    To provide better therapeutic avenues for treating tongue squamous cell carcinoma (TSCC), a series of experiments about the effects of microRNA (miR)-532-3p on TSCC malignant behaviors were carried out. The result showed that miR-532-3p was down-regulated and C-C chemokine receptor 7 (CCR7) was up-regulated in the tumor tissues compared with those in the paired paratumor tissues. Further, expression of miR-532-3p was detected in four TSCC cell lines, TSCCA, TCA8113, CAL-27, and SCC-25. The miR-532-3p mimics and inhibitor were transfected into the CAL-27 and TCA8113 cell lines which were the relatively lowest and highest miR-532-3p expressions, respectively. It was found that the overexpression of miR-532-3p suppressed TSCC cell proliferation, migration, invasion, and promoted apoptosis in vitro, whilst the knockdown of miR-532-3p reversed these behaviors. The bioinformatics predicted that CCR7 was a downstream gene of miR-532-3p, which was confirmed via luciferase assay. Following, the decline of CCR7 in the miR-532-3p mimics group and the rise of CCR7 in the miR-532-3p inhibitor group were also verified. In addition, enhanced cell proliferation, migration and invasion induced by CCR7 were partly restrained by miR-532-3p in TSCC cell. Meanwhile, miR-532-3p attenuated tumourigenesis in vivo due to the reduction of tumor volume and Ki-67 positive rate and the increase of apoptotic cells. Taken together, these findings reveal a pivotal role for the miR-532-3p/CCR7 axis in regulating TSCC, and this novel axis could be suitable for therapeutic intervention in TSCC disease.</p

    Planktonic foraminiferal assemblages of DSDP Hole 75-532

    Qualitative and quantitative analyses of planktonic foraminiferal assemblages from Deep Sea Drilling Project site 532 shed light on hydrographic changes over the Walvis Ridge during the past 500,000 years. From changes in distribution of foraminiferal assemblages, two major hydrographic regimes (coastal and geostrophic branches of the Benguela Current and the Angola Current) can be distinguished at site 532. It is suggested that the hydrographic situation on the northeastern Walvis Ridge was characterized by intensified upwelling and a westward expansion of the coastal upwelling cells during several global cooling pulses. During glacial stages 2-4, the middle part of stage 6, sporadically from the lower stage 8 through upper stage 10, and during stage 12, site 532 was located beneath the coastal branch of the Benguela Current because faunal distribution patterns indicate intensified upwelling. The Angola Current probably intruded the area of study during the lower stages 5, sporadically 6-8, and 11, as documented by the increased abundance of Neogloboquadrina dutertrei

    CaCO3, organic carbon and mass accumulation rates at DSDP Site 75-532

    Detailed carbonate and organic-carbon stratigraphies were constructed from samples collected every 20 cm in a 250-m hydraulic piston core recovered at DSDP Site 532 on Walvis Ridge. This sampling interval represents about one sample every 5000 yr., based on sediment accumulation rates calculated from nannofossil biostratigraphic zones. All samples were analyzed for percent CaCO3, resulting in a detailed carbonate stratigraphy for the past 5.0 m.y. The samples for the top 110 m of section were also analyzed for organic carbon in order to construct a detailed organiccarbon stratigraphy for the last 2.5 m.y. The recovered section has distinct dark-light color cycles with average periodicities of 55, 58, and 30 k.y. for the Quaternary, upper Pliocene, and lower Pliocene, respectively. Periodicities of carbonate cycles are similar to the color cycles; most carbonate minima correspond to the dark parts of color cycles. The average periodicity for carbonate cycles is about 36 k.y. Darker parts of color cycles usually contain higher concentrations of organic carbon, but the organic- carbon record does not follow the cyclicity of the color cycles in detail, at least for the last 2.5 m.y. Organic-carbon cycles have an average periodicity of about 34 k.y. for the Quaternary and upper Pliocene. The cycles of CaCO3 and color have periodicities similar to those reported from carbonate stratigraphies from the northeast Atlantic, Caribbean, and eastern equatorial Pacific. The carbonate cycles at Site 532 are the result of external forcing, probably related to global climate, that affected fluctuations in both sediment supply from the African continental margin and productivity of siliceous organisms. The organic-carbon cycles have similar periodicities and similar changes in periodicities to those of the CaCO3 cycles. Semiquantitative estimates of diatom abundance from smear slides and concentrations of biogenic SiO2 calculated from chemical analyses suggest that upwelling at Site 532 was minor until about 3 m.y. ago. The Benguela-Current upwelling system either began at that time or, more likely, migrated into the area of Site 532, where it prevailed until some time between about 1.2 and 0.5 m.y. ago. The increase and decline of upwelling in the area of Site 532, however, did not disturb the trend of cyclicities of carbonate and organic carbon. The latest change in conditions at Site 532 was an increase in intensity of bottom currents during the past 0.5 m.y. that winnowed nannofossils, diatoms, and clay and left a lag deposit represented by a foraminifer-rich fades

