Lost horizons: Revitalization project of the former military resort in Kupari
Magistrska naloga obravnava problematiko izgube identitete obalnega prostora južne Dalmacije skozi primer zgodbe nekdanjega vojaškega letovišča v Kuparih, enega najzanimivejših kompleksov jugoslovanske turistične dediščine. Izhodišče raziskave je osebno in prostorsko doživljanje turistične arhitekture, ki jo nadgradim z analizo družbenega, zgodovinskega in načrtovalskega konteksta Župe dubrovačke, v kateri se Kupari nahajajo. Naloga sledi plastenju identitete od agrarne in obrobne, preko historicistične in kolektivne, do njenega vrhunca v podobi jugoslovanskega modernizma ter njegovega razpada v času Domovinske vojne.
Posebna pozornost je namenjena tipološkemu razvoju hotelske arhitekture na Jadranu ter vlogi turizma kot ideološkega, gospodarskega in prostorskega katalizatorja. Analiza pokaže, da je jugoslovanski turizem ustvarjal odprte, javno dostopne in krajinsko integrirane prostore, katerih vrednote so danes ogrožene zaradi globaliziranega, potrošniško naravnanega družbeno prostorskega razvoja. Ruševine Kuparov so obravnavane kot nosilke kolektivnega spomina in prostorskega potenciala, ne kot prazna ali neuporabna dediščina.
Na podlagi celovite valorizacije prostorskih, arhitekturnih in krajinskih kvalitet naloga razvije projekt revitalizacije, ki letovišče razume kot hibridno mestotvorno in turistično središče. Predlagana zasnova temelji na ohranjanju, reinterpretaciji in rušitvi, pri čemer so trije hoteli – Grand, Pelegrin in Goričina – obravnavani z različnimi pristopi: diskretno prenovo, rekonstrukcijo in prezentacijo ruševine. Projekt vzpostavlja ravnovesje med turizmom in lokalno skupnostjo ter zagovarja arhitekturo kot medij za ponovno vzpostavitev identitete, kontinuitete in javnega prostora v ranljivem obalnem okolju.This thesis addresses the issue of the loss of identity in the coastal landscape of southern Dalmatia through the case study of the former military resort in Kupari, one of the most compelling complexes of Yugoslav tourist heritage. The research is grounded in a personal and spatial experience of tourist architecture, which is further expanded through an analysis of the social, historical, and planning context of Župa dubrovačka, where Kupari is located. The thesis traces the layering of identity from its agrarian and peripheral origins, through historicist and collective phases, to its peak embodied in Yugoslav modernism and its subsequent collapse during the Croatian War of Independence.
Special attention is devoted to the typological development of hotel architecture along the Adriatic coast and to the role of tourism as an ideological, economic, and spatial catalyst. The analysis demonstrates that Yugoslav tourism produced open, publicly accessible, and landscape-integrated spaces whose values are today threatened by globalized, consumption-driven socio-spatial development. The ruins of Kupari are therefore understood as carriers of collective memory and spatial potential, rather than as empty or obsolete heritage.
Based on a comprehensive evaluation of spatial, architectural, and landscape qualities, the thesis develops a revitalization project that conceptualizes the resort as a hybrid urban and tourist center. The proposed design is based on preservation, reinterpretation, and selective demolition, whereby three hotels—Grand, Pelegrin, and Goričina—are addressed through different approaches: discreet renovation, reconstruction, and the presentation of ruins. The project seeks to re-establish a balance between tourism and the local community and positions architecture as a tool for restoring identity, continuity, and public space within a fragile coastal environment
Comparison of culture-dependent and culture-independent methods for routine identification of airborne microorganisms in speleotherapeutic caves
The effective identification of bacterial and fungal isolates is essential for microbiological monitoring in environments like speleotherapeutic caves. This study compares MALDI-TOF MS and the OmniLog ID System, two high-throughput culture-based identification methods. MALDI-TOF MS identified 80.0% of bacterial isolates to the species level, while the OmniLog ID System identified 92.9%. However, species-level matches between the methods were only 48.8%, revealing considerable discrepancies. For discrepant results, MALDI-TOF MS matched molecular identification at the genus level in 90.5% of cases, while the OmniLog ID System matched only in 28.6%, demonstrating MALDI-TOF MS’s superiority. The OmniLog ID System had difficulties identifying genera from the order Micrococcales. Fungal identification success with MALDI-TOF MS was 30.6% at the species level, potentially improvable with a customised spectral library, compared to the OmniLog ID System’s 16.7%. Metagenomic approaches detected around 100 times more microbial taxa than culture-based methods, highlighting human-associated microorganisms, especially Staphylococcus spp. In addition to Staphylococcus spp. and Micrococcus spp. as indicators of cave anthropisation, metagenomics revealed another indicator, Cutibacterium acnes. This study advocates a multi-method approach combining MALDI-TOF MS, the OmniLog ID System, culture-based, and metagenomic analyses for comprehensive microbial identification. Metagenomic sampling on nitrocellulose filters provided superior read quality and microbial representation over liquid sampling, making it preferable for cave air sample collection
