Effects of RH-5992 on ecdysteroidogenesis of the prothoracic glands during the fourth larval instar of the silkworm, Bombyx mori
Stage-dependent effects of RH-5992 on ecdysteroidogenesis of the prothoracic glands during the fourth larval instar of the silkworm, Bombyx mori, were studied in the present report. When larvae were treated with RH-5992 during the early stages of the fourth larval instar (between day 0 and day 1), initially ecdysteroid levels in the hemolymph were inhibited. However, 24 h after RH-5992 application, ecdysteroid levels were greatly increased as compared with those treated with acetone. The examination of the in vitro prothoracic gland activity upon RH-5992 application during the early stages of the fourth larval instar confirmed a short-term inhibitory effect. When RH-5992 was applied to the later stages of the fourth larval instar, no effects on both hemolymph ecdysteroid levels and prothoracic gland activity were observed. Addition of RH-5992 to incubation medium strongly inhibited ecdysteroid secretion by the prothoracic glands from the early fourth instar, indicating direct action of RH-5992 on ecdysteroidogenesis by prothoracic glands. Four hours after application with RH-5992 on day 1.5, prothoracic glands still showed an activated response to PTTH in both PTTH-cAMP signaling and the extracellular signal-regulated kinase (IRK) signaling. Moreover, addition of RH-5992 to incubation medium did not interfere with the stimulatory effect of the glands to PTTH in ecdysteroidogenesis. These results indicated that both PTTH-cAMP signaling and PTTH-ERK signaling may not be involved in short-term inhibitory regulation by RH-5992
RH-5992—An ecdysone agonist on model system of the silkworm <i>Bombyx mori</i>
137-144RH-5992 is a novel synthetic non-steroidal
ecdysteroid agonist with a high selectivity towards Lepidopteran species. The
effect of ecdysone agonist RH-5992 on larval period, larval weight, silk gland
weight and haemolymph protein profile were examined in the model organism, the larvae
of silkworm, Bombyx mori . The LD50 values were found to be 16.21
and 12.01μg/larva for 72 and 96 hr respectively. In the present study, three sublethal
concentrations of l/5th, 1/10th and 1/20th of LD50
at 72 hr were selected and applied on the mid-dorsal line of the silkworm B.
mori. The maximum mortality of 35% was observed in the group which received
the highest (3.2μg/larva) concentration of RH-5992. The mortality rate was
found to be dose dependent as well as time dependent. Interesting results were
observed in haemolymph profile of the RH-5992 treated larvae as staining intensity
of 30kDa protein decreased significantly whereas the effect was not marked on other
major proteins like storage proteins and vitellogenin polypeptides. From the results,
it is confirmed that RH-5992 causes changes in larval characters and protein profile
of silkworm B.mori It is proposed that RH-5992 may cause negative effect
specifically on reproductive characters like development of ovary and egg production
due to decrease in 30kDa protein
Apple, Control of the Pandemis Leafroller with RH-5992, 1993
Abstract
An experimental insecticide, RH-5992, was evaluated for its ability to control PLR larvae of the overwintering generation. The test was conducted in an apple orchard near Wenatchee. Trees were 10-yr-old spur-type (‘Oregon Spur II’) on seedling roots. Treatments were applied to three-tree plots replicated five times using a RCB design. All treatments were applied with a handgun sprayer at 300 psi to the point of drip, simulating a dilute spray of approximately 400 gpa. Two rates of RH-5992 plus Lorsban and methyl-parathion were applied at the half-inch green tip (HIG) stage of apple bud development (19 Apr). Two rates of RH-5992 were also applied at the pink stage of apple bud development (5 May). Pre-treatment evaluations were made on 19 Apr by examining 20 fruiting buds per tree and recording the number infested. The post-treatment evaluation was made on 20 May when surviving larvae could be easily located in flower clusters and growing shoots. Each tree was sampled and the number of live PLR larvae recorded. On the first day following the 19 Apr application, five spurs were collected from the middle tree within each replicate of each RH-5992 treatment and the untreated plot (25 total per treatment). Spurs were returned to the laboratory and one was placed in a 2-oz plastic portion cup. One 10-d-old PLR larva reared on artificial diet, usually a third instar, was placed in the cup and the lid put in place. After the 5 May applications, 10 spurs were collected from each RH-5992 treatment and the untreated plot (50 total per treatment). At the same time, PLR larvae were collected from the same orchard where the test was conducted. Spurs were returned to the laboratory, and one was placed in a 2-oz plastic portion cup. One 10-d-old PLR larva reared on artificial diet, usually a third instar, was placed in the cup and the lid put in place. In addition, field-collected leafroller larvae were placed on spurs in plastic cups and handled the same as the laboratory-reared larvae. All larvae were kept at 70°F for 7 d, after which they were examined and the number of live or dead (dead included larvae which were alive but with slipped head capsules) larvae was recorded.</jats:p
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Lignin-induced Expression of Aspergillus oryzae 5992 Genes using Suppression Subtractive Hybridization
A previous study reported that a novel Aspergillus oryzae strain (CGMCC5992) can synthesize lignin hydrolytic enzymes for lignin degradation from straw. The present work involves the different gene expression of A. oryzae 5992 grown in media using lignin and glucose as carbon sources by suppression subtractive hybridization. Surprisingly, peroxidase was found in up-regulation genes, which is the key enzyme for degrading lignin. This shows that A. oryzae 5992 can secrete peroxidase in the presence of lignin. The functions of up-regulation genes also included gluconeogenesis, repairs, as well as signal and transporter proteins in the cell membrane. In addition, the down-regulation of genes was closely related to the aerobic metabolism of glucose, the fatty acid synthesis of the cell membrane, and the synthesis and utilization of ATP. Therefore, A. oryzae could regulate metabolism using lignin as carbon source, including lignin degradation promotion, glucose metabolism inhibition, and glucose regeneration
