Mir-3179'un Küçük Hücre Dışı Akciğer Kanseri İnvazyon ve Metastazını Düzenleyici Rolünün İn Vitro ve İn Vivo Olarak Saptanması
No full textKüçük hücreli dışı akciğer kanseri yüksek oranda metastaz yapma yeteneğinden dolayı oldukça agresif bir kanser türüdür ve PI3K/Akt/NF-?B yolağı KHDAK hücre hatlarında genellikle aktifdir. Ayrıca tümör baskılayıcı ya da onkogen olarak görev yapan mikroRNA?lar da dahil olmak üzere birçok genin transkripsiyonel regülasyonunda da etkili olduğu görülmüştür. Daha önceki çalışmalarımızda NF-?B?nin çeşitli mikroRNA?ları pozitif veya negatif olarak regüle ettiğini göstermiştik. Bu çalışmada miR-3179?un KHDAK?inde invazyon ve metastazdaki rolünü bulmayı amaçladık. Öncelikle invasiv kapasitesi yüksek olan A549 ve H1299 hücre hatlarındaki miR-3179 ifadesinin invaziv kapasitesi daha az olan H23 ve H1975 hücre hatlarına kıyasla daha düşük olduğunu gösterdik. A549 hücre hatlarında miR-3179?u overeksprese ettiğimizde, Snail ve N-cadherin ifadesinin azaldığı ve hücrelerin E-cadherin ekspresyonunu yeniden kazandığı görüldü. Ayrıca, miR-3179 ifadesini H23 ve H1975 hücre hatlarında inhibe ettiğimizde, Snail ve N-cadherin ifadesi artarken E-cadherin ifadesi azalmıştır. Bu yüzden miR-3179 EMT?yi ve hücre invazyonunu engelleyebilir. Ayrıca miR-3179?un KHDAK hücrelerinin metastatik kabiliyetini engelleme potensiyelini nude fare modelinde inceledik. Bunun için A549-Empty, A549-miR3179, H23-Empty and H23-miR3179Off hücreleri nude farelerin akciğerlerinin plevral boşluğuna enjekte edildi ve 35 günün sonunda fareler sakrifiye edilerek makro/mikro metastatik nodüller değerlendirildi. Sonuçlarımız, artan miR-3179 ifadesinin A549 hücrelerinde kontrol hücre grubuna kıyasla bu hücrelerin metastatik yeteneğini inhibe ettiğini ve H23 hücrelerinde miR-3179 ifadesinin inhibe edilmesinin yine kontrol hücrelerine oranla bu hücrelere metastaz yeteneği kazandırdığını açıkça göstermiştir. MiR-3179?un KHDAK?inde EMT, invazyon ve metastazı baskılayıcı etkisinin hangi mekanizma ile yaptığını da inceledik. Sonuç olarak miR-3179?un KHDAK hücrelerinde TAB1 genini doğrudan hedef aldığı ve böylece Smad2/3, Akt, p65 fosforilasyonu ile Twist ve Snail ifadesini azaldığı, PTEN ifadesinin ise arttığı görülmüştür. miR-3179 ifadesini inhibe ettiğimizde ise tersine Smad2/3, Akt, p65 fosforilasyonu ile Twist ve Snail ifadelerinin arttığı PTEN ifadesinin ise azaldığı gösterilmiştir. Böylece miR-3179?un invazyon ve metastaz için supresör miRNA olarak görev yaptığını ve KHDAK invazyon ve metastazını TAB1 genini hedef alarak engellediğini gösterdik
Going Beyond Counting First Authors in Author Co-citation Analysis
Get PDFThe present study examines one of the fundamental aspects of author co-citation analysis (ACA) - the way co-citation
counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Reasons for these effects are discussed
