Single-Center Investigation of the Pharmacokinetics of WCK 4282 (Cefepime-Tazobactam Combination) in Renal Impairment
WCK 4282 is a combination product of cefepime (FEP) and tazobactam (TAZ) in a 1:1 ratio currently under development for the treatment of multidrug-resistant Gram-negative bacterial infections. We investigated the effect of renal impairment on the pharmacokinetics (PK) and safety of WCK 4282 in 48 subjects with various degrees of renal function. Subjects were categorized on the basis of their Cockcroft-Gault equation-estimated creatinine clearance (CL
). We enrolled 6 subjects each into those with mild (CL
, 60 to <90 ml/min), moderate (CL
, 30 to <60 ml/min), or severe (CL
, <30 ml/min) renal impairment and those with end-stage renal disease (ESRD) requiring hemodialysis and 24 healthy control subjects (CL
, ≥90 ml/min). Healthy subjects and subjects with mild and moderate renal impairment received a single 90-min infusion of 4 g of WCK 4282 (2 g FEP and 2 g TAZ). Subjects with severe renal impairment and ESRD received 2 g of WCK 4282 (1 g FEP and 1 g TAZ) over 90 min. The plasma exposure of FEP-TAZ increased as renal function decreased. In subjects with mild, moderate, and severe renal impairment and ESRD, the mean exposure (area under the plasma concentration versus time curve from time zero extrapolated to infinity) of FEP and TAZ increased by 1.3- and 1.2-fold, 2.3- and 2.3-fold, 4.7- and 4.0-fold, and 8.5- and 11.6-fold, respectively. The urinary recovery of FEP and TAZ decreased with increasing renal impairment. There were no adverse events reported during the study. The findings suggest that dose adjustments for WCK 4282 will be required according to the degree of renal impairment. A single infusion of WCK 4282 was found to be safe and well tolerated in subjects with normal and impaired renal function. Pseudolaric acid B inhibits the proliferation of human diffuse large B-cell lymphoma cell line SU-DHL-4 via miR-4282
Objective To observe the effect of pseudolaric acid B(PAB) on the proliferation and apoptosis of human diffuse large B-cell lymphoma cell line SU-DHL-4 possibly and to explore the potential mechanism through regulating miR-4282. Methods SU-DHL-4 cells were divided into control group, PAB-L group, PAB-M group, PAB-H group; According to liposome method, miR-NC and miR-4282 were transfected into the cells and were marked as miR-NC group, miR-4282 group; The anti-miR-NC, anti-miR-4282 were transfected into the cells,cultured with 20 μmol/L PAB, denoted as (PAB+anti-miR-NC)group and(PAB+anti-miR-4282)group. Cell pro- liferation was detected by CCK8 assay, and cell apoptosis was detected by flow cytometry. The expression of Ki-67,cleaved caspase-3, cleaved caspase-9 protein was detected by Western blot and miR-4282 expression was detected by RT-qPCR. Results Compared with the control group, Ki-67 protein expression in PAB-L group, PAB-M group, and PAB-H group was decreased while miR-4282 expression, and the apoptosis rate, cell inhibition rate, protein expression of cleaved caspase-3, cleaved caspase-9 were increased (P<0.05). Compared with the miR-NC group, Ki-67 protein expression in miR-4282 group decreased, while miR-4282 expression, the apoptosis rate, cell inhibition rate, protein expression of cleaved caspase-3, cleaved caspase-9 were all increased (P<0.05). Inhibition of miR-4282 expression reduced the effect of PAB on the proliferation and apoptosis of SU-DHL-4 cells. Conclusions PAB may inhibit the proliferation of human diffuse large B-cell lymphoma cell SU-DHL-4 and promote apoptosis through increased expression of miR-4282
Going Beyond Counting First Authors in Author Co-citation Analysis
The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
Variations on the Author
“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
