Producción de autoinductor-2 por Bifidobacterium dentium ATCC 27534 y sus efectos en la adhesión y formación de biopelículas

Introducción: Bifidobacterium dentium es un microorganismo anaeróbico, Gram positivo, acidógeno y acidúrico, que ha sido aislada en la cavidad oral a partir de saliva y lesiones de caries, relacionándose al progreso de la caries dental. Junto a otros microorganismos, B. dentium puede formar parte de la biopelícula en la superficie dental, denominada placa dental, donde diferentes especies bacterianas pueden interactuar a través de quorum sensing. En algunas bacterias Gram positivo orales, se ha descrito el sistema de quorum sensing mediado por autoinductor-2, el cual es producido por la enzima LuxS. Éste ha demostrado tener un papel en el crecimiento, la formación de biopelículas y la resistencia a los ácidos en esos microorganismos. En B. dentium, existe poca información de este sistema. El objetivo de este estudio fue analizar la producción de autoinductor-2 por parte de B. dentium ATCC 27534 y los efectos de éste en la adhesión y formación de biopelículas.
Materiales y métodos: la presencia y actividad de AI-2 producido por B. dentium ATCC 27534 fue detectada mediante el sistema de ensayo reportero Vibrio harveyi ATCC BAA-1117, utilizando el sobrenadante de cultivos de B. dentium ATCC 27534 obtenidos en distintas etapas de crecimiento. El efecto de autoinductor-2 en la adhesión de B. dentium ATCC 27534 se analizó en microplacas de poliestireno de 96 pocillos en presencia o ausencia de DPD (autoinductor-2 sintetizado químicamente), utilizando saliva y medio de cultivo, a las 16 horas. La adherencia bacteriana se midió usando el ensayo MTT y un SynergyHT (Biotek®). Finalmente, el ensayo de formación de biopelículas y efecto del DPD se realizó sobre cubreobjetos de vidrio embebidos en saliva, durante 3 y 5 días, bajo las mismas condiciones que el ensayo de adherencia, y se documentó con un microscopio electrónico de barrido (JSM-IT300, JEOL).
Resultados: Se demostró la presencia de autoinductor-2 en el sobrenadante de cultivos de B. dentium ATCC 27534, indicando que este microorganismo es capaz de producirlo y secretarlo al medio externo. Se determinó, además, que este microorganismo es capaz de presentar adhesión a superficies sólidas, la que

aumenta significativamente en presencia de autoinductor-2. Junto con esto, la presencia de autoinductor-2 exógeno incrementa la formación de biopelículas, mediante el aumento del número de células adheridas a la superficie, así como se pudo observar un aumento cualitativo de los niveles de exopolisacáridos excretados.
Conclusiones: Los resultados indican que el sistema de quorum sensing mediado por autoinductor-2 es importante para controlar los procesos fisiológicos de adhesión y formación de biopelículas en B. dentium ATCC 27534.
Investigaciones adicionales permitirán dilucidar cómo estos microorganismos se comunican entre sí a través de este sistema, así como su participación en la cascada de regulaciones que permiten la permanencia de B. dentium en los sitios de caries.Adscrito a Proyecto Fondecyt N° 11150928

Going Beyond Counting First Authors in Author Co-citation Analysis

counts are defined. Co-citation counting provides the data on which all subsequent statistical analyses and mappings
are based, and we compare ACA results based on two different types of co-citation counting - the traditional type that
only counts the first one among a cited work's authors on the one hand and a non-traditional type that takes into
account the first 5 authors of a cited work on the other hand. Results indicate that the picture produced through this non-traditional author co-citation counting contains more coherent author groups and is therefore considerably clearer. However, this picture represents fewer specialties in the research field being studied than that produced through the traditional first-author co-citation counting when the same number of top-ranked authors is selected and analyzed. Variations on the Author
Get PDF“Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
Appropriate Similarity Measures for Author Cocitation Analysis
Get PDFWe provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature. Note that the objective for the hut study was to evaluate the IRS treatment applications for residual efficacy against Anopheles mosquitoes, including the local An. coluzzii mosquito population. Cows were only used to attract mosquitoes into the huts and no tests were carried out directly on the cows. The author checklist is intended for use with studies where experiments are carried out on animals, which is why we have had such difficulty in completing this for the hut study, as many of the questions do not relate to how the cows were used
