    Going Beyond Counting First Authors in Author Co-citation Analysis

    The present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed

    The amoebal MAP kinase response to Legionella pneumophila is regulated by DupA

    The amoeba Dictyostelium discoideum can support replication of Legionella pneumophila. Here we identify the dupA gene, encoding a putative tyrosine kinase/dual-specificity phosphatase, in a screen for D. discoideum mutants altered in allowing L. pneumophila intracellular replication. Inactivation of dupA resulted in depressed L. pneumophila growth and sustained hyperphosphorylation of the amoebal MAP kinase ERK1, consistent with loss of a phosphatase activity. Bacterial challenge of wild-type amoebae induced dupA expression and resulted in transiently increased ERK1 phosphorylation, suggesting that dupA and ERK1 are part of a response to bacteria. Indeed, over 500 of the genes misregulated in the dupA(-) mutant were regulated in response to L. pneumophila infection, including some thought to have immune-like functions. MAP kinase phosphatases are known to be highly upregulated in macrophages challenged with L. pneumophila. Thus, DupA may regulate a MAP kinase response to bacteria that is conserved from amoebae to mammals

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     [cat] L'ictus és la segona causa principal de discapacitat i mort arreu del món, amb una incidència creixent a causa del canvi demogràfic i l'augment de les malalties cardiovasculars. Recentment, s'ha identificat que el miR-3609 està associat amb un pronòstic funcional favorable en l’ictus isquèmic d’etiologia criptogènica. L'objectiu principal d’aquest treball de fi de màster ha estat identificar els gens diana regulats per aquest miRNA. Per aconseguir-ho, s’han realitzat estudis de transfecció gènica emprant molècules sintètiques mimètiques i inhibidores del miR-3609 endogen en cèl·lules endotelials microvasculars cerebrals humanes immortalitzades (hCMEC/d3). Posteriorment, s’ha estudiat el perfil d’expressió gènica mitjançant l’anàlisi de l’ARNm i de la proteïna en condicions de normòxia i hipòxia. Els resultats assenyalen el gen CRYAB, que codifica per la xaperona αB-cristal·lina, com a gen diana del miR-3609. Aquesta proteïna podria actuar en resposta a la hipòxia per induir un estat proinflamatori en les cèl·lules endotelials cerebrals. A més, s’ha observat que el miR-3609 regula CRYAB, inhibint la seva expressió en condicions d’hipòxia prolongada. Això suggereix que el miR-3609 podria ser una diana terapèutica en el futur per millorar el pronòstic funcional de pacients joves que hagin patit un ictus d’etiologia criptogènica[eng] Stroke is the second leading cause of disability and death worldwide, with a growing incidence due to demographic changes and the increase in cardiovascular diseases. Recently, miR-3609 has been identified as being associated with a favorable functional prognosis in cryptogenic ischemic stroke. The main objective of this master's thesis was to identify the target genes regulated by this miRNA. To achieve this, transfection experiments to introduce synthetic mimetic and inhibitory molecules of endogenous miR3609 into immortalized human brain microvascular endothelial cells (hCMEC/d3) were performed. Subsequently, the gene expression profile was studied through mRNA and protein analysis under normoxic and hypoxic conditions. The results indicate that CRYAB gene, which encodes the αB-crystallin chaperone, is a target gene of miR-3609. This protein may respond to hypoxia by inducing a pro-inflammatory state in brain endothelial cells. Additionally, miR-3609 was observed to regulate CRYAB by inhibiting its expression under prolonged hypoxic conditions. This suggests that miR-3609 could be a therapeutic target in the future to improve the functional prognosis of young patients who have suffered a cryptogenic strok