Tematizacija in prezentacija ljubezni na primeru Pohorskih poti Janka Glazerja
Prispevek izhaja iz opredelitve, da imajo čustva v literarnem besedilu subjektivno in intersubjektivno razsežnost, kar pomeni, da niso povsem subjektivni psihofizični fenomeni, ampak tudi kulturno kodirana. Tematizacija in prezentacija ljubezni v izbranih pesmih Janka Glazerja se pokaže kot preplet pesnikove subjektivnosti, njegove poetike in kulturnih značilnosti obdobja (konvencij, pravil, moralnih norm, literarnih stilov idr.). Čustvo ljubezni je v Glazerjevi poeziji ubesedeno v eksplicitni in implicitni obliki, slednje zlasti z motivi in leksiko Pohorja, metaforami in s prototipsko simboliko letnih časov.This article draws from the assumption that emotions in a literary text contain subjective and intersubjective dimensions, which means that emotions are not entirely psychophysical phenomena, but are also culturally coded. The thematization and presentation of love in selected poems by Janko Glazer reflect an interplay between the poet’s subjectivity and poetics, and the cultural characteristics (conventions, rules, moral norms, literary styles, etc.) of the period. In Glazer’s poetry, love is verbalized in explicit and implicit forms, the latter especially with the themes and lexis of the Pohorje Mountains, metaphors, and the prototypical symbolism of the seasons
Synthesis and determination of structural properties of a non-natural amino acid
Sinteza nenaravnih aminokislin in njihova vgradnja v proteine je vedno bolj zanimivo in intenzivno področje raziskovanja. Cilj je sintetizirati aminokisline z vgradnjo katerih bomo spremenili lastnosti proteinov in npr. dosegli njihovo večjo stabilnost, funkcionalnost in specifičnost. Cilj diplomskega dela je bila sintetiza nenaravne aminokisline – azahistidina, ki je analog naravnega histidina. Sinteza je potekala z uporabo z bakrom katalizirane reakcije med propargil glicinom in hidrazojsko kislino, ki je pri sintezi nastala in situ med natrijevim azidom in metanojsko kislino. Tako pripravljen azahistidin ima na stranski verigi triazolni obroč, ki med drugim lahko predstavlja koordinativno mesto za kovine. S titracijami (1H-1,2,3-triazola) sem preverjala, katere kovine in v kolikšni meri se lahko potencialno vežejo na triazolni fragment te nenaravne aminokisline. Ugotovili smo, da se na 1H-1,2,3-triazol najboljše vežejo kobaltovi in cinkovi ioni.The synthesis of non-natural amino acids and their incorporation into proteins is an increasingly exciting area of research that is developing rapidly. The aim is to synthesize amino acids whose incorporation into proteins changes their properties, e.g. increases their stability, functionality and specificity. The main objective of this thesis was to synthesise a non-natural amino acid, azahistidine, an analogue of the natural histidine. The synthesis was carried out by a copper-catalysed reaction between propargyl glycine and hydrazoic acid formed in situ during synthesis between sodium azide and methanoic acid. The azahistidine prepared has a triazole ring on the side chain, which, among others, can represent the coordinating site for metals. I performed titrations of 1,2,3-triazole (1H-1,2,3-triazole) to test which metals can bind to the triazole fragment of the non-natural amino acid and to what extent. Evaluation of the effects of microRNAs hsa-miR-375-3p, hsa-miR-363-3p, hsa-miR-20b-5p, hsa-miR-133a and hsa-miR-31-3p on the osteogenic differentiation of mesenchymal stem stromal cells
Na diferenciacijo mezenhimskih stromalnih matičnih celic (MSC) vplivajo številni epigenetski dejavniki, med katerimi imajo ključno vlogo mikroRNA (miRNA). MiRNA delujejo kot regulatorji izražanja genov, pomembnih za osteogenezo, in uravnavajo homeostazo skeletnega tkiva. Številne raziskave dokazujejo, da motnje v izražanju specifičnih miRNA niso povezane le z osteogenezo, temveč tudi z razvojem kostnih bolezni, kot je osteoporoza. Slednja predstavlja enega večjih javnozdravstvenih problemov, saj prizadene približno 200 milijonov ljudi po svetu. V magistrski nalogi smo preučevali vpliv izbranih miRNA (hsa-miR-375-3p, hsa-miR-363-3p, hsa-miR-20b-5p, hsa-miR-133a-3p in hsa-miR-31-3p) na osteogeno diferenciacijo MSC. MSC, ki smo jih uporabili kot celični model za raziskavo, so bile pripravljene z lentivirusno transdukcijo tako, da so se izbrane miRNA bodisi zvišano izražale bodisi so bile utišane. V teh celicah smo z metodo kvantitativne verižne reakcije s polimerazo v realnem času analizirali