    Circ_0007841 knockdown confers cisplatin sensitivity to ovarian cancer cells by down-regulation of NFIB expression in a miR-532-5p-dependent manner

    Cisplatin (DDP) is first-line management for ovarian cancer (OC). Previous data have suggested that circular RNA_0007841 (circ_0007841) regulates OC progression; however, there is no data on its role in the sensitivity of OC cells to DDP. RNA expression of circ_0007841, microRNA-532-5p (miR-532-5p) and nuclear factor I B (NFIB) was detected by quantitative real-time polymerase chain reaction in OC patient samples and OC cell lines. Protein expression was checked by Western blotting analysis. Cell viability, proliferation, cell apoptotic rate, migration and invasion were investigated by 3-(4,5)-dimethylthiahiazo (-z-y1)-3,5-diphenytetrazoliumromide, 5-Ethynyl-29-deoxyuridine, flow cytometry analysis, scratch test and transwell assays, respectively. The interactions among circ_0007841, miR-532-5p and NFIB were identified by a dual-luciferase reporter assay. Xenograft mouse model assay was performed to determine the effect of circ_0007841 on DDP sensitivity in vivo. Circ_0007841 and NFIB expression were upregulated, whereas miR-532-5p was downregulated in DDP-resistant OC tissues and cells compared with controls. Circ_0007841 silencing improved DDP sensitivity, inhibited cell proliferation, invasion and migration, but induced cell apoptosis in DDP-resistant OC cells. Circ_0007841 acted as a miR-532-5p sponge and regulated DDP resistance and OC cell malignancy through miR-532-5p in DDP-resistant OC cells. Besides, the overexpression of NFIB, a target of miR-532-5p, remitted miR-532-5p-mediated effects in DDP-resistant OC cells. Circ_0007841 depletion conferred DDP sensitivity to DDP-resistant OC cells in vivo. Further, circ_0007841 was secreted from DDP-resistant OC cells through being packaged into exosomes. Circ_0007841 conferred DDP resistance to DDP-resistant OC cells through the miR-532-5p/NFIB axis, suggesting the potential of circ_0007841 as a therapeutic target for OC.</p

    An investigation of the quantitative composition of the coarse (> 40 µm) and clay (< 2 µm) fraction of HPC 532, DSDP Leg 75, in 1300 m water depth on the eastern Walvis Ridge off Southwest Africa yielded the following results: (1) The sediments reflect a complete Latest Miocene to Recent depositional history. Sedimentation rates vary between 2.3 and 7.8 cm/ka. (2) Preservation of calcium carbonate is subject to strong variations: short-term (< 100,000 years) and long-term (about 1 m.y.) cycles in carbonate dissolution have been observed, with strongest dissolution occurring during periods of lowered sea level. (3) Upwelling influence from the near-coastal upwelling centre has been detected by means of the opal content: interglacial periods show high opal contents, because the Benguela Current turned westward at about 20°S and carried opal-laden upwelled water to the west. Sediments from glacial periods, however, show opal minima. Besides these short-term cyclic variations in opal content, long-term cycles have been found, with maximum upwelling influence in the latest Pliocene/early Quaternary. (4) Each CaCO3 dissolution minimum (maximum) is correlated with an opal maximum (minimum) throughout the sediment sequence. (5) The oceanographic system off southwest Africa remained essentially unchanged since the latest Miocene: sea level rose and fell periodically on a small and on a large scale, and the Benguela Current flowed southeast-northwest and turned to the west at the latitude of Site 532 during interglacial periods, when sea level was high. (6) The climate in the near-coastal area of southwest Africa in the latitude of Site 532 has probably been arid throughout the investigated period

    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    General descriptions, organic contents and ratios of geolipid components at DSDP Holes 75-530B and 75-532

    Distributions of free and bound n-alkanes, n-alkanoic acids, and n-alkanols were determined in order to compare the character of organic matter contained in organic-carbon-rich sediments from two sites sampled by the hydraulic piston corer. Two diatomaceous debris-flow samples of Pleistocene age were obtained from Hole 530B in the Angola Basin. A sample of bioturbated Pleistocene diatomaceous clay and another of bioturbated late Miocene nannofossil clay were collected from Hole 532 on the Walvis Ridge. Geolipid distributions of all samples contain large terrigenous contributions and lesser amounts of marine components. Similarities in organic matter contents of Hole 530B and Hole 532 sediments suggest that a common depositional setting, probably on the Walvis Ridge, was the original source of these sediments through Quaternary, and possibly late Neogene, times and that downslope relocation of these biogenic deposits has frequently occurred
