Characterization of nitrite degradation by Lactobacillus casei subsp. rhamnosus LCR 6013.
Nitrites are potential carcinogens. Therefore, limiting nitrites in food is critically important for food safety. The nitrite degradation capacity of Lactobacillus casei subsp. rhamnosus LCR 6013 was investigated in pickle fermentation. After LCR 6013 fermentation for 120 h at 37°C, the nitrite concentration in the fermentation system was significantly lower than that in the control sample without the LCR 6013 strain. The effects of NaCl and Vc on nitrite degradation by LCR 6013 in the De Man, Rogosa and Sharpe (MRS) medium were also investigated. The highest nitrite degradations, 9.29 mg/L and 9.89 mg/L, were observed when NaCl and Vc concentrations were 0.75% and 0.02%, respectively in the MRS medium, which was significantly higher than the control group (p ≤ 0.01). Electron capture/gas chromatography and indophenol blue staining were used to study the nitrite degradation pathway of LCR 6013. The nitrite degradation products contained N2O, but no NH4(+). The LCR 6013 strain completely degraded all NaNO2 (50.00 mg/L) after 16 h of fermentation. The enzyme activity of NiR in the periplasmic space was 2.5 times of that in the cytoplasm. Our results demonstrated that L. casei subsp. rhamnosus LCR 6013 can effectively degrade nitrites in both the pickle fermentation system and in MRS medium by NiR. Nitrites are degraded by the LCR 6013 strain, likely via the nitrate respiration pathway (NO2(-)>NO->N2O->N2), rather than the aammonium formation pathway (dissimilatory nitrate reduction to ammonium, DNRA), because the degradation products contain N2O, but not NH4(+)
MİKRODALGA FIRINDA SUNİ YAŞLANDIRILAN 6013 ALÜMİNYUM ALAŞIMIN KOROZYON DAVRANIŞININ İNCELENMESİ
Alüminyum alaşımları günümüzde düşük yoğunluk ve yüksek mukavemet elde edilebilen bir alaşım türüdür. Bu alaşımlar özellikle yaşlandırma işlemiyle çökelti oluşturularak daha fazla mukavemetli olur. Bu çalışmada, 6013 alüminyum alaşımını mikrodalga fırında suni yaşlandırarak dayanım değerleri arttırılmıştır. Mikrodalga fırında yaşlandırma yöntemi numuneleri hacimsel ve homojen bir şekilde yaşlanmasını sağlamıştır. 6013 alüminyum alaşımları, mikrodalga fırında saf argon gazı atmosferinde suni yaşlandırma işlemine tabi tutulduktan sonra sertlik analizleri, iç yapı (optik mikroskop, SEM) karakterizasyonu çalışılmıştır. Yaşlandırma ısıl işlemleri sunucu numunelerin içerisinde oluşan fazların varlığı XRD eğrileri ile tespit edilmiştir. Suni yaşlandırılmış numunelerin iç yapısında mevcut olan fazların korozyon analizleri gerçekleştirilmiştir. 6013 Alüminyum alaşımının mikrodalga fırında suni yaşlandırma sürelerine bağlı olarak numunelerde meydana gelen çökeltilerin mekanik özellikleri korozyon direnç değerlerini etkilediği görülmüştür

Pengaruh Post-Weld Heat Treatment T6 Pada Laju Perambatan Retak Fatik Paduan Al 6013
The effects of post-weId heat treatment T6 on the fatigue crack growth rate of aluminum alloy Al 6013 have been investigated. The investigation was performed under three material conditions : base metal, weld metal without heat treatment, and weld metal with heat treatment. Welding process used filler AI 5356 consisting of longitudinal butt joint and transversal butt joint. In addition, the post-weld heat treatment T6 involved solution treatment, quenching, and artificial aging.
The test of fatigue crack growth rate was performed on two types of specimen: single edge crack specimen for longitudinal butt weld joint and center crack tension specimen for transversal butt weld joint. The test was performed on those specimens with a constant load amplitude at stress level of 20% and stress ratio of 0.3. The load frequency was adjusted from 5 to 15 Hz. The results were treated using seven incremental polynomial method according to the ASTM E-647 to obtain the fatigue crack growth rate curve (da/dN vs AK) and Paris equation.
The results show that welding process can increase the fatigue crack growth rate of Al 6013, signed by the increase of n value from 3.25 to 3.93 for longitudinal butt joint, and from 3.09 to 3.70 for transversal butt joint. The post-weld heat treatment also increase the fatigue crack growth rate of AI 6013. The results also show that the strength, elongation, and hardness of Al 6013 decrease after the welding process and the post-weld heat treatment, and there is a change in micro structures that influence the fatigue crack growth rate of Al 6013.