izražanje ključnih osteogenih označevalcev (ALPL, BGLAP, COL1A1 in RUNX2). Te rezultate smo kvantificirali z relativno kvanitifikacijo z metodo standardne krivulje in statistično ovrednotili s Studentovim t-testom ter jih primerjali z barvanjem po Von Kossa. Na podlagi rezultatov smo ocenili vpliv miRNA na osteogenezo MSC. Rezultati magistrske naloge potrjujejo hipotezo, da ima spremenjeno izražanje miRNA pomemben vpliv na osteogeno diferenciacijo. Med preučevanimi miRNA je hsa-miR-363-3p izkazala največji potencial kot biološki označevalec in/ali terapevtska tarča, saj smo z rezultati qPCR dobili statistično značilne razlike v izražanju osteogenih genov v primerjavi s kontrolnimi vzorci, prav tako pa so se rezultati qPCR skladali z rezultati barvanja po Von Kossa. Naši rezultati prispevajo k boljšemu razumevanju vloge miRNA v osteogenezi in njihovemu potencialu v diagnostiki ter terapiji osteoporoze. Kljub obetavnim rezultatom uporaba miRNA v klinični praksi ostaja izziv zaradi njihove nespecifičnosti, nestabilnosti in pomanjkanja učinkovitih dostavnih sistemov. Potrebne so nadaljnje raziskave na kliničnih vzorcih in modelih in vivo za boljše razumevanje ter klinično implementacijo miRNA.The differentiation of mesenchymal stromal cells (MSCs) is influenced by numerous epigenetic factors, among which microRNAs (miRNAs) play a key role. MiRNAs act as regulators of gene expression important for osteogenesis and help maintain skeletal tissue homeostasis. Numerous studies have demonstrated that dysregulation of specific miRNAs is not only associated with osteogenesis but also with the development of bone diseases such as osteoporosis. Osteoporosis represents one of the major public health challenges, affecting approximately 200 million people worldwide. In this master’s thesis, we studied the impact of selected miRNAs (hsa-miR-375-3p, hsa-miR-363-3p, hsa-miR-20b-5p, hsa-miR-133a-3p, and hsa-miR-31-3p) on the osteogenic differentiation of MSCs. The MSCs used as the cellular model for the study were prepared through lentiviral transduction, resulting in either overexpression or knockdown of the selected miRNAs. In these cells we analyzed the expression of key osteogenic markers (ALPL, BGLAP, COL1A1 and RUNX2), using quantitative real-time polymerase chain reaction. The results were quantified by relative quantification using the standard curve method and statistically evaluated using Student\u27s t-test. These results were further compared with Von Kossa staining. Based on the results, we assessed the influence of miRNAs on the osteogenesis of MSCs. The results of the thesis confirm the hypothesis that altered miRNA expression significantly affects osteogenic differentiation of MSCs. Among the studied miRNAs, hsa-miR-363-3p showed the greatest potential as a biological marker and/or therapeutic target, as qPCR results showed statistically significant differences in the expression of osteogenic genes compared to control samples, and qPCR results were consistent with Von Kossa staining. Our results contribute to a better understanding of the role of miRNAs in osteogenesis and their potential in the diagnosis and treatment of osteoporosis. Despite the promising findings, the use of miRNAs in clinical practice remains a challenge due to their nonspecificity, instability, and lack of efficient delivery systems. Further research using clinical samples and in vivo models are necessary for better understanding and to enable the clinical implementation of miRNAs

Prispevek prinaša vpogled v romanja in romarske procesije predjožefinskih župnij Zgornje Savinjske doline v času baroka. Na podlagi podatkov iz matrikul in vizitacij je predstavljen koledar romarskih procesij, v nadaljevanju pa so sistematično obravnavane vse romarske cerkve tega področja.The article provides an insight into the pilgrimages and pilgrimage processions that took place in pre-Josephinian parishes of the Upper Savinja Valley during the Baroque period. Based on information from registers and visitation records, the calendar of pilgrimage processions is presented, followed by a systematic discussion of all the pilgrimage churches in the area

Trail Trap: a variant of partizan edge geography
We study a two-player game played on undirected graphs called Trail Trap, which is a variant of a game known as Partizan Edge Geography. One player starts by choosing any edge and moving a token from one endpoint to the otherthe other player then chooses a different edge and does the same. Alternating turns, each player moves their token along an unused edge from its current vertex to an adjacent vertex, until one player cannot move and loses. We present an algorithm to determine which player has a winning strategy when the graph is a tree and partially characterize the trees on which a given player wins. Additionally, we show that it is NP-hard to determine if Player 2 has a winning strategy on Trail Trap from the starting position, even for connected bipartite planar graphs with maximum degree 4.
