Cd-accumulating Salix caprea regarding their potential to support heavy metal phytoextraction. Methods and Results: Three different media allowed the isolation of 44 rhizosphere strains and 44 endophytes, resistant to Zn ⁄Cd and mostly affiliated with Proteobacteria, Actinobacteria and Bacteroidetes ⁄ Chlorobi. 1-Aminocyclopropane- 1-carboxylic acid deaminase (ACCD), indole acetic acid and siderophore production were detected in 41, 23 and 50% of the rhizosphere isolates and in 9, 55 and 2% of the endophytes, respectively. Fifteen rhizosphere bacteria and five endophytes were further tested for the production of metal-mobilizing metabolites by extracting contaminated soil with filtrates from liquid cultures. Four Actinobacteria mobilized Zn and ⁄ or Cd. The other strains immobilized Cd or both metals. An ACCD- and siderophore-producing, Zn ⁄ Cd-immobilizing rhizosphere isolate (Burkholderia sp.) and a Zn ⁄ Cd-mobilizing Actinobacterium endophyte were inoculated onto S. caprea. The rhizosphere isolate reduced metal uptake in roots, whereas the endophyte enhanced metal accumulation in leaves. Plant growth was not promoted. Conclusions: Metal mobilization experiments predicted bacterial effects on S. caprea more reliably than standard tests for plant growth-promoting activities. Significance and Impact of the Study: Bacteria, particularly Actinobacteria, associated with heavy metal-accumulating Salix have the potential to increase metal uptake, which can be predicted by mobilization experiments and may be applicable in phytoremediation

    It's a long way to the top: Plant species diversity in the transition from managed to old-growth forests

    Questions: Do vascular plant species richness and beta-diversity differ between managed and structurally complex unmanaged stands? To what extent do species richness and beta-diversity relate to forest structural attributes and heterogeneity?. Location: Five national parks in central and southern Italy. Methods: We sampled vascular plant species composition and forest structural attributes in eight unmanaged temperate mesic forest stands dominated or co-dominated by beech, and in eight comparison stands managed as high forests with similar environmental features. We compared plant species richness, composition and beta-diversity across pairs of stands (unmanaged vs managed) using GLMMs. Beta-diversity was quantified both at the scale of each pair of stands using plot-to-plot dissimilarity matrices (species turnover), and across the whole data set, considering the distance in the multivariate species space of individual plots from their centroid within the same stand (compositional heterogeneity). We modelled the relationship between species diversity (richness and beta-diversity) and forest structural heterogeneity and individual structural variables using GLMMs and multiple regression on distance matrices. Results: Species composition differed significantly between managed and unmanaged stands, but not richness and beta-diversity. We found weak evidence that plant species richness increased with increasing levels of structural heterogeneity and canopy diversification. At the scale of individual stands, species turnover was explained by different variables in distinct stands, with variables related to deadwood quantity and quality being selected most often. We did not find support for the hypothesis that compositional heterogeneity varies as a function of forest structural characteristics at the scale of the whole data set. Conclusions: Structurally complex unmanaged stands have a distinct herb layer species composition from that of mature stands in similar environmental conditions. Nevertheless, we did not find significantly higher levels of vascular plant species richness and beta-diversity in unmanaged stands. Beta-diversity was related to patterns of deadwood accumulation, while for species richness the evidence that it increases with increasing levels of canopy diversification was weak. These results suggest that emulating natural disturbance, and favouring deadwood accumulation and canopy diversification may benefit some, but not all, facets of plant species diversity in Apennine beech forests

    Water use assessment in muskmelon by the Penman-Monteith "one-step" approach

    The use of the "one-step" approach of Penman-Monteith (P-M) to assess crop water use has its limit in the lack of user friendly methodologies for a daily assessment of crop canopy resistance, rc. The model proposed by Monteith [Monteith, J.L., 1965. Evaporation and environment. In: Fogg, G.E. (Ed.), The State and Movement of Water in Living Organism. Soc. Exp. Biol. Symp. 19, 205-234] to estimate rc (rc =  100/LAI), although it is simple, requires the knowledge of LAI value during growing cycle. This work aims to propose an easy-to-use methodology for irrigation scheduling, to assess the values LAI gets during the growing cycle and thus for canopy resistance assessment. In muskmelon crop, grown with and without plastic film mulching, canopy resistance was measured by P-M formula inversion, clarifying the unknown resistivity term, being crop evapotranspiration known by lysimeter-based measurements. Measured canopy resistance was compared with the one estimated by Monteith model (1965), both in its original version and in the form we simplified in terms of LAI value during crop cycle. Thus, we compared the evapotranspiration assessed by the "one-step" use of P-M equation, with those estimated by the classical "two-step" approach, using crop coefficients, and with that directly measured by lysimeters. In particular in the mulched crop the "one-step" approach of P-M overestimates water use only by 4%, while the "two steps" approach underestimates water use by 17%. Despite both the methodologies proposed for LAI calculation and the Monteith model to assess canopy resistance extremely simplify more complex processes, they were able to give a good accuracy to assess muskmelon water use by the P-M "one-step" equation. Comparing the "one-step" and the more used "two-step" approaches it came out that, despite the latter is better correlated to the data measured by a lysimeter, it does not achieve a more accurate assessment compared to the "one-step" approach; in particular, in the mulched crop, the "two-step" approach significantly underestimates water use, while its estimation is reliable with the "one-step" approach.Evapotranspiration Canopy resistance Aerodynamic resistance Crop coefficient

P. bovis infects cows throughout the lactation cycle, including dry periods, and can persist in the udder and the environment for a long time. Since P. bovis does not respond to treatments with antibiotics, the suggested sanitary measure to restrict the spread is culling infected animals. A point-of-care test for early detection of the causative agent is critically needed to guide farm management and the appropriate treatment of mastitis. Loop-mediated isothermal amplification (LAMP) is a highly specific molecular method, time-saving, cost-effective and easy to perform in limited settings. This study aimed to develop a LAMP assay for P. bovis detection directly from milk samples; it was employed in conjunction with a commercial DNA extraction kit which was previously used to extract DNA from milk specimens containing microbes. The LAMP assay detected P. bovis DNA within 1 h in milk samples spiked with P. bovis at a concentration of 50 cells/μL, enabling on-farm disease monitoring and decision-making based on a reliable diagnosis. The LAMP method will contribute to the accurate and rapid identification of P. bovis in asymptomatic or recurrent mastitis cases and consequently aid the implementation of targeted control measures and the reduction of losses in milk production. © 2024, The Author(s), under exclusive licence to Springer Nature B.V.journal articl
