Evolution of HLA-B*5703 HIV-1 escape mutations in HLA-B*5703-positive individuals and their transmission recipients.
HLA-B*57 is the class I allele most consistently associated with control of human immunodeficiency virus (HIV) replication, which may be linked to the specific HIV peptides that this allele presents to cytotoxic T lymphocytes (CTLs), and the resulting efficacy of these cellular immune responses. In two HIV C clade-infected populations in South Africa and Zambia, we sought to elucidate the role of HLA-B*5703 in HIV disease outcome. HLA-B*5703-restricted CTL responses select for escape mutations in three Gag p24 epitopes, in a predictable order. We show that the accumulation of these mutations sequentially reduces viral replicative capacity in vitro. Despite this, in vivo data demonstrate that there is ultimately an increase in viral load concomitant with evasion of all three HLA-B*5703-restricted CTL responses. In HLA-B*5703-mismatched recipients, the previously described early benefit of transmitted HLA-B*5703-associated escape mutations is abrogated by the increase in viral load coincident with reversion. Rapid disease progression is observed in HLA-matched recipients to whom mutated virus is transmitted. These data demonstrate that, although costly escape from CTL responses can progressively attenuate the virus, high viral loads develop in the absence of adequate, continued CTL responses. These data underline the need for a CTL vaccine against multiple conserved epitopes

Hydrolytic Degradation of Estane 5703
Relative to the unaged Estane{reg_sign} 5703, heating decreases the modulus and ultimate stress for both the dry and humid samples. The sample aged in dry air, quickly recovered to its original mechanical properties. The reversibility of the mechanical properties is a result of the reorganization of the hard segments into domains. For the Estane 5703 samples aged in humid air, the mechanical properties only partially recovered with time. Molecular weight data confirm that differences in the extent of the recovery for the samples aged under humid environments are likely due to the hydrolytic scission of the polyester soft segments. Since the tensile moduli are similar for the samples aged in both the dry and humid environments, it is believed that the phase behavior of the hard domains dictate the mechanical properties at low extensions. However, at higher elongations, the mechanical properties are dominated by the soft segments. Both the influences of morphological changes and chemical changes induced by heating must be considered in multiphase polymers when accelerated aging is utilized to predict long term polymer properties. After accelerated aging experiments, the morphology must be allowed to recover to equilibrium conditions in order to assess the extent of degradation and aging. Also, the effect of different humidities on the rate of hydrolytic degradation are presented

Thermal aging of nitroplasticized Estane 5703
In support of the Lifetime Prediction modeling effort, we have been investigating the aging processes that could impact the lifetime of PBX 9501. PBX 9501 is composed of 95% HMX and 5% polymeric binder. The polymeric binder is composed of 50% nitroplasticizer (NP) and 50% Estane{reg_sign} 5703 with a small quantity of stabilizer. Estane{reg_sign} 5703 is a segmented poly(ester urethane) with mechanical properties derived from phase separation of hard and soft segments along the polymer backbone. Since the binder has a significant effect on the composite mechanical properties, it is essential to know how the binder changes with time. Typically, polymer lifetime predictions are determined from extrapolation of properties after the material has been exposed to elevated temperatures and/or reactive environments for varying periods of time. For multiphase polymers, this accelerated aging methodology is very difficult to interpret since elevated temperatures alter the physical structure of the polymer, as well as, accelerate the chemical degradation reactions. Accelerated aging studies of nitroplasticized Estane have shown an increase in the molecular weight. The increase in molecular weight is most likely due to polymer chain branching reactions that eventually leads to formation an insoluble cross-linked gel. The decreased chain mobility caused by branching may also affect phase separation, which in turn, may change the mechanical properties. The HLA-Cw*1801 specificity, a Cw7/Cw4 hybrid allele, has recently been described in association with B*8101 (formerly B“DT”). In this study, the new Cw*1802 variant, differing from Cw*1801 at exon 5. is found associated with B*5703 in Bubi individuals from Equatorial Guinea. Confirmatory complete coding regions of B*5703 and B*3910 are also reported
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
