    Cryopreservation of yellowtail flounder (Pleuronectes ferrugineus) semen in large straws

    Semen from yellowtail flounder was cryopreserved in 0.25 ml and flat 1.7 ml straws. Semen frozen in 1.7 ml straws was collected from males that had been stimulated to increase semen output by treatment with a GnRH analogue. The semen extender was a modification of a diluent that had previously been used to cryopreserve salmonid semen with 10% propylene glycol added as a cryoprotectant. The semen was frozen in liquid nitrogen vapour and stored in liquid nitrogen. Freezing rates inside the straws were measured using a thermocouple. In a preliminary fertility trial, there were no significant differences in egg fertilization rates and hatching rates between freshly collected semen and semen frozen in 1.7 ml straws. A larger trial compared the fertility of freshly collected semen, semen frozen in 0.25 ml straws and semen frozen in 1.7 ml straws. The mean fertilization rates for these groups were 64.7, 59.2 and 54.4% respectively; the hatching rates were 36.7, 52.5 and 42.8% respectively.     Fresh storage of rainbow trout (Oncorhynchus mykiss) semen using a non-aqueous medium

    Several media for the storage of fresh semen from rainbow trout at 4 degrees C were evaluated. Semen was diluted with an equal volume of fluorocarbon, a 1:1 fluorocarbon:extender emulsion, a 2:1 fluorocarbon:extender emulsion, or with extender, or was stored undiluted. The semen was stored under moisture-saturated or non-saturated air. Efficacy of treatment was evaluated using motility rating, percentage of dead spermatozoa as determined by fluorometry, leakage of lactate dehydrogenase (LDH) activity into seminal plasma, and fertility trials during the storage period. Unlike motility ratings and fluorometry, measurement of LDH leakage to estimate sperm viability during storage with fluorocarbon did not give clear results due to the separation of the non-aqueous fluorocarbon from the aqueous seminal plasma phase. Semen diluted 1:1 with fluorocarbon maintained sperm viability at an acceptable level for 37 days.     Resonance Coupling in Si@WS2Core-Ω Shell Nanostructure

    Realizing strong laser–matter interaction in a heterostructure consisting of two-dimensional transition metal dichalcogenides (TMDCs) and an optical nanocavity is a potential strategy for novel photonic devices. In this paper, two core-Ω shell nanostructures, Si@WS2 core-Ω shell nanostructure on glass/Si substrates, are briefly introduced. A strong laser–matter interaction occurred in the Si@WS2 core-Ω shell nanostructure when it was excited by femtosecond (fs) laser in the near-infrared-1 region (NIR-1, 650 nm–950 nm), resulting in a resonance coupling between the electric dipole resonance (EDR) of the Si nanosphere (NS) and the exciton resonance of the WS2 nanomembrane (NMB). The generation of resonance coupling regulates the resonant mode of the nanostructure to realize the multi-dimensional nonlinear optical response, which can be utilized in the fields of biological imaging and nanoscale light source

    Evaluation of protein quality in fish meals by chemical and biological assays

    Fish meals (herring, menhaden, and anchovy), commercially available in Atlantic Canada and a Norwegian fish meal (Norse-LT94Reg.) formulated to contain 0, 16, 28 or 40% protein were evaluated for protein quality by in vitro assays and by growth studies with Atlantic salmon fingerlings. Pepsin digestibility and multienzyme digestibility were useful in predicting protein quality. Assays for total volatile basic-nitrogen, available lysine, sulphydryl groups, and disulphide bonds, particularly when used alone, were of limited value. Atlantic salmon fingerlings (initial weight 7.65 +or- 0.14 g) fed on diets containing 2 steam-dried herring meals and Norse-LT94Reg. for 70 days were evaluated on the basis of weight gain, protein efficiency ratio, net protein ratio, net protein utilization, and slope assay. Fish fed Norse-LT94Reg. gained 11-60% more weight than fish fed on the Canadian meals. The protein quality of the Canadian fish meals was lower than that of Norse-LT94Reg.. Pepsin digestibility ranked the fish meals in the same order as the biological tests, but the biological testing of feedstuffs to determine protein quality is recommended as the final method of comparison.     Genetic impacts of hybrid catfish farming (Clarias macrocephalus x C. gariepinus) on native catfish populations in central Thailand

    Escaped hybrid catfish (female Thai walking catfish, Clarias macrocephalus x male African catfish, C. gariepinus) from farms in central Thailand may interbreed with C. macrocephalus individuals in the wild. We assessed genetic introgression of C. gariepinus genes into four wild and two broodstock populations of C. macrocephalus based on diagnostic alleles at six allozyme loci and one microsatellite locus. A total of 22 out of 515 C. macrocephalus individuals examined had C. gariepinus alleles. One individual had C. gariepinus alleles at all six loci while the 21 remaining individuals had C. gariepinus alleles at one to two loci. In each population, the diagnostic markers detected between one and five individuals (1% to 11% of the sample) bearing hybrid genotypes. Farmers' inadvertent use of introgressed C. macrocephalus individuals as broodstock in producing hybrid catfish could result in loss of hybrid vigor for growth or disease resistance. We also evaluated existing genetic diversity within and among wild and broodstock C. macrocephalus populations using three microsatellite DNA markers. Compared to other Clarias species, C. macrocephalus had moderately high genetic variation within and among populations. Within each population, the number of alleles per locus ranged from 4 to 18 and heterozygosities ranged from 0.34 to 0.89. Narathivas, a southern population had the lowest microsatellite variation, allelic diversity (three alleles per locus) and heterozygosity (HO=0.34). Within the central Thailand populations, the Ang Thong broodstock population expressed the lowest genetic diversity. Pair-wise exact tests suggested genetic divergence among most studied populations (31 of 36 tests, p<0.01). Cavalli-Sforza's chord genetic distances indicated strong genetic divergence between the Narathivas population (i.e., southern Thailand) and the rest of the sampled populations. Within central and northern Thailand, the Ang Thong Broodstock (central), Ayutthaya (central), Phitsanulok 1 (northern) and Phitsanulok 2 (northern) populations were genetically diverged from the rest of the central populations. Our results suggested low levels of genetic introgression within examined wild and broodstock populations of C. macrocephalus and moderately high genetic diversity within and among these populations.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed.
