    Accumulating evidence has indicated that abnormal microRNAs (miRNAs) serve critical roles in carcinogenesis and development of osteosarcoma (OS). The purpose of the present study was to elucidate the relationship between miR-766-3p and development of osteosarcoma and explore the potential mechanism. In this study, we found that miR-766-3p was the most downregulated miRNA by analyzing GSE65071 from the GEO database. miR-766-3p was lowly expressed in OS tissue samples and cells, and high miR-766-3p expression repressed the malignant level of OS, including cell proliferation, EMT, migration, and invasion in vitro and in vivo. B-Cell Lymphoma 9-Like Protein (BCL9L) was negatively associated with miR-766-3p expression in OS cells and tissue samples and was validated as the downstream target by luciferase reporter assay and western blotting. Rescue experiment indicated that BCL9L could restore the influence of miR-766-3p on OS cells. In addition, the β-Catenin/TCF-4 signal pathway was demonstrated to be related to the miR-766-3p/BCL9L axis. In summary, miR-766-3p, a negative regulator of BCL9L, plays the role of tumor metastasis suppressor via the β-catenin signaling pathway in the progression of OS.</p

    CircFKBP3 absence alleviates oxygen glucose deprivation-induced function loss of human brain microvascular endothelial cells <i>in vitro</i> via governing the miR-766-3p/TRAF3 axis

    Brain microvascular endothelial cell (BMEC) functions loss is a key event in the development of ischemic stroke, which may be affected by the dysregulation of circular RNAs (circRNAs). We aimed to unveil the role of circRNA FKBP Prolyl Isomerase 3 (circFKBP3) in cell models of ischemic stroke. Cell models of ischemic stroke were constructed in human BEMCs (HBMECs) with the treatment of oxygen glucose deprivation (OGD). Quantitative real-time PCR (qPCR) and western blotting were conducted for expression analysis of circFKBP3, miR-766-3p and TNF receptor associated factor 3 (TRAF3). CCK-8, transwell, wound healing, flow cytometry, tube formation and ELISA assays were implemented to monitor cell viability, migration, apoptosis, angiogenesis and inflammation production. The putative binding relationship between miR-766-3p and circFKBP3 or TRAF3 was validated by dual-luciferase, RIP and pull-down assays. CircFKBP3 expression was elevated in OGD-treated HBMECs. OGD suppressed HBMEC viability, migration, angiogenesis, and provoked cell apoptosis and inflammation production, while knockdown of circFKBP3 attenuated these effects. CircFKBP3 interacted with miR-766-3p, and circFKBP3 absence-repressed HBMEC function loss and inflammation were recovered by miR-766-3p inhibition. CircFKBP3 targeted miR-766-3p to regulate TRAF3 expression. MiR-766-3p enrichment-repressed HBMEC function loss and inflammation were recovered by TRAF3 overexpression. CircFKBP3 absence alleviated OGD-induced function loss and inflammatory responses of HBMECs via governing the miR-766-3p/TRAF3 axis. CircFKBP3 expression is elevated in OGD-treated HBMECs. OGD-induced HBMEC function loss and inflammation are alleviated by circFKBP3 absence. CircFKBP3 directly targets miR-766-3p to regulate TRAF3 expression.</p

    Circular RNA CircSHKBP1 accelerates the proliferation, invasion, angiogenesis, and stem cell-like properties via modulation of microR-766-5p/high mobility group AT-hook 2 axis in laryngeal squamous cell carcinoma

    Laryngeal squamous cell carcinoma (LSCC) is a common malignancy in head and neck. Circular SHKBP1 (circSHKBP) exerts momentous functions in the occurrence of many cancers including LSCC. Thus, we investigated the oncogenic capacities of circSHKBP1 in LSCC, and revealed the underlying mechanism as a competing endogenous RNA. The expression levels of circSHKBP1, miR-766-5p, and high mobility group AT-hook 2 (HMGA2) were examined by quantitative real-time PCR and their influences on the overall survival were measured by Kaplan–Meier method. The correlations between circSHKBP1 and miR-766-5p or HMGA2 were detected by Spearman’s rank correlation analysis. In vitro, the influences of circSHKBP1/miR-766-5p/HMGA2 axis on the tumorigenesis of LSCC were examined by CCK-8, transwell, sphere formation, and angiogenesis assays, respectively. circSHKBP1 expression was up-regulated in the LSCC specimens and cell lines. And elevated circSHKBP1 expression was closely linked to poor prognosis. Silencing circSHKBP1 expression restrained cell proliferation, invasion, angiogenesis, stem cell-like properties and tumor growth. We observed that miR-766-5p was down-regulated and negatively correlated to circSHKBP1 in LSCC samples. However, HMGA2 was highly expressed and positively associated with circSHKBP1 in these specimens. Importantly, the levels of circSHKBP1, miR-766-5p, and HMGA2 were closely associated with patients’ clinical parameters including lymph nodes metastasis and TNM stages. Mechanistic analysis clarified that circSHKBP1 sponged miR-766-5p to regulate HMGA2, the target of miR-766-5p. Moreover, miR-766-5p inhibition and overexpression of HMGA2 rescued the tumor-suppressing roles of circSHKBP1 downregulation in LSCC. In conclusion, circSHKBP1 accelerated the tumorigenesis of LSCC via modulating HMGA2 by targeting miR-766-5p.</p

    Valanginian to Barremian Benthic Foraminifera from ODP Site 766 (Leg 123, Indian Ocean)

    A Valanginian to Barremian bathyal foraminiferal assemblage (118 taxa belonging to 51 genera) is documented from ODP Site 766, drilled near the foot of the Exmouth Plateau off northwest Australia. The majority of taxa are cosmopolitan species previously described from the boreal assemblages of northern Europe and the northern margins of the Tethys, but 32 taxa could not be assigned to previously described species and are left in open nomenclature. The chronostratigraphy of this section is based upon nannofossils, palynomorphs, and radiolarians. The foraminiferal succession is divided into four assemblages based on the first and last occurrences of characteristic benthic foraminifera: 1) An upper Valanginian Lenticulina ouachensis - Textularia bettenstaedti assemblage with Ammodiscus tenuissimus, Bulbobaculites sp., and Aaptotoichus clavellatus. The last occurrences of these taxa are observed near the transition between the Valanginian and Hauterivian. 2) A lower Hauterivian assemblage is characterized by Lenticulina heiermanni and Saracenaria forticosta. 3) An upper Hauterivian assemblage contains Planularia crepidularis, Patellina subcretacea, and Reinholdella hofkeri. A major faunal discontinuity is observed at the top of this assemblage between Cores 766A-32R and 766A-30R, probably associated with a depositional gap between the late Hauterivian and the Barremian. 4) The overlying Barremian assemblage is characterized by Gavelinella barremiana, Glomospira spp., and Pseudogaudryinella sp. This record enables long-distance comparisons of the stratigraphic ranges of benthic foraminiferal species in the austral bioprovince with their reported ranges in the North Atlantic region and the northern Tethys. Although the assemblages at Site 766 consist of ca. 74% cosmopolitan species, they differ from Tethyan assemblages in the rarity or absence of index taxa such as Praedorothia spp., Lenticulina nodosa and L. eichenbergi, as well as ornamented, palmate morphotypes such as Citharina spp., Flabellina spp., and Frondicularia spp. They differ from the typical boreal assemblages by the lack of diverse agglutinated taxa. As a result, benthic foraminiferal zonations established for the Lower Cretaceous of the northern Tethys and Tethyan DSDP sites could not be applied at Site 766.     Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
