Abstract 5513: Notch directly represses transcription by recruitment of PRC2 to the ternary complex
Abstract
It is well established that Notch functions as a transcriptional activator through the formation of a ternary complex that comprises Notch, Maml and CSL. This ternary complex then serves to recruit additional transcriptional cofactors that link to higher order transcriptional complexes. The mechanistic details of these events remain unclear. In this study, we report that the Notch ternary complex can also serve to direct the formation of a repressor complex to terminate genes expression of select target genes. Herein we demonstrate that p19Arf and Klf4 are transcriptionally repressed directly by Notch. Data indicate that Notch recruits PRC2 and Lsd1 to these promoters, which leads to changes in the state of the epigenetic landscape and repression of transcription. The demethylase activity of Lsd1 is a pre-requisite for Notch-mediate transcriptional repression. Furthermore, we identified a stable Notch transcriptional repressor complex containing Lsd1, PRC2 and the Notch ternary complex. This study demonstrates proof of concept for a novel function of Notch and provides further insight into the mechanisms of Notch mediated tumorigenesis. This finding provides rationale for the targeting of epigenetic enzymes to inhibit Notch activity or use in combinatorial therapy to provide a more profound therapeutic response.

Identification of a novel HLA-B*55 variant (B*5513) from a Korean family.
The novel allele, B*5513, was identified from three siblings of a Korean family. Direct DNA sequencing analysis revealed that B*5513 differed from B*5502 by a single-nucleotide substitution at codon 116 (TTA-->TTC) resulting in an amino acid change from leucine to phenylalanine. Preparation and Characterization of PEBAX-5513/AgBF4/BMIMBF4 Membranes for Olefin/Paraffin Separation
In this study, we investigated a poly(ether-block-amide)-5513 (PEBAX-5513)/AgBF4/1-butyl-3-methylimidazolium tetrafluoroborate (BMIMBF4) composite membrane, which is expected to have a high stabilizing effect on the Ag+ ions functioning as olefin carriers in the amide group. Poly(ethylene oxide) (PEO) only consists of ether regions, whereas the PEBAX-5513 copolymer contains both ether and amide regions. However, given the brittle nature of the amide, the penetration of BMIMBF4 remains challenging. The nanoparticles did not stabilize after their formation in the long-term test, thereby resulting in a poor performance compared to previous experiments using PEO as the polymer (selectivity 3; permeance 12.3 GPU). The properties of the functional groups in the polymers were assessed using Fourier transform infrared spectroscopy, scanning electron microscopy, and thermogravimetric analysis, which confirmed that the properties endowed during the production of the film using the ionic liquid can impact the performance
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.
